    Notes: A new optical method using a Pockels device is developed to simultaneously measure two directional components of an electric field. In this method, an output light beam from a Pockels device is split into two beams and then each of the beams is transmitted through a polarizing plate whose polarization direction is different, depending on the measured field component. The relation between two measured field components and the required polarization direction of the polarizing plate is theoretically derived for representative materials of the Pockels device and is also examined experimentally. A field sensor is made on the basis of this new idea and is used to investigate a nonuniform electric field. An electric field changing spatially by 20% per centimeter can be measured with an accuracy of 5% using the Pockels sensor. The sensor is successfully applied to measure an impulse electric field between a sphere-to-sphere gap.

To determine whether the antihypertensive response in deoxycorticosterone acetate (DOCA) salt-treated rats was mediated by kinins on the luminal side of renal tubules or in the circulation, selective urinary kininase inhibitors were administered to normal Brown Norway Kitasato (BN-Ki) rats and kininogen-deficient Brown Norway Katholiek (BN-Ka) rats.2. Kinins were degraded by neutral endopeptidase (NEP) and carboxypeptidase Y-like kininase (CPY) in urine, but were inactivated mainly by angiotensin-converting enzyme (ACE) in the plasma.3. Ebelactone B inhibited CPY, while poststatin inhibited CPY and NEP.4. Daily administration of poststatin (5 mg/kg per day, s.c.) for 3 days reduced blood pressure (BP) in DOCA salt-treated BN-Ki rats, but not in BN-Ka rats.5. Ebelactone B (5 mg/kg per day, s.c.) also reduced BP in BN-Ki rats, which was accompanied by increased urinary sodium excretion, but had no effect on BP in BN-Ka rats.6. Lisinopril (5 mg/kg per day, s.c.) had no effect on BP in either rat strain.7. Arterial kinin levels in BN-Ki rats increased significantly (2.2–4.6 pg/mL) with captopril (10 mg/kg, s.c.). However, arterial kinin levels that induced hypotension following the infusion of bradykinin (1000 ng/kg per min, i.v.) were 110- fold higher than endogenous arterial kinin levels attained following captopril.8. These results suggest that inhibition of kinin degradation on the luminal side of the renal tubules may effectively attenuate hypertension.

    Notes: Abstract Experimental pleurisy was induced by intrapleural injection of carrageenin or kaolin in three strains of rat: Brown Norway-Katholiek (B/N-Ka), Brown Norway-Kitasato (B/N-Ki) and Sprague-Dawley (SD). B/N-Ka rats (kininogen-deficient) showed significantly less pleural fluid accumulation and exudation rate than SD rats or than B/N-Ki rats (normal). The result indicates the involvement of the plasma kallikrein-kinin system in these pleurisies and a role of high-molecular-weight kininogen is suggested.

    Notes: Abstract  Biochemical and gross pathological effects of diquat were studied with special attention to cysteine proteinase inhibitor level which was often increased in acute and chronic disorder. Diquat was fed continuously to rats at the dose of 1000 ppm in the diet. After 10 days, anorexia and severe diarrhea were observed but epistaxis and hypokinesia were not apparent. The rats were killed after feeding the diet for 13.5 days and plasma components such as acute phase reactant proteins and some vitamins which act as antioxidants were examined. The results showed that α-cysteine proteinase inhibitor (α-CPI) increased to 9-fold and vitamin C radical increased to 1.6-fold, whereas α1 proteinase inhibitor (α1-PI) decreased to 0.9-fold and vitamins C and E were the same as the control. Among three components of α-CPI, the T kininogen level in intoxicated rat plasma was about 20-fold, whereas the high molecular weight kininogen level was about 2-fold of the control. Diquat also enhanced the cysteine proteinase inhibitor (CPI) level to 20-fold in kidney and to 7- to 10-fold in the other organs. The large increment of T kininogen in these organs was also confirmed immunologically. The kidney showed a granular degeneration and its weight increased to 1.2-fold of control. The other organs showed neither gross pathological alteration nor weight change, compared with the control. The diquat distribution was highest in spleen and next highest in kidney among several organs. These results were compared with those caused by paraquat.

