    Notes: Resistance to sulfanilamide has persisted in a strain of Chilomonas Paramecium for 255 transfers (63 months) in a drug-free medium. In attempts to modify resistance, stocks derived from sulfonamide-resistant and normal strains have been acclimatized to and then maintained in media containing p-aminobenzoic acid at 5.0, 10.0, 15.0 and 20.0 mgJ100 ml. Each PABA-acclimatized strain was more susceptible to sulfanilamide than its parent stock. In other words, sulfanil-amide-resistant strains lost their resistance and normal strains became hypersensitive. One strain, adapted first to sulfanilamide, subsequently to PABA (15 mgJ100 ml) and again to sulfanilamide, showed a loss of and finally a restoration of sui-fonamide-resistance (but to a degree somewhat lower than the original level).

    Notes: SUMMARY. Strains of Chilomonas paramecium differing in degree of resistance to sulfanilamide have been established through acclimatization to this sulfonamide at 50, 100 and 200 mg. %. Resistant strains differ from the normal stock in their enhanced sensitivity to p-aminobenzoic acid. In the normal stock, sulfanilamide inhibition is reversed at an SA/PABA ratio of 10,000 but not at 20,000; in the least resistant strain, at a ratio of 400,000 but not at 800,000. In resistant strains inhibition is reversed by folk acid, methionine, adenine, cytosine and thymine; in the normal stock, none of these metabolites produces reversal. In high concentrations of PABA (10–20 mg. %) growth of the normal stock is only retarded, whereas the strain least resistant to sulfanilamide fails to recover from exposure to 20 mg. % PABA. The strain most resistant to sulfanilamide is most susceptible to PABA in high concentrations. The data suggest that resistance to sulfanilamide in C. paramecium may depend mainly upon an accelerated synthesis of PABA.

    Notes: SYNOPSIS. Chilomonas paramecium developed significant resistance to INH at concentrations up to 150 mg/100 ml. Once established, the resistant strains grew subnormally in drug-free medium and were more susceptible than the normal strain to sulfanilamide, but were much more resistant to PABA (10-20 mg/100 ml). In addition, INH-adapted strains were less responsive than the controls in PABA reversal of SA-inhibition. Strains adapted to sulfanilamide, sulfapyridine, and nicotinamide were less resistant to INH than the normal strain.

    Notes: Presently the Heavy Ion Fusion Virtual National Laboratory is researching ion sources and injector concepts to understand how to optimize beam brightness over a range of currents (50–2000 mA argon equivalent). One concept initially accelerates millimeter size, milliamp beamlets to 1 MeV before merging them into centimeter size, ampere beams. Computer simulations have shown the final brightness of the merged beams is dominated by the emittance growth of the merging process, as long as the beamlets' ion temperature is below a few electron volts. Thus, a radio frequency multicusp source capable of high current density can produce beams with better brightness compared to ones extracted from a colder source with a large aperture and lower current density. Initial experiments with such a source have successfully demonstrated simultaneously high current density, ∼100 mA/cm2, and fast turn on, ∼1 μs. Results from these experiments are presented as well as progress and plans for the next set of experiments for these sources. © 2002 American Institute of Physics.

    Notes: Abstract: Dermatitis herpetiformis (DH) is a blistering skin disease characterized by cutaneous deposits of IgA and an associated, most often asymptomatic, gluten sensitive enteropathy (GSE). Gluten sensitive enteropathy is also seen in patients that do not have skin disease or cutaneous IgA deposits, but do have significant gastrointestinal (GI) complaints. Patients with DH and with GSE without skin disease have similar small bowel morphologic changes and HLA associations and both the skin disease and the GI symptoms can be controlled by a gluten free diet. It is not known what factors allow almost all patients with DH to continue to eat gluten and not develop symptomatic gastrointestinal disease. We have examined the expression of the Vβ T-cell receptor (TCR) in the small bowel of patients with DH (n=11) and of patients with both symptomatic (n=10) and asymptomatic (n=7) GSE without skin disease to determine if differences in the pattern of TCR Vβ expression are associated with differences in the clinical manifestations of these diseases. TCR Vβ expression was analyzed using RT-PCR from small bowel biopsies. Patients with DH and those with GSE without skin disease that were on a gluten free diet and asymptomatic were found to express 6.6 and 5.6 out of 20 Vβ families respectively, with no single family preference. Examination of peripheral blood lymphocytes from these patients did not reveal any restriction of TCR Vβ family expression. In contrast, patients with symptomatic GSE expressed 12.6 Vβ families (P〈 0.05), with no consistent preferential expression of any single Vβ family between patients. Patients with DH, who are continuing to ingest wheat, show a more restricted pattern of TCR Vβ utilization, similar to that of treated patients with GSE without skin disease, and significantly different from GSE without skin disease patients eating gluten. These findings suggest that the restricted nature of the TCR Vβ expression may play a role in the different clinical manifestations of dermatitis herpetiformis and isolated gluten sensitive enteropathy.

    Notes: Growth of Tetrahymena pyriformis in a peptone medium is inhibited slightly to extensively by comparable concentrations of oxalate, citrate, and ethylenediaminetetraacetic acid (in ascending order). Single additions of Ca, Cu, Zn, and Mg, in increasing order of effectiveness, reversed citrate inhibition. EDTA inhibition was completely reversed by single additions of Ca or of certain other metals (Al, Co, Cu, Fe, Mn, Ni, Zn) forming more stable EDTA complexes. Reversal by added Mg was slight. The data suggest physiological importance of Ca for T. pyriformis.Pre-treatment of peptone solution with a cation exchanger (Natrinil) resulted in little or no growth; growth was restored almost to normal by the addition of Mg. The growth-response showed a linear relationship to quantity of Mg.

    Notes: Summary Adaptation to two antipurines, 8-azaguanine and 6-mercaptopurine, has resulted in increased resistance to the inducing agent without modifying an original resistance to sulfanilamide in a strain of Chilomonas paramecium. In the case of isoniazid, a sulfonamide-resistant strain proved to be much more susceptible than the normal strain. Adaptation has reduced this susceptibility but, so far, has not brought sensitivity back to the level of the normal strain. However, this adaptation of a sulfonamide-resistant strain to isoniazid has not appreciably decreased the original resistance to sulfanilamide. The effects of 5-bromouracil and 2,6-diaminopurine were negligible and in each case “adaptation” caused no apparent change in a previous sulfonamide-resistance. Benzimidazole, although moderately inhibitory for all strains tested, also did not modify a preexisting sulfonamideresistance.
