    Notes: The extracellularly secreted endopeptidase elastase (LasB) is regarded as an important virulence factor of Pseudomonas aeruginosa. It has also been implicated in the processing of LasA which enhances elastolytic activity of LasB. In order to Investigate the role of LasB in virulence and LasA processing, a LasB-negative mutant, PA01E, was constructed by insertional mutagenesis of the LasB structural gene, lasB, in P. aeruginosa PAO. An Internal 636 bp lasB fragment of the plasmid pRB1803 was ligated into a derivative of the mobilization vector pSUP201–1. The resulting plasmid, pBRMOB-LasB, was transformed into Escherichia coli and transferred by filter matings to the LasB-positive P. aeruginosa strain, PA01. Plasmid integration in the lasB site of the chromosome was confirmed by Southern blot analysis. Radioimmunoassay and immunoblotting of PA01E supernatant fluids yielded no detectable LasB (〈1 ng ml-1 LasB). The absence of LasB in PA01E was further proven by the inability of its culture supernatant fluid to cleave transferrin or rabbit immunogiobulin G (IgG) after a 72 h incubation. The residual proteolytic activity of PA01E culture supernatant fluid was attributed to alkaline proteinase (Apr), since it was totally inhibited by specific antibodies against Apr. Residual elastolytic activity in culture supernatant fluid of PAO1E was due to the LasA fragment and to the combined action of the LasA fragment with Apr on elastin. The sizes of purified LasA from PA01 and PA01E were identical (22 kDa). These results show that, besides LasB and the LasA fragment, Apr may also act on elastin in the presence of the LasA fragment and that the proteolytic processing of LasA in P. aeruginosa is independent of LasB.

    Notes: Abstract A purification method was developed to isolate Legionella pneumophila cytotoxic protease in a form suitable for biological assays. Culture supernatant of a clinical isolate of L. pneumophila, Knoxville 1 strain, was used as the starting material. The protease was purified by FPLC on a Mono Q column followed by ultrafiltration. The isolated proteolytic enzyme has a specific activity of 90 azocasein units/mg protein and is a 42 kDa monomeric protein as determined by SDS-PAGE and gel filtration chromatography. It is heat-labile and toxic to a variety of cells e.g. McCoy, SIRC, HeLa, and rhabdomyosarcoma cells, baby hamster and green monkey kidney cells, and human embryonic lung fibroblasts.

    Notes: Zusammenfassung An in situ belassenen Mägen weißer weiblicher Ratten vom Stamm Sprague-Dawley wird mit einer photographischen Registriermethode postmortale Magenperistaltik aufgenommen. Es wird eine neue Peristaltikauswertung nach Anzahl, Gesamtamplitude, Zeitdauer und Stärke beschrieben. Je nach Todesart und Giftdosis ergeben sich deutliche Unterschiede im Ablauf der Peristaltik, die zudem noch vom Füllungszustand des Magens bei Todeseintritt abhängig ist.

    Description / Table of Contents: Zusammenfassung Zur Klärung der Frage, ob ein negativer Befund der gaschromatographischen Blutuntersuchung auf Bestandteile der „chemischen Keule“ beweist, daß keine oder allenfalls nur leichte Tränengasexposition vorgelegen hat, wurden Tierversuche durchgeführt. Im Blut von 10 Meerschweinchen, die den Komponenten der chemischen Keule 1–6 Std ausgesetzt worden waren, konnten die Trägergase 1,1,2-Trichlor-1,2,2-trifluoräthan (Frigen 113) und 1,1,1-Trichloräthan — auch noch 23 Std nach Expositionsende sowie nach 18wöchiger Lagerung der Blutproben — deutlich, der Wirkstoff Chloracetophenon (CN) dagegen überhaupt nicht nachgewiesen werden. In vitro-Versuche zeigten, daß das sehr reaktionsfähige CN relativ rasch mit Blutbestandteilen reagiert. Blutuntersuchungen auf CN müssen daher möglichst sofort nach der Entnahme durchgeführt werden. Bei Einatmung von Bestandteilen der chemischen Keule, d. h. bei der vergleichsweise leichtesten Einwirkungsart des Chloracetophenon, dürfte der Wirkstoff infolge schneller Reaktion mit Proteinen der respiratorischen Oberfläche der Lungen gar nicht erst in das Blut gelangen.
    Notes: Summary To answer the question whether a negative result of gas chromatographic blood analysis for components of chemical mace proves that no or at most only slight tear gas exposure can have occurred, animal experiments were carried out. In the blood of 10 guinea pigs, which were exposed to the contents of chemical mace for 1–6h, the solvants 1,1,2-trichloro-1,2,2-trifluorethane (freon 113) and 1,1,1-trichloroethane could easily be detected—even 23 h after the end of exposure or after a storage of the blood samples for 18 weeks—whereas the lacrimator chloracetophenone (CN) could not be found at al. In vitro experiments showed that CN relatively quickly reacts with components of blood. Therefore, blood samples should be analyzed for CN after withdrawal as soon as possible. In case of inhalation of the contents of chemical mace, i.e., after the comparatively mildest form of CN application, most probably no traces of the lacrimator at all can pass into the blood due to the quick reaction of CN with proteins of the respiratory surface of the lung.

