Notes: [Auszug] An excised nodule from an experimentally infected calf was used for the inoculation of monolayers of bovine testis cells. Some cytopathic changes were observed already at the first passage, but they were much more marked at the second passage, and consisted of intranuclear inclusions and the ...

Notes: [Auszug] A five-month-old bovine foetus was presented for bacteriological examination, and, as brucellosis is widely spread in native cattle in the Congo, it was at first suspected that it was infected with this pathological agent. The stomach contents and various parts of the internal organs were seeded on ...

Notes: Summary A rubella virus strain (Cendehill strain) was carried through serial passages in primary rabbit kidney tissue cultures at 34° C and at 28° C. A cytopathic effect was consistently observed in these cells, when low serum concentrations were used in the medium. Experimental batches of freezedried rubella virus vaccine were produced at several passage levels.

Notes: Summary The primary rabbit kidney cell propagated Cendehill strain of rubella virus was studied in vivo at different passage levels. The low passage levels consistently evoked significant amounts of antibodies in parenterally inoculated rabbits and monkeys. At the higher passage levels, however, the virus completely lost its immunogenicity for rabbits. A similar phenomenon was observed in monkeys. Following intramuscular inoculation of 51st passage virus a high percentage of monkeys failed to develop any detectable antibodies, as measured by serum-neutralization or hemagglutination inhibition techniques. None of the monkeys inoculated by the intranasal route with virus from the same passage level showed any serologic response. This rubella virus strain, which has been markedly modified in its biological properties for monkeys and rabbits, still proved highly immunogenic in vaccinated children.

Notes: Summary Thein vivo properties of the “Cendehill” strain of attenuated rubella virus were further studied. The absence of seroconversion in rabbits inoculated subcutaneously was confirmed. When the virus was given intravenously most rabbits seroconverted and virus could be recovered from the organs of inoculated animals. Virulent virus given subcutaneously even in minute amounts, regularly evoked antibodies. When given by the combined intrathalamic, intraspinal and intramuscular routes, the “Cendehill” strain evoked antibodies in about one third ofCercopithecus monkeys in 17 to 19 days. When cortisone was given simultaneously with the virus the seroconversion rate in monkeys inoculated under similar conditions dropped to about 3%. A depressive effect of cortisone on the production of antibodies was also observed when virulent virus or the attenuated RA 27/3 virus were used. Comparative tests with virulent virus and “Cendehill” virus were also carried out in the following species: rats, mice, ferrets, gerbils and guinea-pigs. They all responded serologically to a single parenteral inoculation of wild virus. Mice, ferrets, gerbils and guinea-pigs failed to seroconvert when inoculated with “Cende-hill” virus. The rat was the only species in which a seroconversion could be regularly obtained.

Notes: Conclusions The above reported results allow us to classify the “Wavre” isolate in the picorna group. Most porcine enteroviruses that have been described have not been studied extensively on their hemagglutinating activity (Betts, 1964), but the attempts that have been reported have given negative results (Beran, 1958;Moscovici, 1959;Webster, 1959;Jastrzebski, 1961;Switzer andL'Ecuyer, 1960;Morimoto et al., 1962;Izawa et al., 1962;Kelly, 1962;Kasza, 1965;Kubin, 1965). The virus isolated byHahnefeld from a porcine tissue culture had no hemagglutinating properties.Greig andGirard (1963) described the characteristics of a hemagglutinating virus causing encephalomyelitis in swine, but this virus differs markedly from our isolate and was considered by the authors to belong to the parainfluenza group.

Notes: Summary Three ts mutants derived from herpesvirus hominis type 2 were inoculated into rabbits and mice. Intradermal or intramuscular inoculation failed to induce any nervous symptoms in rabbits, whereas the parent virus strain showed marked neuropathogenicity. Although no detectable circulating antibodies developed as a result of the inoculation of the mutants, the animals showed a high degree of protection against a subsequent challenge with virulent virus. This protection was more marked in those animals which had received the ts mutants by the intradermal route. In mice also, peripheral inoculation of a ts mutant resulted in a morbidity markedly lower than that caused by the parent strain. Intradermal inoculation of the ts strain provided complete protection against a subsequent challenge with virulent virus either by the intracerebral or by the intradermal route. Intramuscular inoculation gave partial protection.
