    Notes: The N-acetylcysteine, cysteine and gluathione conjugates of r-7,t-8-dihydroxy-t-9,10-oxy-7,8,9,10-tetrahydrobenzo[a]pyrene as well as sulphuric and glucuronic acid conjugates of hydroxybenzo[a]pyrenes have been studied by fast-atom bombardment mass spectrometry (FABMS). Dynamic FABMS has been shown to be advantageous over conventional static FABMS giving more intense spectra with less material in both positive-and negative-ion modes. The spectra show quasmolecular ions and fragment ions indicative of the aglycome and/or the conjugating group.

    Notes: Urinary metabolites of bis(2-ethylhexyl) phthalate were studied using fast-atom bombardment mass spectrometry (FAB-MS). It was found that the ß-glucosides of mono(2-ethyl-5-hydroxyhexyl) phthalate and mono(2-ethyl-5-oxohexyl) phthalate gave radical molecular unions, M -·, under negative-ion FAB conditions. This was also shown to occur for the methyl ester of mono(2-ethylhexyl) phthalate ß-glucuronide, an artefact formed during the work-up procedure.

    Notes: Taurine-conjugated bile acids are an important group of biological metabolites. When investigated by negative-ion fast-atom bombardment collision-induced dissociation mass spectroscopy they show charge-remote fragmentations of the [M—H]- pseudomolecular ion. These fragmentations provide information on the positions of ring substituents remote from the charge site. In the present work we have compared the negative-ion fast-atom bombardment collision-induced dissociation spectra of six different conjugated bile acids.

    Notes: Mono(2-ethylhexyl) phthalate was given to guinea pigs and mice and its metabolites were isolated from urine. A procedure consisting of sorbent extraction, ion-exchange chromatography and reversed-phase high-performance liquid chromatography was used in the purification scheme. The metabolites were analysed by fast atom bombardment mass spectrometry. It is concluded that the purification procedure is very mild and that fast atom bombardment is a useful ionization technique for intact conjugates of metabolites of bis(2-ethylhexyl) phthalate.

    Notes: The conjugated bile acids taurochenodeoxycholate and taurodeoxycholate are indistinguishable from analysis of their fast-atom bombardment spectra. Both the positive- and negative-ion spectra show prominent pseudomolecular ions, but little fragmentation. Collision-induced dissociation of the negative pseudomolecular ions reveal structural differences between the two isomers which are evident in the resultant daughter-ion spectra. Thus the two isomers can be differentiated.

    Notes: Rats, germfree and conventional, were dosed with 14C-labelled benzo[a]pyrene. Faeces and urine were collected. Metabolites in faces were effectively extracted with a new method using a combination of solvents and solid sorbents. Metabolites in urine were extracted with octadecylsilane-bonded silica. The metabolites were fractionated into groups by chromatography on a cation exchanger (SP-LH-20 or SP-Sephadex C-25) and an anion exchanger (TEAP-LH-20). Some of the groups were further purified by column chromatography and analysed by HPLC and TLC. The analyses show a complex pattern of metabolism. A large part of the metabolites (9-24% depending on animal type and route of excretion) had amphoteric properties, e.g. like glutathione and cysteine conjugates. The abundance of conjugates sensitive to β-glucuronidase and sulphatase was low. The relative amount of acidic conjugates in faeces was much higher in the germfree than in the conventional rats indicating the influence of the intestinal flora on the metabolism. The results support the view that the mercapturic acid pathway is a quantitatively important metabolic route for benzo[a]pyrene in rats. The methods of extraction and group fractionation were designed to be generally applicable to the analysis of lipophilic xenobiotics and their metabolites.

    Notes: Substantial evidence has suggested that a nonsterol product of mevalonic acid (MVA) is essential for the initiation of DNA synthesis in mammalian cells. Several possible isoprenoid candidates have been suggested, but the identity of this compound still remains unknown. In this study we have isolated and purified MVA products from SV40-transformed human fibroblasts and identified fractions with a growth-stimulatory effect. The cells were labelled with [14C]MVA in the presence of inhibitors of 3-hydroxy-3-methylglutaryl coenzyme A (HMG-CoA) reductase. After lipid extraction, the [14C]MVA-labelled lipids were subjected to high performance liquid chromatography and size-exclusion chromatography, and the effect of the fractionated eluate on the DNA synthesis of arrested MVA-depleted target cells was tested. Thereby we found a fraction of [14C]MVA-labelled lipids with a substantial stimulatory effect on DNA synthesis. The chromatographic behavior suggested that the growth-stimulating fractions contained dolichol-20. This was confirmed by mass spectrometric analysis. Similar results were obtained when lipids from hepatocellular carcinoma cells and a sample from breast tumor were isolated and analyzed by the same procedure. The mechanisms by which these compounds induce DNA synthesis are unknown. Recent data obtained in our laboratory have provided evidence that dolichyl groups are covalently linked to tumor cell proteins, which implicates a new biological function for long-chain polyisoprenoid alcohols (Hjertman et al. [1997] FEBS Lett 416:235-238). In this study we demonstrate that tumor cells containing dolichol-like growth-stimulatory lipids also contained dolichylated proteins. This raises the question whether the growth-stimulatory dolichol-like lipids serve as substrates for the dolichylation reaction. J. Cell. Biochem. 71:502-514, 1998. © 1998 Wiley-Liss, Inc.