    Notes: Abstract We evaluated the utility of in-situ hybridization (ISH) for the rapid diagnosis of sepsis. We applied this approach to polymorphonuclear neutrophil (PMN)-rich smears from patients with suspected bacterial infection. Positive results by ISH were obtained in the smears of 123 of 292 patients (42%), while only 32 of the 292 (11%) were positive by blood culture. These findings indicate that ISH is almost four times more sensitive than the culture method for the detection of sepsis. ISH results are obtained within 1 day, while 1 day to 2 weeks is required for the results of blood culture. Blood culture and ISH methods detected the same bacteria in two patients. ISH also successfully identified the same bacteria in blood and PMN-rich body fluid (bronchoalveolar lavage samples) in 6 patients. In 19 patients, ISH of blood detected the same bacteria as those found in subcultures from other sources (e.g., stool, sputum, nasal cavity). We discuss these results in comparison with blood culture results in terms of evaluating a rapid approach to the management of patients with sepsis.

    Notes: Primary monolyer cultures of adult rat hepatocytes can be induced to undergo DNA synthesis in serum-free medium in the presence of insulin, glucagon, and epidermal growth factor (three factors). We have found that hepatocyte DNA synthesis is affected not only by an endogenous stimulant produced by the hepatocytes and released into the culture medium. Serum has a strong inhibitory effect on hepatocyte DNA synthesis. Partially purified human platelet extract (“platelet inhibitor”) inhibits the three-factor-induced DNA synthesis in a concenration-dependent manner. Pure βTGF at 0.5 ng/ml as well as HPLC-purified PDGF at 10 ng/ml completely inhibit the three-factor-induced DNA synthesis. Determination of the time required for the presence of the three factors and the platelet inhibitor to exert their effects indicated that the inhibition of DNA synthesis is caused not by competition of the platelet inhibitor with any of the three factors but through an independent pathway. Hepatocyte DNA synthesis is density-dependent and is greater if medium is not changed during the course of an experiment than if medium is changed daily. Hepatocyte-conditioned medium is also affective in stimulating DNA synthesis beyond the level induced by the three factors. These results suggest that an endogenous stimulant for hepatocyte DNA synthesis is produced by the hepatocytes themselves. Our studies demonstrate that hepatocyte DNA synthesis is subject to both stimulatory and inhibitory controls. Unlike the three factors, the endogenous stimulant can overcome the inhibition by the platelet inhibitor, suggesting the importance of these factors in the physiological control of hepatocyte DNA synthesis.

Auf Grund von Untersuchungen der Eigenschaften dieser Polyamide wird der Einfluß des Tetrahydrofuranringes auf die Eigenschaften der Polykondensate diskutiert.Die Einführung des Tetrahydrofuranrings in die Polyamidkette hat nut wenig Einfluß auf Schmelzpunkt und Kristallinität, erhöht jedoch beträchtlich die Löslichkeit, Feuchtigkeitsaufnahme und das Farbstoffbindevermögen. Das Mischpolyamid 66/6T (Gew.-Verhältnis 4 : 1) gibt Fasern mit guten mechanischen Eigenschaften und stark verbesserter Anfärbbarkeit.
    Notes: A homologous series of polyamides from tetrahydrofuran-2,5-dipropionic acid and polymethylenediamine (nylon nT, n = 2, 4 ∼ 10) and two series of copolyamides (nylon 66/6T and 69/6T) have been prepared. Based on studies of the properties of these polymers the effects of tetrahydrofuran ring on polymer properties have been discussed.The introduction of tetrahydrofuran ring into polyamide chain has little influence on melting point and christallinity of the polymer, but fairly increases the solubility, moisture regain, and receptivity towards dyes. The copolyamide 66/6T (wt. ratio 4 : 1) gives a fibre having good mechanical properties and greatly improved dyeing properties.

    Notes: New fiber-forming polyamides containing heterocyclic groups were prepared form furan 2,5-dipropionic acid and the corresponding tetrahydrofuran compound which were found to be prepared easily from furfural and levulinic acid. For example, poly-decamethylenetetrahydrofuran 2,5-dipropionamide (intrinsic viscosity 0.74) gave a fiber having, when cold drawn to 350%, tensile strength 3.1 g./denier and elongation 25%.