    Notes: Summary The aim of the present paper was to find an analytic method which, in one operation, allows to detect in organic matter (blood and tissues) as many pesticides as possible. Steam distillation as a method of isolation with susbsequent infra-red spectrophotometric identification proved to be useful in special cases only. A chromatographic procedure, however, allowed to isolate the pesticides: aldrin, chlordane, DDT, dieldrin, endrin, heptachlor, hexachlorcyclohexane, methoxychlor, parathion, malathion, phosdrin and thiodan from organic matter and to identify them by means of the infra-red spectra.

    Description / Table of Contents: Summary In contrast to the unstretched muscle, the stretched muscle may decrease in size postmortally. The cause of this difference in behaviour is presumably the difference in the ATP metabolism. Therefore, we determined the ATP as well as creatine phosphate, glycogen, glucose, and lactic acid under aerobic and anaerobic conditions in the unstressed and stressed M. gastrocnemius of the rat at various times after death. 1. The ATP contents of the gastrocnemius varied distinctly under different experimental conditions. The highest content was found in the stressed muscle under aerobic conditions, as of approximately 30 min p.m. Under nitrogen, the ATP content of the stressed muscle was always lower than the content of the unstressed muscle. 2. The creatine phosphate and glycogen catabolism in the unstretched muscle always occurred faster than in the stretched muscle. While the creatine phosphate catabolism under nitrogen and air occurred virtually at the same rate of speed, glycogenolysis under anaerobic conditions showed a marked acceleration as of approximately 2 hours p.m. The lactic acid content of the unstressed muscle rose faster than in the stressed muscle; under nitrogen the increase was stronger than under air. 3. Based on the results of these experiments it appears probable that aerobic processes occur also postmortally in the muscle und supply energy for the postmortal contractions.
    Notes: Zusammenfassung Der gedehnte Muskel kann sich im Gegensatz zum ungedehnten Muskel postmortal verkürzen. Als Ursache dieses unterschiedlichen Verhaltens wurden Unterschiede im ATP-Stoffwechsel vermutet. Wir bestimmten daher ATP sowie Kreatinphosphat, Glykogen, Glucose und Milchsäure unter aeroben und anaeroben Bedingungen im belasteten und unbelasteten M. gastrocnemius der Ratte zu verschiedenen Zeiten nach dem Tode. 1. Die ATP- Gehalte des Gastrocnemius unterschieden sich unter den verschiedenen Versuchsbedingungen deutlich. Der höchste Gehalt wurde von etwa 30 min p.m. an im belasteten, aerob gehaltenen Muskel gefunden. Unter Stickstoff lag der ATP-Gehalt des belasteten Muskels immer unter dem Gehalt des unbelasteten Muskels. 2. Der Kreatinphosphat- und Glykogenabbau waren im ungedehnten Muskel stets schneller als im gedehnten. Während Kreatinphosphat an Luft und unter Stickstoff praktisch gleich schnell abgebaut wurde, war die Glykogenolyse unter anaeroben Bedingungen von etwa 2 Std p.m. an deutlich beschleunigt. Der Milchsäuregehalt stieg im unbelasteten Muskel rascher an als im belasteten und unter Stickstoff stärker als an Luft. 3. Auf Grund der Versuchsergebnisse erscheint es wahrscheinlich, daß im Muskel auch postmortal aerobe Prozesse in geringem Umfang ablaufen und die Energie für die postmortalen Kontraktionen liefern.
