    Notes: A multinational co-trial was organized to determine if electron spin resonance (ESR) spectroscopy could be used to monitor foods exposed to ionizing radiation. The bones of chicken legs, frog legs and pork rib bones were prepared and distributed as unknowns to the participating laboratories. In every instance, non-irradiated bones were correctly identified as such. Moreover, irradiated bones were not only correctly identified, but relatively good estimates of the absorbed dose were obtained. An intercomparison of the different approaches used by each laboratory is discussed, and recommendations for future trials are presented.

    Notes: Background Multiple mediators including prostaglandin D2 and leukotriene B4 have been shown to increase in nasal secretions during the early response to nasal challenge with antigen.Objective Our objective was to investigate the time course of prostanoid and leukotriene B4 release into nasal secretions on both the ipsilateral and contralateral side after a unilateral nasal allergen challenge.Methods We performed a controlled, randomized trial. Six volunteers were challenged unilaterally with antigen or diluent in a randomized order and discs were used to collect nasal secretions from both nostrils at 2 min intervals for 20 min after the challenge. Prostanoids and leukotriene B4 (LTB4) in recovered nasal secretions were measured by combined capillary gas chromatography-negative ion chemical ionization mass spectrometry (GC/MS).Results Nasal allergen challenge resulted in a significant and immediate increase in symptoms and sneezing. PGD2 was significantly elevated above diluent values (0.6 ± 0.6pg) 30s after removal of the allergen disc (P〈0.05), reached its peak (423.2 ± 182.4pg) at 2 min and then slowly decreased. PGD2 also increased on the contralateral side after unilateral allergen challenge, reaching peak values about six times lower than on the ipsilateral side (70.8 ± 2l.7pg at 6 min). Levels of 9a, Ub-PGF2 after antigen provocation became significantly higher than after diluent (0 ± 0pg) on the ipsilateral side at 2 min (17.2 ± 5.9 pg), and reached peak levels at 4 mm (25.1 ± 8.0 pg). LTB4 also increased significantly on the side of ehallenge. For the other prostanoids measured (PGF2, PGF2α TxB2, 6kPGF1α), no significant changes in either ipsilateral or contralateral secretions were observed after allergen challenge.Conclusions Our study described the kinetics of PGD2 and LTB4 release as well as the contralaterai release of PGD2.

    Notes: Background Subjects with occupational asthma (OA) often report nasal symptoms, but nasal reactions to inhalation challenges with occupational agents have not been well characterized. Methods Fifteen subjects with OA (eight due to high-molecular-weight agents - flour and guar gum - and seven due to isocyanates) underwent inhalational challenges using closed-circuit devices (dry particles for high-molecular-weight agents and gas generator for isocyanates) on two occasions, 2-4 weeks apart in a random fashion. On one occasion, they inhaled through the nose and, on another, through the mouth. The FEV, was monitored for up to 8 h afterward, and symptoms were documented with a standardized questionnaire on nasal symptomatology, assessment of nasal resistance by rhinomanometry, and nasal lavage for the examination of cells and mediators. Results Inhaling through the mouth and through the nose:〈list xml:id="l1" style="custom"〉1)yielded similar asthmatic responses (25 ± 8% and 22 ± 10% maximum changes in FEV1)2)more than doubled the peak nasal symptoms and nasal resistance when the maximum daily response was compared with prechallenge results.This increase occurred on the days of inhalational challenges through the mouth and through the nose. There were some significant responses assessed by nasal lavage in terms of cells and mediators, again with no differences between the days of challenges through the mouth and through the nose. Conclusions Inhaling occupational agents of high or low molecular weight, including isocyanates, whether through the mouth or nose:〈list xml:id="l2" style="custom"〉1)results in a similar asthmatic response2)causes a significant nasal response in terms of symptoms and an increase n i nasal resistance3)causes some significant changes in inflammatory cells and mediators.

    Notes: Several antifoam agents were evaluated for the ability to control foam in the production of Bacillus thuringiensis-based biopesticides using sewage sludge as a raw material. Experiments were conducted in shake flasks as well as in 15 l fermentors with controlled parameters. Polypropylene glycol (PPG), the most commonly used antifoam agent in B. thuringiensis fermentation, inhibited cell growth, sporulation and decreased the entomotoxicity yield even at a concentration of 0.1% (v/v) in sewage sludge medium. About 40% reduction in entomotoxicity was observed when PPG was used at 0.3% (v/v). The impact of PPG on sporulation and toxin synthesis in tryptic soy yeast broth (TSYB) medium was also studied. The inhibitory effects were less severe in TSYB than in sludge medium. Another silicone-based antifoam agent, “Antifoam A”, showed less severe effect on growth and stendotoxin production. The problem of the inhibitory effect of chemical antifoam agents on growth and endotoxin production was minimised substantially with the use of vegetable oils such as canola, olive, and peanut oils. Canola and peanut oil stimulated both sporulation and δ-endotoxin synthesis. The stimulus effect varies with the monounsaturated fat contents of oils.
