    Notes: Summary A simple microprecipitin test employing phase contrast microscopy has been described for poliovirus antigens and antisera. The antigens, consisting of concentrates of the 3 types of poliovirus, exhibited typespecific reactions in high titer with rabbit antisera heated twice at 56° C for 30 minutes, heterotypic crossings occuring, if at all, only at low dilutions of the sera. Potent antigens were also obtained from attenuated virus strains. Highly purified Type 1 (Mahoney) virus (fraction D) reacted with rabbit antiserum in a similar manner as the crude virus concentrate. Heating of precipitin antigens of all 3 types at 56° C for 30 minutes resulted in almost complete destruction of their reactivity. Positive precipitin reactions were not obtained with the serum of monkeys hyperimmunized with Type 1 poliovirus; with monkey antisera for Types 2 and 3 polioviruses positive reactions occurred only in a very limited zone, which disappeared after two repeated heatings at 56° C for 30 minutes. Precipitin reactions were readily obtained with certain human sera. Heating of the human sera partly reduced their precipitin titers. Prolonged incubation of the reaction mixtures increased the precipitin titers. Un concentrated virus from tissue culture fluids (monkey kidney, ERK) proved to be an unsatisfactory precipitin antigen in spite of its high infectivity titer.

    Notes: Summary The occurrence and development of microprecipitin antibodies to poliovirus antigens was investigated with sera derived from triplenegative persons, triple-positive naturally, immune persons, persons immunized by killed or live poliovirus vaccines, persons with clinical poliomyelitis caused by each of the 3 types of poliovirus, and persons infected with various Coxsackie and ECHO viruses. No positive reactions were obtained in sera of 11 triple-negative children and adults. Seven of 44 (16 per cent) sera from healthy, naturally immune children and adults gave positive reactions. Sera from persons who ingested live, attenuated poliovirus vaccine all developed precipitin antibody, which however was detected later than the neutralizing antibody. The response to 4 doses of Salk vaccine was less frequent and marked, and occurred most often in persons who had naturally acquired neutralizing antibodies for the corresponding type of poliovirus prior to vaccination. Thirty-two of 36 patients with poliomyelitis developed a positive precipitin reaction which was already, as a rule, present in maximum titers at the end of the first week after onset of illness. Precipitin antibody titers began to decline within a few weeks after onset of illness. Similar rapid declines were observed after infection with live virus vaccine, and after inoculation of Salk vaccine. The polio-precipitin reaction was type-specific in patients apparently infected for the first time with poliovirus, but heterotypic reactions occurred frequently in patients with pre-existing heterotypic neutralizing antibodies. There was some indication that Coxsackie A 9 and the polioviruses may share a minor, common microprecipitin antigen. The available data indicate that the microprecipitin antibody is different from both the neutralizing and complement-fixing antibodies. The advantages and limitations of the polio-precipitin test as a diagnostic procedure are discussed.

    Notes: Abstract Male 8 to 20-week-old NMRI mice (an outbred strain) infected with the encephalomyocarditis virus (EMCV) plaque variant (PV) 7 consistently develop a distinct myocarditis with a relatively low mortality (21%). Myocarditis occurs in essence independent of the virus dose applied, and other internal organs are not affected. Nevertheless, 3.5-week-old NMRI mice perished within 5 days of virus inoculation and exhibited disseminated myofibrillar degeneration (MFD); this obviously virus-induced myocardial damage was accompanied by scanty inflammatory infiltrates. EMCV PV7 infection of adult male C57B1/6 and DBA/2 mice causes myocarditis comparable to that seen in NMRI mice. In DBA/2 mice, however, the virus-induced myocardial necrosis is complicated by subtotal calcification. This strain has a genetically determined “spontaneous” calcification of the myocardium, as shown by the study of uninfected controls. EMCV PV7-infected NMRI mice appear a promising model for study of long-term effects of viral myocarditis, possibly including cardiomyopathy. Furthermore, this outbred mouse strain offers the possibility of examining the pathogenesis of direct viral cytolysis and its relation to MFD as well as immunologically mediated cell damage.
