    Notes: Abstract Advances in the elaboration of novel genomic types of beta-galactosidase-positive Enterobacteriaceae and comprehensive studies of their habitats have resulted in an innovative approach to the assessment of the merits and shortcomings of the thermotrophic and fecal species Escherichia coli and all other coliforms as markers of the microbiological safety of water. As one of the consequences, it is recommended to abolish the "technical" designation fecal coliforms because their current method of detection will result in the isolation of thermotrophic organisms that have been demonstrated, beyond a doubt, to be of environmental, rather than uniquely enteric origin. Additional population studies have demonstrated that none of the coliforms can function as reliable markers for all enteric pathogens (index organisms sensu Ingram), nor be of use in validating adequate processing for safety of raw water, which represents the indicator function of markers, as defined by Ingram. Future studies along these lines will have to provide the data required to assess the suitability of additional markers for the reliable monitoring of drinking water for microbiological safety.

    Notes: Abstract Many states within the United States as well as many other countries have promulgated regulations addressing public health, consumer protection, and truthin-labeling aspects of the extraction, bottling, and labeling of commercially bottled springwater intended for public consumption. Many of these regulations are inconsistent, suggesting a need for more uniform standards in acceptable extraction methods and legal/technical definitions of “spring” and “springwater.” An objective of the extraction or collection method is protecting the quality and integrity of the springwater, especially against microbial contamination. A summary of microbiological issues associated with groundwater and springwater is presented. Acceptable extraction methods can be either surface collection boxes/houses at the discharge point of the spring or a subsurface borehole or gallery interception system. Although extraction wells can provide total protective isolation of the water, a potential concern with that method is providing assurance that the extracted water is in fact the same water that feeds the adjacent spring. Criteria for testing this requirement are suggested in the paper.

    Notes: Abstract During the past eight decades, a large number of studies have examined the possible relationship between blood type and infection. Many publications reflect uncritical attempts to mathematically link unstratified or random data. The interaction of pathogen and erythrocyte membrane may reflect antigenic similarity, adhesion through specific receptors, or modulation of antibody response. Anthropological surveys suggest that the geographic and racial distribution of human blood groups reflects susceptibility of populations with specific blood types to the plague, cholera, smallpox, malaria and other infectious diseases.

    Notes: Abstract The gelatin test has been utilized for many years as a characteristic to separate the genusSerratia from other members of the tribe Klebsielleae. Gelatin is a large protein matrix that cannot diffuse into bacterial cells. Microbes that attack gelatin do so by producing extracellular proteinases. The measurement of gelatinase has suffered from the lack of a defineable endpoint and the inability to quantitate the enzyme. A method was developed utilizing an azodye-labelled collagen substrate that could measure the extracellular proteinase ofSerratia. The test was easy to perform, inexpensive, and potentially quantifiable. The azodye test corresponded completely with the gelatinase tests.

    Notes: Abstract Bile-esculin medium has been used for many years for the presumptive identification of group D Streptococcus. The test is based on the ability of a bacterium to grow in the presence of 40% bile and produce esculinase. 2935 strains of Enterobacteriaceae were inoculated onto bile—esculin agar slants and incubated at 35 C. Esculin hydrolysis was determined after 24 and 48 hours. At 24 hours of incubation esculin hydrolysis was limited to the generaKlebsiella, Enterobacter, Serratia, and the speciesP. vulgaris, P. rettgeri, andC. diversus. Not all strains of these species were positive, however. All other members of the family were negative. At 48 hours of incubation 37% ofE. coli gave a positive reaction; all other Enterobacteriaceae which were negative at 24 hours remained negative. Esculin hydrolysis is a valuable test for the taxonomic classification of the family Enterobacteriaceae.
