    Notizen: Several evaluation methods for soybean chilling tolerance at the reproductive stage were examined under artificial climatic conditions. Comparisons were made on plants in control and treated plots using three cultivars differing in the level of chilling tolerance. In all methods, plants were grown at 22/17°C (day/night) until first flowering, and then transferred to growth chambers at 24/17°C for control and 15/15°C for chilling treatment, respectively. A method, in which plants were grown at 20/16°C after 4 weeks of the different temperature treatments, proved comparable to the conventional one, in which chilling tolerance at the flowering stage is evaluated using natural and artificial conditions. Another method, in which the plants in chilling treatment plots were grown at 15°C until maturity, also proved usable to evaluate genotypic differences in chilling tolerance independently of maturity time.

    Notizen: Although it is known that the compositions of low-molecular weight glutenin subunits (LMW-GSs) are important factors for bread-making quality of wheat, it is still not clear which LMW-GSs confer improved bread-making quality and how those LMW-GSs interact with high-molecular weight (HMW) GSs. Using a hard red winter wheat line with good bread-making quality and a Japanese wheat cultivar with poor quality as well as their progeny we identified LMW-GSs associated with the bread-making quality. One such LMW-GS, KS2, which had a molecular weight of 42 kDa and was allelic to HS1, was associated with bread-making quality. Furthermore, by using four recombinant inbred lines with different HMW-GS and LMW-GS combinations, KS2 and HMW-GS 5+10 showed interaction effects on the bread-making quality. Two-dimensional polyacrylamide gel electrophoresis (Page) analysis showed that KS2 consists of two protein components and that HS1 is composed of three components. The N-terminal amino acid sequences of the five components were identical to the most frequently analysed sequence of LMW glutenin components.

    Notizen: Plant regeneration was achieved through anther culture of three wild species of Hordeum (H. murinum, H. marinum and H, bulbosum). Calli or embryoids were formed from microspores in anthers cultured on a medium containing 6-benzylammopurine (BAP) and ficoll. These calli or embryoids regenerated green or albino shoots and roots after transfer to regeneration media. Green plantlets which developed on regeneration media were transferred to soil where they showed further growth.

    Notizen: Abstract We report the generation of transgenic barley plants via PEG-mediated direct DNA uptake to protoplasts. Protoplasts isolated from embryogenic cell suspensions of barley (Hordeum vulgare L. cv ‘Igri’) were PEG-treated in a solution containing a plasmid which contained the neomycin phosphotransferase (NPT II) gene under the control of the rice actin promoter and the nos terminator. Colonies developing from the treated protoplasts were incubated in liquid medium containing the selective antibiotic G418. Surviving calli were subsequently transferred to solid media containing G418, on which embryogenic calli developed. These calli gave rise to albino and green shoots on antibiotic-free regeneration medium. NPT II ELISA revealed that approximately half of the morphogenic calli expressed the foreign gene. In total, 12 plantlets derived from NPT-positive calli survived transfer to soil. Southern hybridization analysis confirmed the stable transformation of these plants. However, the foreign gene seemed to be inactivated in plants from one transgenic line. Most of the transgenic plants set seed, and the foreign gene was transmitted and expressed in their progenies, which was ascertained by Southern hybridization and NPT II ELISA.
