    Notes: The aim of the present study was the quantification of proteins present in crevicular fluid obtained from individual sulci, and to this effect simplified micromethods for the quantification of individual proteins were developed. Unstimulated, resting crevicular fluid from healthy crevicular spaces (i.e. in the absence of clinically detectable inflammation) of dogs and humans was collected for 3 minutes, either in microcapillary glass tubes or with the aid of filter paper strips. Quantification of proteins was done by radial immunodiffusion without the pooling of samples or by immunoelectrophoresis, without eluting from the papers. The concentrations of albumin and fibrinogen in crevicular fluid so determined proved to be significantly lower than plasma concentrations. These results strengthen the concept that capillary dynamics in healthy gingivae are similar to those in general tissue capillary beds and that gingival fluid is a plasma transudate. However, due to ever present sub-clinical inflammation and other factors, it is improbable that fluid from even healthy sulci is a true transudate.

    Notes: Abstract Ipsapirone is a new pyrimidinylpiperazine ligand specific for 5-HT1A receptors, with potential therapeutic use in affective disorders. Because 5-HT is involved in the regulation of sleep, we investigated the effect of ipsapirone hydrochloride on sleep patterns in 18 normal, healthy subjects of both sexes. Compared to placebo, ipsapirone 5 mg administered by mouth three times daily for 14 days decreased rapid eye movement (REM) sleep duration and, by the tenth day of treatment, began to reduce slow wave sleep (SWS) duration. The decrease in REM sleep occurred in the first 3 h of sleep. The latency to REM sleep was increased from the first night following ipsapirone administration, remained increased throughout the 14 days of administration, and fell to equal latency on placebo immediately administration ended. Subjective assessments of sleep revealed no differences between ipsapirone and placebo. Our experiments confirm a role of 5-HT1A receptors in sleep. The effects of ipsapirone on the sleep patterns of patients with affective disorders still need to be determined.

    Notes: Summary We investigated the effect of a 12-week physical training programme on the sleep of nine unfit women. Data were collected at 0, 4, 8 and 12 weeks of the training programme. Changes in fitness were assessed by changes in maximal oxygen consumption ( $$\dot V_{O_{2\max } } $$ ) and onset of blood lactate turn point (LTP). Lean body mass (LBM) was calculated from total body potassium measurements. The all-night sleep recordings were made following days during which the subjects carried out their normal daily routines and did no extra exercise. Although cardiorespiratory fitness improved significantly as indicated by an increase in both $$\dot V_{O_{2\max } } $$ and LTP, there was no change in LBM. The improvement in cardiorespiratory fitness did not result in any changes in the sleep parameters measured. Other workers have reported an improvement in sleep quality in eight army recruits during their basic training. The male recruits showed improved cardiorespiratory fitness and an increase in muscle bulk. These results suggest that increased fitness only facilitates sleep when there is an associated increase in LBM. Alternatively it may be that the response of sleep to improved fitness is sex-linked.
