Introduction of Digitoxose Units and Directed β-Linkages in Syntheses of 2′-DeoxydisaccharidesBy condensation of methyl digitoxoside (1) and triacetylglucal (5) following the N-iodosuccinimide procedure predominantly α-linked disacharides are obtained, yielding more of the α, 1 → 3 than of the α, 1 → 4 derivative 6 and 10, respectively. An orthoester reaction of 1 gives a mixture of 3 and 4, which under comparable conditions leads to more α, 1 → 4 and less α, 1 → 3 disaccharide 11 and 7, respectively. In both these reactions the presence of the corresponding β-compounds can be demonstrated. - The conformational equilibrium of methyl diacetylglucuronal (15) in solution is evidently shifted towards the 5H4(D) half-chair conformation. Its N-iodosuccinimide condensation with (3 + 4) gives in addition to little of the α, 1 → 4 compound 16 the β, 1 → 3 and α, 1 → 3 disaccharide 17 and 18 in a ratio of approximately 3:2. Modification reactions with 18 lead to the tetradeoxydisaccharide obtained as a crystalline triacetate 24. - In the linkage of 15 with the allo epoxide 25 the β, 1 → 4 disaccharide 27 and its α, 1 → 4 anomer 26 are prepared in the ratio of approximately 3:2. Reductive dehalogenation with concomitant ester reduction of 27 gives 29 and further selective tosylation 31. By simultaneous iodine exchange at C-6′ and nucleophilic epoxide opening in the reducing ring the isomers 32 and 33 (2:1) result. With a final reductive deiodination of 32 the synthesis of methyl 4-O-(β-D-olivosyl)-α-D-digitoxoside (35) is achieved. 35 is compared with derivatives of the terminal disaccharide lactone from flambamycin.

Durch Kondensation von Methyldigitoxosid (1) mit Triacetylglucal (5) nach dem N-Iodsuccinimid-Verfahren werden vorwiegend α-verknüpfte Disaccharide erhalten, wobei mehr α, 1 → 3- als α, 1 → 4-Verbindung 6 bzw. 10 anfällt. Das durch Orthoester-Reaktion aus 1 gewonnene Gemisch der Monoacetate 3 und 4 ergibt unter vergleichbaren Bedingungen überwiegend α, 1 → 4- und weniger α, 1 → 3-Derivat 11 bzw. 7. Bei beiden Umsetzungen lassen sich die entsprechenden β-Verbindungen in geringer Menge nachweisen. - Im Fall von Methyldiacetylglucuronal (15) liegt das Konformerengleichgewicht in Lösung deutlich auf der Seite der 5H4(D)-Halbsesselkonformation. Seine N-Iodsuccinimid-Kondensation mit (3 + 4) gibt neben wenig α, 1 → 4-Verbindung 16 das β, 1 → 3- und das α, 1 → 3-Disaccharid 17 bzw. 18 im Verhältnis von ca. 3:2. Ausgehend von 18 werden Modifikationsreaktionen vorgenommen, die das Tetradesoxydisaccharid als kristallines Triacetat 24 liefern. - Bei der Verknüpfung von 15 mit dem allo-Epoxid 25 werden die β, 1 → 4-und die α, 1 → 4-Verbindung 27 bzw. 26 im Verhältnis von ca. 3:2 dargestellt. Reduktive Dehalogenierung sowie Esterreduktion von 27 führen zu 29 und anschließende selektive Tosylierung liefert 31. Durch simultanen Iodaustausch an C-6′ und nucleophile Epoxidöffnung im reduzierenden Ring werden die Isomeren 32 und 33 (2:1) erhalten. Mittels abschließender reduktiver De-Iodierung von 32 erreicht man die Synthese von Methyl-4-O-(β-D-olivosyl)-α-D-digitoxosid (35), das mit Derivaten des terminalen Disaccharidlactons aus Flambamycin verglichen wird.
