    Notes: Summary The red anthraquinone derivative isolated from several Drechslera and Curvularia species has been identified as cynodontin (1,4,5,8-tetrahydroxy-2-methylanthraquinone), and not as 1,4,5,8-tetrahydroxy-2,6-dimethylanthraquinone as reported earlier. Both pigments showed similar physico-chemical properties. A good distinction could be obtained by gas-liquid chromatography.

    Notes: Abstract A procedure is described to culture microorganisms under aerobic and anaerobic conditions in infusion bottles internally coated with a solid agar medium, in view of headspace sampling of volatile components or extraction of cellular constituents. This method is far less laborious than conventional petri-dish procedures and reduces the risk of contamination. Biochemical analysis of microorganisms usually involves time-consuming and laborious procedures including sterilization of media, inoculation, culturing, killing, harvesting, washing, freeze-drying, extraction and, finally, sampling for separation and characterization of components (Booth, 1971; Constantinescu, 1974). To simplify the procedure, a ‘single bottle technique’ was introduced for liquid media (Weijman, 1978), using regular infusion bottles as applied in blood banks and hospitals. The present paper describes a similar technique for use with agar-containing solid media, as an alternative for or an addition to the use of petri dishes, Roux bottles or roll-tubes (Hirsch, 1948). Procedure: Infusion bottles made of borosilicate glass were used with the following specifications: 500 ml capacity, 21 cm in height and 7.7 cm outside diameter. Bottles containing 50–150 ml medium with 3% agar (w/v) were autoclaved for 20 min at 120°C and cooled to ambient temperature. During cooling the bottles were rotated in a horizontal position at about 50 rpm to allow smooth and gradual coating of the inner surfaces. Subsequently, the flasks were inoculated and incubated (Fig. 1). The bottles can be closed with cotton wool plugs

    Notes: Abstract 72 Strains belonging to 44 species of ascomycetous black yeasts were analyzed for their coenzyme Q systems. Prevalent were Q-10 and dihydrogenated Q-10 systems. Members of the Dothidealean suborder Dothideineae have Q-10 (H2), while those belonging to the suborder Pseudosphaeriineae mostly have Q-10. The anamorph genus Exophiala Carmichael and the teleomorph genus Capronia Sacc. seem to be heterogenous.

In dit artikel worden nieuwe diagnostische benaderingen beschreven die gebaseerd zijn op de detectie van specifieke ionen door middel van pyrolyse-massaspectrometrie (Py-MS), direct-probe-massaspectrometrie (DIP-MS) en gaschromatografie-massaspectrometrie (GC-MS). De voor- en nadelen van deze methoden worden behandeld en vergeleken met de dunnelaag-chromatografische methode. GC-MS is een veelbelovende methode vanwege de grote specificiteit, gevoeligheid en geschiktheid voor automatisering. Op den duur kunnen de MS-methoden leiden tot een verkorting van de totale expertisetijd.
    Notes: Abstract Current diagnosis of gangrene in potato tubers, caused byPhoma exigua var.foveata, is based on isolation and identification of the causal organism or on thin-layer chromatographic (TLC) detection of the anthraquinone pigment pachybasin (1-hydroxy-3-methylanthraquinone). In this paper alternative diagnostic approaches are described based on detection of specific ions by pyrolysis mass spectrometry (Py-MS), direct probe mass spectrometry (DIP-MS) and gas chromatography-mass spectrometry (GC-MS). The advantages and disadvantages of these methods are discussed in comparison with the TLC approach. GC-MS is a promising technique because of its high specificity, sensitivity and suitability for automation. Moreover, the MS approaches may lead to a reduction of total expertise time.
