    Notes: Summary Leukocyte-derived inhibitory activity inhibiting the entry of normal progenitor cells of granulocytes and macrophages (CFU-GM) into the S-phase of a cell cycle was investigated in 16 patients with different forms of myelodysplastic syndrome (MDS). The presence of this inhibitory activity was analysed in medium conditioned with low-density cells obtained from peripheral blood of MDS patients. The inhibition rate was measured by 3H-thymidine suicide technique with subsequent cultivation of pretreated cells in semisolid agar medium. Low-density cells from MDS patients of various types were studied: from the twelve patients with refractory anaemia (RA or RAS) only three were positive, one patient with chronic myelomonocytic leukaemia (CMML) was negative while one patient with refractory anaemia with excess of blasts (RAEB) and two patients with RAEB in transformation (RAEB-T) were positive with respect of the described test. In two patients with RA, who underwent a long-term investigation, the production of leukocyte-derived inhibitory activity preceded the development of disease into RAEB or RAEB-T. In five positive cases, supernatants were incubated with antiserum against human placental ferritin; with one exception, the inhibitory activity was neutralized.

    Notes: Summary Monoclonal antibody 2A4 against human heart ferritin and a polyclonal antibody against human placental ferritin inactivated the suppressor activity present in media conditioned by cells of patients with myelodysplastic syndrome (MDS). These antibodies also neutralized the inhibitory activity released by cells of myeloid leukemic ML-2 cell line. In contrast, a monoclonal antibody against the ferritin L subunit did not neutralize the inhibitory activity of the media conditioned either by the cells of patients with MDS or by the ML-2 cell line. Neutralization of the inhibitory activity present in supernatants of cells of MDS patients and the ML-2 cell line by monoclonal antibodies against the H subunit of ferritin strongly indicates that the inhibitory activity resides in the acidic isoferritin molecule.

    Notes: Summary Conditioned media from the human myeloid leukemic cell line ML-2 contain a factor that inhibits the entry of normal CFU-GM into S phase of mitotic cycle as measured by the 3H-TdR suicide technique. This factor was detected in conditioned media prepared by incubating 5×106 ML-2 cells/ml or 1×106 ML-2 cells/ml in serum-free RPMI for 5 or 24 hours respectively, and was isolated by ultrafiltration through an XM300 Diaflo membrane followed by chromatography on Sepharose 6B. Ferritin, prepared from human placenta, had the same inhibitory effect on CFU-GM. Antibodies against human placental ferritin completely inactivated the inhibitory effect of both human placental ferritin and the factor released from ML-2 cells. The inhibitory activity produced by the cell-line ML-2 was considered as LIA (leukemia cell-derived inhibitory activity) earlier found in HL-60 cell line and AML and CML cells.

    Notes: Abstract In tests with 72 human donors, the outcome of the xenogeneic human-mouse MLR was influenced more by the general capacity of the human responder cells to react than by the MHS genotype of the mouse stimulating cells. While the response of the human lymphocytes of the same donor to the lymphocytes of six congenic mouse strains with differentH-2 haplotypes was generally rather similar, there were pronounced differences among individual human donors in the reactivity in these tests. These differences correlated with HLA, since the low reactivity was associated with the presence of the antigens HLA-AW25, HLA-B18, and HLA-DW2. The effect of the presence of antigens AW25, B18 was much more pronounced in males than in females. The low capacity to react in xenogeneic MLR in individuals with antigens HLA-AW25 and HLA-B18 may be the result of the general immune insufficiency associated with these antigens, which also includes C2 defficiency and association with immunopathological diorders.

    Notes: Abstract Our data demonstrate the granulopoietic toxicity of aflatoxin B1 (AFB1)in vivo and show an impact of this mycotoxin on the production of some humoral regulatory factors dealing with the granulopoietic developmental pathway (CSA, IL-1, IL-2). The dose of AFB1 studied represented approximately 1/5 of LD50 for young male rats. An early suppressive effect of AFB1 towards CFU-GM was transient in treated animals. The peak in granulopoietic activity was preceded in time by an increased CSA and IL-1 formation. Elevated IL-2 synthesis and increased T cell activation paralleled the peak in granulopoietic activity.
