    Notes: Abstract Sex pheromone traps forArgyrotaenia pulchellana Haw. (Lep. Tortricidae) were loaded with two low pheromone doses (0.1 and 0.01 mg) and tested in apple and pear orchards of northern Italy's Emilia-Romagna Region to estimate the relationship between male catches and subsequent larval infestation. While the lower dose captured fewer males, it registered an enhanced correlation to larval populations and a decrease in the number of episodes of high catches associated with no infestation. In all likelihood, the pheromone traps baited at the lower dose have a smaller active space and captured only makes within the orchard. Lures with 0.01 mg of pheromone can be replaced every 6 weeks, just as the 1 mg lures now in widespread use in Italy. A threshold of 65 adults captured from the beginning of flight is proposed for the Emilia-Romagna IPM project.

    Notes: Summary Uridine-H3 was incorporated into meristem tips of both healthy and PLRV-infected potato plants, of the cultivars, Majestic and Primura. Autoradiograms of tips pretreated with actinomycin D showed that 13 of 14 and 11 of 14 respectively, were virus free in the dome and first 4 leaf primordia, 1 and 3 were infected in the 4th leaf primordium, and 1 of Primura also in its 3rd primordium. The highest plantlet yield from cultures in vitro of meristem tips that included 4 leaf primordia (7 to 9 plantlets per meristem), was obtained by growing them in sequence on 3 sequential media, each based on Murashige and Skoog basic medium complemented with various hormones. The 2nd medium, containing benzylaminopurine (0.5 mg/l) and gibberellic acid (0.5 mg/l), elicited callus and the subsequent formation of adventitious buds. Of the plantlets of the cvs Vivaks, Primura and Majestic, 88, 91 and 100 %, respectively, were PLRV-free. Propagation in vitro, by the single-node cutting technique and tuberlet production, were successful.

    Notes: Summary Using sensitive Dot-ELISA, potato virus S (PVS) and potato virus M (PVM) contents per single potato meristem tip of 4-leaf primordia were determined, and were correlated with the percentage of virus-free plantlets obtained by meristem-tip culture preceded by chemo-and thermotherapy of the plants. Treatment of PVS-infected cv. Majestic plants for 26 days at 35°C resulted in 85% of the meristems having less than 10pg of virus, and 89% of the derived progenies were similarly virus-free. When stem cuttings were dipped for 15 days in various chemicals, the mixture of 5 mg/l ribavirin+5mg/l DHT was the most effective. For cvs Majestic and Primura, 90 and 72% of the meristems had less than 10pg of virus, and resulted in 74 and 67% virus-free progenies respectively. In PVM infection of the two cultivars, the same mixture yielded 70 and 66% meristems with less than 10pg of virus, and 69 and 68% virus-free progenies.

    Notes: Summary The potato leafroll virus (PLRV) coat protein (CP) gene was directly cloned from the total RNA extracted from virus-infected plants. First strand cDNA synthesis was not necessarily specific; it was equally efficient using either random or CP-specific primers. The viral sequence encoding the coat protein was specifically amplified from the total population of cDNA molecules by polymerase chain reaction (PCR), using specific primers bordering the CP gene. The unique amplified product thus obtained was cloned blunt-end into the pT7T318U plasmid vector, and the authenticity of the cloned gene verified by sequence analysis. This cloning strategy obviates the need for virus purification. Sequence comparison of the CP gene of the Italian isolate and those of five other PLRV isolates revealed a close similarity to the three European and the Canadian isolates, and a more distant relationship with the Australian one.
