    Notes: The physiological role of topoisomerase III is unclear for any organism. We show here that the removal of topoisomerase III in temperature sensitive topoisomerase IV mutants in Escherichia coli results in inviability at the permissive temperature. The removal of topoisomerase III has no effect on the accumulation of catenated intermediates of DNA replication, even when topoisomerase IV activity is removed. Either recQ or recA null mutations, but not helD null or lexA3, partially rescued the synthetic lethality of the double topoisomerase III/IV mutant, indicating a role for topoisomerase III in recombination. We find a bias against deleting the gene encoding topoisomerase III in ruvC53 or ΔruvABC backgrounds compared with the isogenic wild-type strains. The topoisomerase III RuvC double mutants that can be constructed are five- to 10-fold more sensitive to UV irradiation and mitomycin C treatment and are twofold less efficient in transduction efficiency than ruvC53 mutants. The overexpression of ruvABC allows the construction of the topoisomerase III/IV double mutant. These data are consistent with a role for topoisomerase III in disentangling recombination intermediates as an alternative to RuvABC to maintain the stability of the genome.

    Notes: Summary Crosses were set up, using the b2 locus of Ascobolus immersus, to detect and measure the restoration of the parental genotype of a chromatid from hetero-duplex. From the results it can be concluded that: (1) restoration occurs, and its frequency is comparable to the frequency of conversion; (2) the relative frequency of conversion and restoration on the two homologous chromatids is that expected if the disparity in conversion to mutant and to wild-type of the marker is caused by the decision whether to excise the mutant or wild-type chain. This decision is the same on both chromatids. However, when all data are pooled, a slight, but significant, excess of conversion over restoration is found. The cause of this in discussed; (3) the absence of an excess of restoration over conversion is taken to imply that, in this system, the length of heteroduplex formed in an event is continuous; (4) these findings support the interpretation of tetrads where a crossover is separated from conversion, that they arise by independent correction giving both conversion and restoration in a heteroduplex tract continuous with the crossover.

    Notes: Summary Analysis of unselected and selected (prototroph-containing) tetrads, carrying two or three alleles of the his1 locus in yeast, led to the following conclusions: (1) Conversion at this locus is strongly polarized and asymmetrical. (2) In cases where a crossover is separated from a converted marker by an unconverted segment, these effects are parts of the same event. (3) The distribution of the various classes of tetrad with a crossover associated with conversion at his1 shows that the crossover is at the variable end of the event within the gene. (4) Reciprocal intragenic recombination results from restoration correction in crossover events which end between the alleles. Its occurrence is marker dependent, not distance dependent. (5) A deficiency in reciprocal intragenic recombination is interpreted as an allele-specific disparity in the ratio of conversion to restoration. (6) Marker effects are interpreted in terms of reciprocal influence of markers on each other's correction from heteroduplex, and of interference by heterozygosity with the migration of the cross-strand exchange.

    Notes: Summary Heteroduplex repair models of recombination predict that restoration of the parental genotype from heteroduplex will have the same frequency as conversion to the genotype of the homologue. We have reported previously (Savage and Hastings 1981) that the proportion of intragenic reciprocal events in the his1 locus of yeast is too low for this expectation. In this study, two classes of recombination event involving longer lengths of his1 are compared in a series of crosses using a constant right-hand marker. This comparison gives a value for conversion: restoration for the left-hand markers which is independant of the method used before. The values obtained by the two methods are significantly correlated, confirming that there is a disparity in the ratio of conversion to restoration, and that this disparity is seen as a deficiency of restoration for five alleles of his1.

    Notes: Summary A possible method of manipulating allelopathy would be to develop crop varieties showing an increased tolerance to allelopathic chemicals. We therefore examined four flax (Linum usitatissimum) varieties and two wild Linum species in the presence of p-coumaric acid and four barley (Hordeum vulgare) varieties in the presence of p-coumaric acid, scopoletin and wild oat (Avena fatua) extract. Analysis of variance indicates significant interaction between variety and treatment for shoot and root growth for seedling flax, shoot growth for older flax, and root growth for seedling barley. These differences in tolerance between varieties could be exploited to develop-varieties with greater tolerances to the allelochemicals produced by weeds or in crop residues and therefore potentially more tolerant of the presence of weeds.
