    Notes: The first measurements of vibrational Raman optical activity (ROA) have been performed employing a forward scattering geometry. Forward-scattered ROA spectra of (+)-trans-pinane and (-)-β-pinene are presented as first examples. Comparison with the corresponding backward-and right-angle-scattered spectra confirmed that the bond polarizability theory of ROA is a good approximation for trans-pinane, and has identified unequivocally a large isotropic contribution to the ROA in β-pinene bands at 716 and 765 cm-1 ascribed to interactions between the olefinic methylene twist and a skeletal mode of the pinane structure. Unless large isotropic contributions are present, which is unusual, forward scattering is by far the least favourable of the ROA measurement strategies, which might explain the failure of recent attempts to observe ROA in coherent anti-Stokes Raman scattering.

    Notes: The vibrational Raman optical activity (ROA) spectra of the simple amino acids L-serine, L-cysteine, L-valine, L-threonine and L-isoleucine in aqueous solution between 600 and 1600 cm-1 measured in backscattering are presented and discussed. Complete vibrational assignments are suggested for serine and cysteine and limited assignments for the others. Bands originating in Cα*H deformations and the symmetric CO2- stretch appear to provide the best signatures for stereochemical studies. The results show that ROA provides a new perspective on amino acid conformation in aqueous solution.

    Notes: A simple and fully computer-controlled backscattering incident circular polarization Raman optical activity (ICP ROA) instrument suitable for chioptic studies of biologically significant molecules in aqueous solution is described. It consists of a fast single-stage monochromator equipped with a holographic edge filter, a highly efficient holographic difiaction grating and a thinned back illuminated thermoelectrically cooled charge-coupled device detector as the key spectral elements. A longitudinal electro-optic modulator is employed to switch between orthogonal circular polarization states in the incident laser radiation within a backscattering geometry. A thick Lyot depolarizer is used for the depolarization of the backscattered Raman radiation. High-quality backscattering ICP ROA spectra of (1S)-( - )-trans-pinane, (1R)-( + )-trans-pinane, L-alanine, L-alanyl-L-alanine, lysozyme, D-glucose, α-D-cyclodextrin and 2′-deoxycytidine are presented as typical examples of the excellent performance characteristics.
