Summary The investigations of the isolated pregnant rat and mouse uteri wereisometrically andisotonically recorded with the respective aids of a mechanical lever on a smoked drum and of a mechano-electrical transducer on a direct recorder. Thus the following results were found: 1. Isoxsuprine significantly diminishes the amplitude, duration and frequency of contractions in direct proportion to the doses used. 2. It decreases the tonus of both; a) spontaneous activity; b) and hypertonic contractions induced by oxytocin in statistically significant degrees which were directly proportional to doses given. 3. The maximally-effective concentrations with total inhibition in all cases consisted of a) for rats: 10−5 g; b) for mice: 1,5×10−7 g of Isoxsuprine per ml of organ-bath solution. 4. The minimally-effective concentrations for the rat and mouse were near 10−9 g of Isoxsuprine per ml of organ-bath solution. 5. The individual parameters of uterine activity were found to be percentwise, differentially effected by very specific concentrations, i.e. each parameter of uterine activity has a different threshold. 6. The Isoxsuprine induced effects are reversible. The reversibility can be obtained via a) waiting for the spontaneous return of activity; b) by rinsing the Isoxsuprine containing organ-bath and c) by counteracting it with oxytocin.

Zusammenfassung Die Untersuchungen mit Isoxsuprine (Duvadilan®) am isolierten schwangeren Ratten- und Mäuseuterus in vitro (isometrische und isotonische Registrierung) führten zu folgenden Resultaten: 1. Isoxsuprine verringertkonzentrationsabhängig und statistisch signifikant Amplitude, Kontraktionsdauer und Frequenz. 2. Es senktkonzentrationsabhängig und statistisch signifikant den Tonus a) bei Spontanwehentätigkeit, b) bei oxytocininduzierter hypertoner Wehentätigkeit. 3. Die maximal-effektive Konzentration mit totaler Hemmung in allen Fällen beträgt a) für die Ratte 10−5 g und b) für die Maus 1,5×10−7 g Isoxsuprine/ml Badlösung. 4. Die minimal-effektiven Konzentrationen liegen bei Ratte und Maus bei 10−9 g Isoxsuprine/ml Badlösung. 5. Die einzelnen Qualitäten der Uterusaktivität werden durcheine definierte Konzentrationprozentual unterschiedlich beeinflußt, d.h. haben verschiedene Schwellenkonzentrationen. 6. Die durch Isoxsuprine hervorgerufenen Effekte sind reversibel. Die Reversibilität kann erzielt werden durch a) Abwarten des spontanen Wiedereinsetzens der Aktivität, b) durch Spülen des isoxsuprinehaltigen Organbades und c) durch Antagonisierung mit Oxytocin.
