Configuration of the Stereoisomeric meso-1,3-Di-[2-piperidyl]-propan-2-ols and the Behavior of Tetrahydro[1,3]oxazines during Hydrolysis with Acids1)The stereoisomeric meso-1,3-di-[2-piperidyl]-propan-2-ols (4 and 5) are converted to the tetrahydro[1,3]oxazine derivatives 8c and 9c by condensation with formaldehyde. The rates of hydrolysis of these derivatives in acetic acid solution are compared with those of the analogous tetrahydro[1,3]oxazine derivatives of the stereoisomeric β-aminoketones 6a, b and 7a, b of known configuration. The tetrahydro[1,3]oxazine derivative of the compound previously referred to as α-meso-1,3-di-[2-piperidyl]-propan-2-ol has thus been shown to have a stability similar to those of the compounds prepared from (+)-allosedridine(6a) or (±)-norsedamine (6b), both of which have the erythro configuration. In contrast, the compounds of the β-series resemble the tetrahydro[1,3]oxazine derivatives of (-)-sedridine (7a) and of (±)-norallosedamine (7b), both of which have the threo configuration and are hydrolysed much faster. It thus follows that α-meso-1,3-di-[2-piperidyl]-propan-2-ol has the erythro configuration 4 whilst the β-compound has the threo configuration 5. - Simple tetrahydro[1,3]oxazines in protonated from or in the form of their quaternary salts are not hydrolysed.

Die stereoisomeren meso-1.3-Di-[2-piperidyl]-propan-2-ole (4 bzw. 5) werden durch Kondensation mit Formaldehyd in die Tetrahydro-[1.3]oxazin-Derivate 8c und 9c übergeführt. Deren Hydrolysegeschwindigkeit in essigsaurer Lösung wird mit derjenigen analoger Tetrahydro-[1.3]oxazin-Derivate aus den stereoisomeren β-Amino-ketonen 6a, b und 7a, b bekannter Konfiguration verglichen. Die Tetrahydro-[1.3]oxazin-Derivate aus der bisher als α-meso-1.3-Di-[2-piperidyl]-propan-2-ol bezeichneten Verbindung haben sich dabei als ähnlich stabil erwiesen wie die aus dem erythroid-konfigurierten (+)-Allosedridin (6a) sowie (±)-Norsedamin (6b) erhaltenen. Die Verbindungen der β-Reihe werden dagegen entsprechend den aus dem threoid-konfigurierten (-)-Sedridin (7a) sowie (±)-Norallosedamin (7b) gewonnenen Tetrahydro-[1.3]oxazin-Derivaten wesentlich schneller gespalten. Daraus folgt für das α-meso-1.3-Di-[2-piperidyl]-propan-2-ol die erythroide Konfiguration 4 und für die β-Verbindung die threoide Konfiguration 5. - Einfache Tetrahydro-[1.3]oxazine werden in protonierter Form oder als quartäre Salze nicht hydrolytisch zerlegt.
