Selective Substitutions in C-SilylpyrazolesThe addition of diazomethane or diazoacetic ester to mono(trimethylsilyl)acetylene (3) leads to 3(5)-(trimethylsilyl)pyrazole (8) or 5(3)-(ethoxycarbonyl)-3(5)-(trimethylsilyl)-pyrazole (9). Moreover the reaction of diazomethane with acetyl(trimethylsilyl)acetylene (4) was found to give 4-acetyl-3(5)-(trimethylsilyl)pyrazole (10) and 3(5)-acetyl-4-(trimethylsilyl)pyrazole (11) in 70 and 30% yield, respectively. These results can be considered as evidence for the directive influence of the trimethylsilyl group. - C-Silylpyrazoles can be desilylated selectively: replacement of silyl groups in 4-position by hydrogen is smoothly affected by conc. sulfuric acid. In this way 8, 9 and 14-16 are obtained in high yields. - Treatment of 5 with conc. nitric acid in a certain range of temperature yields 8 as well as the hitherto unknown 4-nitro-3(5)-(trimethylsilyl)pyrazole (18), the latter resulting from replacement of the silyl group in 4-position by a nitro group. 18 is quantitatively formed from 8 by nitrating acid. - Silyl groups in 3(5)-position, but not those in 4-position, are removed by nucleophilic agents. In this way 12, 15, and 19-22 are formed using sodium ethoxide or alkali hydroxide.
Notes:
