Des plantules de mais sont cultivées à partir de la graine, à 1'obscurite ct i la lumiere, sur des solutions tamponnées (pH5,8) d'hydrazidemalfrque (HM) de concentrations allant de 10′ a 10−3 M. AUX faibles concentrations (10° et 10−6 M), I'HM stimule la croissance et la formation des racines; aux autres concentrations, it est inhibiteur. L'inhibition de croissance provoquee par I'HM est accompagnee d'une augmentation de poids sec et de la teneur en anthocyanes.L'HM n'influence pas in vitro la destruction de I'auxine par des extraits de mais, mais les extraits de plantes traitees par cette substance degradent l'hormone moins rapidement; parallelement, l'activite catalasique est diminuee mais l'activite péroxydasique est augmented. Le mode d'action de I'HM sur la croissanee et la rhizogenpse, compart à celui de la lumierc, est discute en rapport avec ses effets sur le raetabolisme auxinique.

    Notes: A fully habituated (auxin- and cytokinin-independent) nonorganogenic (HNO) sugarbeet (Beta vulgaris) callus produces very little ethylene as compared with a normal (N) hormone-requiring callus of the same strain. Both callus types react by growth changes to application of inhibitors of ethylene biosynthesis and ethylene action, of 1-aminocyclopropane-1-carboxylic acid (ACC) as the immediate precursor of ethylene, to transfer from light to darkness, and also to application of exogenous ethylene or an ethylene trapper. This indicates their growth dependency upon their endogenously biosynthesized ethylene and also their sensitivity to exogenous gas. However, the sensitivity was generally higher for the HNO callus producing naturally less ethylene. The weaker reaction of the HNO callus to the exogenous ethylene was attributed to its hyperhydric status (a water layer surrounding the cells). Because low ethylene production appears as a general characteristic of habituated cell lines, the causal and/or consequential relationships of this low ethylene production with other characteristics of habituated tissues (absence of exogenous hormones in the culture media, deficiency of cell differentiation, accumulation of polyamines in neoplastic tissues) are discussed.

    Notes: Micropropagated poplar shoots rooted 100% on a rooting medium (A) containing NAA, but they did not root in the absence of auxin (NA). Putrescine, but not spermidine and spermine, promoted rooting up to 42% when added to the NA medium. Cyclohexylamine (CHA), an inhibitor of spermine synthase, also promoted (up to 36%) rooting in the absence of auxin. The inhibitors of polyamine biosynthesis DFMA (α-difluoromethylarginine) and DFMO (α-difluoromethylomithine), aminoguanidine (AG) and methylglyoxal-bis-guanylhydrazone (MGBG), inhibited rooting when applied in the presence of auxin and had no effect in its absence.The rooting inductive phase (in the presence of auxin) was determined by periodical transfer of shoots from A to NA medium, and by changes in peroxidase activity, to be 7 h. Putrescine (not spermidine and spermine) accumulated to a maximum during the inductive phase. Both putrescine and CHA promoted rooting on NA medium when applied during the first 7 h. In contrast DFMA and AG inhibited rooting during this period. The results point to the involvement of putrescine and its Δ1-pyrroline pathway, in the inductive phase of rooting in poplar shoots.

    Notes: A previous paper (Galaud et al. 1993. Physiol. Plant. 87: 25–30) reported a rapid demethylation of DNA after mechanical rubbing of young Bryonia dioica internodes, leading to growth inhibition. In order to assess an eventual causative role of DNA demethylation in the mechanisms underlying thigmomorphogenesis, the effects of the anti-DNA methylation drugs, 5-azacytosine (5-azacyt) and 5-azacytidine (5-azacyd) were investigated. Treatment with 5-azacyd induced a decrease of the methylated DNA level. This was accompanied by an increase of peroxidase activity and of ethylene production. On the contrary, 5-azacyt had no significant effect on growth, peroxidase activity and ethylene production. The use of 5-azacyd as an inducer of DNA demethylation is discussed in relation to the control of gene expression

    Notes: The content of polyamines and proline was much lower in a normal (N) callus of Beta vulgaris L. than in a fully habituated hyperhydric (H) callus. The H callus also contained more glutamate and had a higher glutamate dehydrogenase activity. The excess of glutamate, in this chlorophyll-deficient callus, was linked to accumulation of proline and polyamines. Experiments with α-difluoromethylornithine (DFMO) and α-difluoromethylarginine (DFMA) showed that both ornithine decarboxylase and arginine decarboxylase participated in the synthesis of polyamines (especially spermidine and putrescine) and removal of ammonia. It is hypothesized that the H callus was subjected to ammonia stress from the start of the culture. Experiments with gabaculine, an inhibitor of ornithine aminotransferase, showed that this enzyme linked proline degradation to polyamine synthesis through the production of ornithine. This disturbed nitrogen metabolism appeared to be characteristic of the fully habituated callus and might explain the low growth of this hyperhydric tissue.

