Abstract The H-subunit of the photosynthetic reaction center of Rhodobacter capsulatus was synthesized in vitro by programming a R. capsulatus cell-free translation system with isolated H-protein mRNA. When intracytoplasmic membrane vehicles of R. capsulatus were added cotranslationally, newly synthesized H-protein sedimented on sucrose gradients with the membranes. The majority of the membrane-associated H-protein was resistant towards extraction with 6 M urea or 0.2 M Na2CO3 at pH 10.5. The urea-extractibility, however, increased significantly when membranes had been treated with trypsin prior to the insertion of the H-subunit, suggesting that the membrane assembly of the H-protein is dependent on (a) proteinaceous factor(s).
