    Notes: Trace element concentrations in igneous rocks are frequently lognormally distributed, and Shaw (1961) suggested that minor minerals in igneous rocks might also be lognormally distributed. There is accumulating evidence to show that bulk magnetic susceptibility (BMS) often closely follows a lognormal distribution in fresh igneous rocks but, because BMS is dependent upon oxide-grain size and mineralogy, it is not obvious why this should be so. We have adopted Shaw's simple theoretical model as an argument to account for the lognormal distribution of BMS. The obvious prevailing conditions must be that the original melt was uniform, that pressure, temperature and oxygen fugacity were constant and that the minerals were formed at equilibrium. In addition, because BMS is also a function of grain size and mineralogy, the distribution of these latter parameters must be insensitive to changes in P, T, etc. at equilibrium. It appears that there must be a limit to the lognormal law in trying to apply it to minerals of greater concentration. However, BMS proxies for oxide content, and because it does closely follow the lognormal distribution it thus appears that, in crystalline igneous rocks, the lognormal law is obeyed by minerals of concentrations at least up to a few percent. Knowledge of the mean and variance of such distributions can be useful in a variety of petrological applications, especially in drillcore logging; a simple example is presented.

    Notes: Abstract Effects of deforestation upon slopes in limestones and in volcanic rocks in the Benson River valley, northern Vancouver Island, have been investigated quantitatively. Postlogging soil erosion and vegetal regeneration success were assessed by measuring soil depth, percent bare rock and moss cover, and the numbers and diversity of trees, shrubs, and plants on 25 sampling sites, each containing ten measuring quadrats selected at random. Sixteen sites were on the Quatsino Formation, a well-karstified limestone, and nine on the Karmutsen Formation of basaltic lavas. Eight sites were of virgin forest, 16 were logged between 1970 and 1983, and one (on limestone) was logged in 1911. Both bedrock types were significantly affected by the cutting. There was greater loss of soil and an increase in bare rock on the limestones. Erosion was increased significantly by burning on the limestones but not on the volcanics. Within-group comparisons on the limestones determined that steeper slopes and harder burned areas suffered the most and are slowest to regenerate. Volume of timber on the 1911 site was 19 percent of that in similar uncut forest sites. It appears that complete recovery on the barren limestone slopes will require at least some centuries.

    Notes: Abstract  A protoplast fusion strategy has been applied to advance aspects of a potato breeding programme. A sub-population of somatic hybrids, selected for agronomic potential, between tetraploid Solanum tuberosum cv. Brodick and a diploid EBN2 accession, S. sanctae-rosae was subjected to detailed molecular analysis. This study reports the use of simple sequence repeats (SSRs) to identify nuclear hybrid genomes and PCR and DNA-DNA analysis to determine organelle composition in somatic hybrids derived from these parents. SSR analysis revealed somatic hybrids containing the genetic background of S. tuberosum cv. Brodick with some specific markers from S. sanctae-rosae. One somatic hybrid contained the chloroplasts derived from S. sanctae-rosae, and several hybrids had detectable RFLP mitochrondrial DNA profiles, indicating genetic re-arrangements. We also examined the use of DNase I sensitivity to the genomic and ribosomal RNA sequences in these somatic hybrids as an indicator of changes in chromatin structure. Chromatin and DNAse I analysis showed differential sensitivity to increasing levels of nuclease; DNA from several somatic hybrids was found to be resistant to DNase I compared to the parental plants. The significance of the findings to somatic cell genetics and plant breeding studies is discussed.

    Notes: Abstract Somatic hybrids between Solanum tuberosum cv 'Brodick' and S. sanctae-rosae were selected for having elevated chromosome numbers, intermediate morphology and potato cyst nematode resistance (derived from S. sanctae-rosae). DNA was extracted from this material and subjected to inter-SSR PCR directed by oligonucleotide primers comprising 5′-anchored di- and tri-nucleotide repeat motifs. PCR products were separated by polyacrylamide gel electrophoresis and visualised by silver staining. An average of 30.31 bands per primer were generated in each parent and, on average, 8.19 of these bands were polymorphic. S. sanctae-rosae-specific bands were observed in the hybrid material as was evidence of deletions, substitutions and rearrangements. The relative merits of this technique for the analysis of somatic hybrid material are discussed.

    Notes: Summary E. coli spheroplasts can be used to deliver DNA vectors into plant protoplasts. The use of fluorescent dyes showed that 25–100% of the protoplast population was associated with 1–9 spheroplasts following incubation with several fusogens. Electron microscopy demonstrated spheroplasts attached to protoplasts via a plasma membrane protrusion after high pH/Ca2+ treatment, but PEG-high pH/Ca2+ promoted endocytosis of spheroplasts into a plasma membrane bounded vesicle. Ultrastructural profiles showed that fusion between spheroplasts and protoplasts did not occur. Immunofluorescence studies detectedE. coli antigens associated with tobacco protoplasts, and after fusogen treatment the antigens were dispersed within the peripheral cytoplasm. The elimination of residual contaminatingE. coli cells from protoplasts was achieved by lysozyme and antibiotic treatment, thus allowing DNA vector assessment in axenic culture.

    Notes: Abstract Ribosomal RNA genes were exploited as markers to identify somatic hybrids between Solanum tuberosum cv. Brodick and wild diploid Solanum species, S. megistacrolobum, S. sanctae-rosae and S. sparsipilum and DNA methylation as a possible regulatory factor in gene expression was investigated. Specific restriction enzyme/probe combinations revealed useful polymorphisms in the conserved coding and variable intergenic spacer regions of the ribosomal RNA genes. Some intermediate ribosomal RNA gene profiles indicate hybridity whereas others were characteristic of S. tuberosum cv. Brodick. This evidence is suggestive of somatic exchange/re-arrangement between the NOR region of S. sanctae-rosae and S. tuberosum cv. Brodick. Ribosomal RNA gene copy number analysis of the somatic hybrids did not reveal hexaploid values suggesting that these products are not symmetric hybrids derived from the parental diploid and tetraploid plants. The results indicate site-specific methylation of ribosomal RNA gene sequences for the parental plants; while some somatic hybrids display a reduction, others show an increase. The significance of the findings for somatic cell genetics and plant breeding studies is discussed.

    Notes: Summary Ribosomal gene (rDNA) probes have been used to assess genomic changes in plants of (1) S. tuberosum cv Desiree subjected to slow growth and (2) S. tuberosum cv Golden Wonder recovered from cryopreservation. Restriction fragment length polymorphisms (RFLPs) were detected in two out of 16 DNA samples extracted from plants derived from slow growth, control plants did not show RFLP differences. Plants recovered from cryopreserved shoot-tips of Golden Wonder were unchanged in their ribosomal gene RFLP profile compared to the untreated controls. The use of rDNA probes as tools to assess stability, and the possible detrimental effects of slow growth, somaclonal variation, cryoprotectants, and freezing injury are discussed.
