    Notes: Serial measurements of peripheral plasma concentrations of progesterone and oestradiol-17β were made in 23 women with singleton pregnancies who were given intravenous salbutamol in an attempt to inhibit pre-term labour. Mean levels of both hormones fell significantly during salbutamol infusion but returned to pre-treatment levels after therapy was stopped. Plasma human placental lactogen concentrations did not change during salbutamol infusion, suggesting that the fall in steroid hormone levels was unlikely to be an effect of altered uterine or placental blood flow.

    Notes: Prostaglandin E (PGE), prostaglandin F (PGF) and 13, 14-dihydro-15-keto-prostaglandin F (PGFM) have been measured in umbilical cord plasma obtained immediately after delivery of the baby before clamping of the cord. In general the prostaglandin levels followed the pattern PGFM〉PGE〉PGF. A significant arterio-venous difference was demonstrated only for PGE with raised venous levels (P〈0.01). In cord blood samples obtained from infants whose mothers had received epidural anaesthesia, no arterio-venous difference for PGE could be demonstrated although the mean levels were not significantly different from controls. The concentrations of prostaglandins in Umbilical cord plasma proximal to the placenta were found to rise continuously from the time of delivery of the baby with no significant changes after cord clamping or placental delivery. The possible physiological significance of these findings is discussed.

    Notes: Concentrations of prostaglandin F (PGF) have been measured, by radioimmunoassay, in samples of amniotic fluid from patients with normal pregnancies. Amniotic fluid contained significantly more PGF at term prior to the onset of labour than in mid-pregnancy (p〈0.01), and significantly more during spontaneous labour, at a cervical dilatation of three cm. or less, than before the onset of labour (p〈0.001). During spontaneous labour PGF concentrations in amniotic fluid varied between 1.2 and 39.5 ng. per ml., and increased significantly with advancing cervical dilatation (p〈0.001). Levels of PGF in the early first stage of oxytocin-induced labour were higher than the levels obtained before the onset of labour (p〈0.025), but lower than those obtained during spontaneous labour (p〈0.05). It was demonstrated, by gas liquid chromatography with electron-capture detection, that PGF2α is the major PGF component in amniotic fluid during labour.

    Notes: Progesterone concentrations have been determined in a total of 175 peripheral plasma samples obtained serially from 20 women undergoing mid-trimester abortion. In ten patients abortion was induced by intra-amniotic administration of 50 mg. of prostaglandin F2α (PGF2α) alone; in ten others it was induced with 50 mg. of intra-amniotic PGF2α with 80 g. of urea. Mean instillation-abortion intervals were 29.0 hours in the group receiving PGF2α alone and 12.1 hours in the groups receiving PGF2α and urea. In eight of the patients treated with PGF2α and urea, and in four of those treated with PGF2α alone, there were significant decreases in progesterone level (determined by calculation of correlation coefficients) during the instillation-abortion interval. When plotted against instillation-abortion times, calculated rates of decrease in peripheral plasma progesterone levels showed a statistically significant regression (p〈0.05). This indicates that progesterone levels drop most rapidly in patients with shorter instillation-abortion times. There was no relationship between initial progesterone levels and instillation-abortion times.

    Notes: Summary Polyclonal rabbit antisera raised against oxytocin, bovine neurophysin I and vasopressin were used, together with an immunogold complex, to localise the peptides in ultrathin sections of ovine corpus luteum. The only organelle which consistently showed gold labelling was the secretory granule of the large luteal cell. In non-consecutive sections of the same large luteal cell all the granules showed a similar level of labelling after oxytocin or neurophysin I antisera: however no immunolabelling was detected for vasopressin. Oxytocin and neurophysin seem to be rapidly lost after secretion since exocytosed granule cores showed no labelling above background levels.
