    Notizen: Abstract We describe a microfluidic device for separating cells according to their dielectric properties by combining 2-dimensional dielectrophoretic forces with field-flow-fractionation. The device comprises a thin chamber in which a travelling-wave electrical field is generated by a planar, multilayer microelectrode array at the bottom. Under the balance of gravitational and dielectrophoretic levitation forces, cells introduced into the device are positioned at different equilibrium heights in a velocity profile established inside the chamber, and thereby transported at different velocities by the fluid. Simultaneously, cells are subjected to a horizontal travelling-wave dielectrophoretic force that deflects them across the flow stream. The 2-dimensional dielectrophoretic forces acting on cells and the associated velocities in the fluid-flow and travelling-field directions depend sensitively on cell dielectric properties. The responses of cultured MDA-435 human breast cancer, HL-60 human leukemia and DS19 murine erythroleukemia cells, and of peripheral blood mononuclear (PBMN) cells were studied as functions of the frequency and voltage of the applied electric signals, and of the fluid flow rate. Significant differences were observed between the responses of different cell types. Cell separation was demonstrated by the differential redistribution of MDA-435 and PBMN cells as they flowed through the device. The device can be readily integrated with other microfluidic components for microscale sample preparation and analysis.

    Notizen: The dicarbonyl methylglyoxal reacts with the lysine residues of proteins to give brown colored products with a small oxygen-dependent free radical content. Electron spin resonance (ESR) experiments employing ethylene diamine as a model for protein lysine residues are reported. The prevention of imine formation between methylglyoxal and the amine models either by pretreatment of methylglyoxal with glutathione or by methylation of the amine led to brightly colored reaction products and a much higher radical content. From an analysis of the ESR spectra it is clear that the reactions between methylglyoxal and the simple model amines developed far beyond those in the protein-methylglyoxal systems. It is proposed that this difference is accounted for by the enormous steric effects of the proteins. The relevance of the observations to the “browning” reaction in foods is briefly discussed.

    Notizen: The ability of three cultured cell lines (NRK, 6M2, and 54-5A4) derived from rat kidney to quench a population of ascorbyl and 2,6-dimethoxy-p-semiquinone free radicals has been investigated by electron spin resonance spectroscopy. The radical scavenging action of normal rat kidney (NRK) cells was the weakest, that of 6M2 cells (reversibly transformed phenotype) was four times stronger than NRK, and that of 54-5A4 cells (irreversibly transformed mutants of 6M2) was 10 times greater than NRK. Free radical quenching experiments were also performed on Chinese hamster ovary (CHO) cells. A much slower scavenging rate was observed for CHO cells of normal phenotype grown in the presence of dibutyryl cyclic AMP than was found for cells of transformed phenotype grown in its absence. The free radical quenching kinetics of the various cell lines studied directly paralleled their state of transformation.

    Notizen: The proton hyperfine coupling constants for the family of methoxyl-substituted semiquinones in aqueous solution have been determined by electron spin resonance (ESR) spectroscopy. These data and the electrochemical midpoint potentials for the corresponding series of substituted quinone/hydroquinone couples are discussed. Neither the additivity principle describing quinoidal hyperfine coupling constants nor the Hammett substituent relationship predicting electrochemical behavior are obeyed by these methoxyl-substituted quinones. It is shown, however, that the experimental data can be rationalized in terms of neighboring group interactions that occur between adjacent methoxyl substituents. Such perturbations apparently lead to a breakdown of hyperconjugation between the electronic bonding structure of the quinone ring and the methyl group of the methoxyl moiety. It is pointed out that ubiquinone is a dimethoxyl compound whose properties are determined by such neighboring group interactions. This observation may have important consquences for the fabrication of ubiquinone analogs and quinone-based drugs.

    Notizen: The electrophoretic mobilities (EPMs) and semiquinone reductase activities of two clones of Friend murine erythroleukemia (MEL) cells were investigated as a function of treatment with the inducing agents dimethylsulfoxide (DMSO) and hexamethylene bisacetamide (HMBA) As reported previously by others, the inducible clone DS19 lost its ability to grow in soft agar and expressed hemoglobin as judged by benzidine/H2O2 staining after 96 hours of treatment with 1% DMSO or 4 mM HMBA. In addition, its EPM fell by 14%, its semiquinone reductase activity by 40%, and its mean diameter by 10%. The second clone, R1, retained its ability to grow in soft agar and lacked hemoglobin expression after treatment with HMBA and DMSO, characterizing it as noninducible. However, R1 did demonstrate alterations in EPM, semiquinone reductase activity, and cell diameter that closely paralleled those found in DS19. Such responses were not seen in three non-MEL cell lines exposed to HMBA or DMSO, suggesting that clone R1 responded to these inducing agents in a cell-line specific manner but that its ability to complete the sequences necessary for differentiation may be blocked at an unknown point distal to the block characteristic of untreated cells. The data show that while a reduction in EPM, semiquinone reductase activity, and cell diameter accompany induced differentiation in MEL cells, such changes can occur in the absence of a commitment to terminal differentiation.
