    Notes: Experiments were performed to investigate the role of adherent (A) cells in graft-versus-host (GVH)-induced immunosuppression. GVH reactions (GVHR) were induced in adult, F1 hybrid mice by intravenous injections of parental lymphoid cells. Spleen cells (SC) from mice experiencing a GVHR (GVH mice) were stimulated with phytohaemagglutinin (PHA), concanavalin (Con A), and lipopolysaccharide(LPS). SC taken in the early phase of the GVHR (early GVHR) responded normally to LPS but did not respond to PHA and Con A. SC taken in an advanced phase of the GVHR (advanced GVHR) did not respond to PHA, Con A, or LPS. The influence of A cells from GVH mice (GVH-A cells) on the response of normal non-adherent cells to sheep erythrocytes (SRBC), PHA, and LPS was investigated. A cells from early GVHR, used in appropriate numbers, stimulated the responses to SRBC and to PHA; in excess they inhibited both responses. They had no effect on the response to LPS. A cells from advanced GVHR, even in low numbers, suppressed the responses to SRBC, PHA and LPS. The lymphoregulatory activities of GVH-A cells seemed to be mediated by soluble factors. The results indicate that the GVHR evokes complex non-specific regulatory interactions between A cells and lymphocytes.

    Notes: As part of a study of the effectiveness of beclomethasone dipropionate in perennial rhinitis five immunologically related parameters were measured. These included skin tests, total IgE, specific IgE, nasal and peripheral blood eosinophil counts. In addition, nasal biopsies were obtained in twenty out of sixty patients.It was apparent that patients with perennial rhinitis formed a heterogeneous group who could be divided into three groups based on the extent of immunological abnormality detected. Group 1 consisted of twenty patients who displayed no abnormality in any of the five parameters studied. Group 2 consisted of twenty-six patients in whom specific IgE was not detected but who had varying degrees of abnormal findings in the other laboratory parameters. The remaining fourteen patients formed a third group in whom specific IgE was detected in addition to abnormalities in other parameters.In general, patients with detectable specific IgE had a higher frequency of abnormal parameters, including abnormal histology of their nasal biopsy, than patients in whom no specific IgE was detected. The heterogeneous nature of perennial rhinitis is discussed. Despite the heterogeneous nature of the total patient group, the majority of patients (80%) treated with beclomethasone dipropionate improved compared to 20% improved in the placebo group. There was no difference in the drug effect between the three groups of patients.

    Notes: Résume L'effet d'une injection de 100 mg/kg de 3,4-diméthoxyphényléthylamine (DIMPEA) sur l'excrétion urinaire des catécholamines a été étudié chez le rat en cage métabolique. Il est montré que le DIMPEA produit une augmentation significative de l'excrétion urinaire d'acide homovanillique (HVA), le principal métabolite de la dopamine. Le mécanisme d'action plus précis a été étudié chez le chien et il est proposé que le DIMPEA produit une accélération du «turnover» de la dopamine par une augmentation de synthèse de l'amine, elle-même consécutive à un bloc au niveau des terminaisons dopaminergiques.

    Notes: Abstract The present study has examined the inhibition of synaptosomal45calcium uptake by trimipramine, oxaprotiline and doxepin, and their stereoisomers, in synaptosomes from the rat hippocampus. No significant difference in potency could be established for inhibition of net depolarization-induced45calcium uptake for any pair of antipodes, and the IC50 values for calcium channel blockade were in the vicinity of 30 µM for this group of compounds. Trimipramine, doxepin and oxaprotiline also inhibited the45calcium uptake mediated by Na+-Ca2+ exchange, with IC50 values of 71 µM, 110 µM, and 100 µM, respectively. The similar potency of doxepin isomers for inhibition of voltage-dependent calcium channels is in harmony with their reported similar potency in the clinic. A slight difference in potency is reported between the isomers of oxaprotiline in the behavioral despair test in rats, and the dextrorotatory isomer of trimipramine is reported to be a much more potent antidepressant than the levorotatory isomer: these order of potencies do not correspond perfectly with the similar potency of the antipodes against voltage-dependent calcium channels. The present study of stereoisomeric tricyclic antidepressants therefore fails to provide unequivocal support for the hypothesis that calcium channel blockade by tricyclic antidepressants is involved in their therapeutic effect.

    Description / Table of Contents: Sommario 33 pazienti epilettici con TAC normale (gruppo 1) e 31 pazienti con atrofia cerebellare e del tronco (gruppo 2) sono stati inclusi in questo studio randomizzato. Non ci sono differenze statistiche significative tra i due gruppi in correlazione con l'età, l'educazione e il numero di crisi di grande male o di altro tipo. Le analisi statistiche però hanno dimostrato che il gruppo 2 ha più lunga storia di epilessia con conseguente più lunga durata di assunzione della fenitoina. La messa a punto neuropsicologica ha rivelato un più basso rendimento di questo gruppo in tutti i tests eseguiti, tranne che per alcuni subtests della scala Wais, per i subtests del Wechsler Memory Scale per il richiamo immediato e ritardato e per i tempi di movimento visivi e uditivi nei quali non si constatavano differenze tra i due gruppi. L'analisi di questi dati dimostra che il cervelletto interferisce nelle funzioni complesse di comportamento rappresentate dall' organizzazione visuo-spaziale per un compito concreto, di pianificazione e programmazione di attività giornaliera e per la rapidità dei processi di informazione.
    Notes: Abstract Thirty-three outpatient epiletics with normal CT scans (group 1) and 31 patients with cerebellar and brain stem (CBS) atrophy (group 2) were randomly included in this study. There were no significant statistical differences between the groups with regard to age, education, and number of grand mal and other seizures. Statistical analyses showed that group 2 had a longer history of epilepsy with a consequently longer duration of phenytoin (PHT) consumption. Neuropsychological assessment revealed lower performance by this group on the following measures: full I. Q. scale, verbal I.Q. scale, performance I.Q. scale, information, arithmetic, block design, object assembly, digit symbol, Stroop test forms I and II, the B-M dexterity test, and the simple visual and auditory reaction time. No significant differences were observed between the two groups for the remaining 5 subtests from the WAIS scale, for the immediate recall and the delayed recall subtests belonging to Wechsler memory scale as well as for visual and auditory movement time. Analysis of the composite scores of neuropsychological performance showed that the cerebellum interferes with the following complex behavioral functions: (ì) visuo-spatial organization for a concrete task, a function related to the cerebelloparietal loops' (ii) planning and programming of daily activities, a function related to the cerebello-frontal loops; and (iii) the speed of information processing, a mainly subcortical function.
