    Notes: Since all long-distance trade in the Roman world travelled by water, Roman harbour design and construction have special importance. Harbour excavation must be supplemented by analysis of the components of the hydraulic concrete, structural analysis of the cementing materials, and consideration of the design of the wooden formwork. The authors have begun collecting large cores from concrete blocks at Roman harbours and other maritime structures, analysing the materials used, the method of placement, and the structural characteristics of the resulting concrete. These data have provided new information on the engineering properties of Roman concrete, the process of funding and execution, and the trade in the volcanic ash which was the crucial component of hydraulic concrete.© 2004 The Nautical Archaeology Society

    Notes: Abstract A major impediment to the implementation of displacement chromatography has been the lack of suitable displacer compounds. Recently, it has been shown that low molecular weight dendritic polymers, protected amino acids and antibiotics can be successfully employed for displacement purification in cation-exchange systems. In this paper, a variety of low molecular weight anionic displacers are identified for the resolution of a bovine β-lactoglobulin mixture into two closely related forms (A and B). A Dynamic Affinity plot is employed to evaluate the affinity of these low molecular weight compounds under various displacement conditions. In contrast to large polyelectrolyte displacers, the efficacy of these low molecular weight displacers are shown to be dependent on displacer concentration. In fact, the Dynamic Affinity Plot qualitatively predicts the transition from a displacement to a desorption regime with these low molecular weight displacers. In addition to the fundamental interest generated by low molecular weight displacers, it is likely that these displacers will have significant operational advantages as compared to large polyelectrolyte displacers. Furthermore, the ability to carry out selective displacement chromatography with these low molecular weight displacers offers significant potential for developing robust large scale displacement processes.

    Description / Table of Contents: Résumé On a développé une éprouvette d'essai de rupture en mode II à partir de recherches expérimentales et numériques en vue d'être appliquées au bois. On a focalisé le développement sur une éprouvette compacte de cisaillement, conçue de manière à mettre en valeur l'orthotropie extrême des propriétés élastiques et de la résistance du bois. On a établi la ténacité à la rupture sous mode II à partir de données d'essais sur des échantillons de pin méridional et de pin Douglas, combinées à une analyse par éléments finis à singularité quart-points. On montre à évidence que ce type d'éprouvette convient à des recherches sur la rupture de matériaux à orthotropie particulièrement élevée.
    Notes: Abstract A mode II fracture specimen has been developed from experimental and numerical investigation for applications with wood. Development centered on the compact shear specimen and addressed the extreme orthotropy of both the elastic and strength properties of wood. Mode II fracture toughness was assessed from test data gathered for southern pine and Douglas-fir specimens combined with use of quarter-point singular finite elements. Evidence is provided that this specimen is suitable for investigators to employ in fracture studies with highly orthotropic materials.

    Notes: Although the ability to carry out simultaneous concentration and purification in a single displacement step has significant advantages for downstream processing of pharmaceuticals, a major impediment to the implementation of displacement chromatography has been the lack of suitable displacer compounds. An important recent advance in the state of the art of displacement chromatography has been the discovery that low-molecular-weight dendritic polymers can be successfully employed as displacers for protein purification in ion-exchange systems. In this article, protected amino acid esters (based on arginine and lysine) are shown to be useful displacers for protein purification in cation-exchange systems. A dynamic affinity plot is employed to evaluate the affinity of these low-molecular-weight compounds under dis-placement conditions. In contrast to large polyelectroyte displacers, the efficacy of these low-molecular-weight displacers was shown to be dependent on both the initial carrier salt concentration and the displacer concentration. In addition to the funcamental interest generated by low-molecular-weight displacers, it is likely that these displacers will have significant operatioal advantages as compared with large polyelectrolyte displacers. © 1995 John Wiley & Sons, Inc.

    Notes: Nonlinear adsorption plays an important role in determining the chromatographic behavior of proteins in preparative ion-exchange chromatography. In this article, the steric mass action (SMA) isotherm is used in conjunction with a mass transport model to describe nonlinear cation-exchange chromatography. Excellent agreement is observed between simulated and experimental step gradient separations of the proteins α-chymotryp-sinogen A, cytochrome C, and lysozyme. A systematic method of selecting the optimum step gradient program for a given separation problem is presented and employed to study optimization of step gradient chromatography under conditions of high mass loading. This article includes consideration of the effects of the adsorption properties of the feed stream, the feed stream concentration, protein solubility, and otherconstraints on the optimum separation conditions.© John Wiley & Sons, Inc.

    Notes: Enzymatic peptide synthesis was investigated using carboxypeptidase Y immobilized with glutaraldehyde on 10 μm microparticulate amino-silica. Carboxypeptidase Y was immobilized with 98.5% recovery of active enzyme to yield the immobilized enzyme having 0.55 units esterase activity/mg amino-silica support. The stability of the immobilized enzyme was examined as a function of pH, temperature, and reactant concentrations. Immobilized Carboxypeptidase Y was used in stirred batch and recirculating packed-bed reactors for peptide synthesis. Packed-bed reactors (40 × 4.6 mm, 60 × 4.6 mm) were used to catalyze the synthesis of 170 mg N-benzoyl-L-arginyl-L-methioninamide, 380 mg N-benzoyl-L-arginyl-L-methionyl-L-leucinamide, and 200 mg N-benzoyl-L-arginyl-L-methionyl-L-leucyl-L-phenylalaninamide in 8, 3, and 1 hour, respectively, as intermediates in the synthesis of L-methionyl-L-leucyl-L-phenylalanine. No inactivation of the immobilized enzyme was observed during the course of the reactions. The N-benzoyl-L-arginyl group served to increase the water solubility of the peptides and was removed by immobilized trypsin at the end of synthesis to obtain the final product. While the first two syntheses were conducted with aqueous reaction mixtures, the synthesis of N-benzoyl-L-arginyl-L-methionyl-L-leucyl-L-phenylalaninamide was carried out in a reaction mixture containing dimethylformamide to avoid precipitation of the product. HPLC and amino acid analysis confirmed the high purity and amino acid composition of the final product.