    Notes: The content and pattern of soluble isoperoxidases were determined in epidermal explants taken from different internodes of tobacco plants in the vegetative and floral states. There were qualitative and quantitative differences in the isoperoxidases, with a decrease in content and fewer bands being observed acropetally, i.e., in going from the base of the stem towards the apex. Epidermal explants from floral branches were grown in in vitro culture, with various media moditications, to form de novo floral or vegetative buds, roots or callus. Changes in soluble isoperoxidases were followed electrophoretically in relation to these varying morphogenetic pathways. In each of them, the number of bands increased on both the anodic and cathodic sides with time in culture. Compared to each other these four morphogenetic programmes were different in their peroxidase zymograms, mainly through varying kinetics in the development of activity of the isoenzymes. The changes observed during root and vegetative bud formation agree with previously published data, and the changes during floral bud formation agree with those observed in vivo.

    Notes: The changes in germination, peroxidase activity and isoperoxidase spectrum have been studied in apple embryos at 5°C (stratification) and at 20°C in the presence or absence of seed coats. The embryo dormancy is progressively released at 5°C, but not at 20°C. The peroxidase activity in embryos covered with seed coats is very low at 5°C as well as at 20°C which corresponds to a restricted number of isoenzymes. In isolated embryos the peroxidase activity increases significantly. This is due to an increase in both the number and the activity of the isoperoxidases and it is more pronounced at 20°C than at 5°C.The obtained results suggest that the soluble peroxidases are not involved in the process of the release of embryo dormancy. The variations observed are attributed to the growth process following germination, which can occur even at low temperature.

    Notes: Effects of gibberellic acid and kinetic on α-amylase production during the germination of barley. - The action of gibberellic acid and kinetin, alone or combined at different concentrations, has been studied on α-amylase production in whole barley seedlings and in embryoless endosperms in course of the six first days of development in the dark. The classic activation of α-amylase synthesis by gibberellic acid has been confirmed both in whole seeds and in embryoless endosperms. Kinetin inhibits α-amylase synthesis after the third day of germination but has no effect on isolated endosperms. When gibberellic acid and kinetin are given simultaneously gibberellic acid stimulated during the three first days just as it does alone, kinetin inhibits after the third day also as it was alone so that the two regulators act, without interactions, at different stages in the time. These effects of kinetin are be independent. A critical examination of the techniques used in the literature in the stud of amylase is made.

    Notes: A generalized two-step and interdependent control of basic and acidic peroxidases (EC 1.11.1.7) is observed in plant responses to different physical and chemical stimuli. An interpretative model consisting of a pathway of reactions is presented on the basis of our own data and the literature. Stress-induced membrane depolarization would generate different species of free radicals and peroxides, which in turn initiate lipid peroxidation. The degradation of cell membranes is suggested to bring about rapid changes in ionic fluxes (especially release of K+ which would result in an enhanced endogenous Ca/K ratio) and in leakage of solutes (among them electron donors such as ascorbic acid and phenolic substances). The increased intracellular relative calcium level results in: 1) activated secretion of basic peroxidases into the free space where, in association with the electron donors and maybe with the circulating indole-3-acetic acid (IAA), they eliminate the peroxides; and 2) facilitated binding of basic peroxidases to membrane structures allowing a role as 1-aminocyclopropane-1-carboxylic acid (ACC)-oxidases. The resulting IAA and ACC oxidase-mediated changes in ethylene production would further induce (this time through the protein synthesis machinery) an increase in activity of phenylalanine ammonia-lyase (EC 4.31.5) and acidic peroxidases. The resulting lignification and cell wall rigidification determines the growth and/or the developmental response to the initial stress.
