    Notes: Abstract: In the presence of 1 mM spermine, accumulations of 3H labelled inositol phosphates elicited by quisqualate (100 μM) and 1 -aminocyclopentane-trans- 1, 3-dicarboxylate (t-ACPD, 300 μM) were significantly enhanced by 21 and 26%, respectively, without a significant alteration in the accumulation elicited by l-glutamate (10 mM) or DL-α-amino-3-hydroxy-5-methyl-4-isoxalone propionate (10 μM). Analysis of concentration-response data indicated that the presence of spermine led to an increase in the maximal response to t-ACPD without altering the EC50 value. The stimulatory effect of spermine on the accumulation of t-ACPD-elicited 3H-inositol phosphates was not reversed by ifenprodil or diethylenetriamine (putative polyamine site antagonists), by agents that activate or inhibit protein kinase C, or by calcium channel blockade, but was abolished in the presence of elevated extracellular calcium ion concentration. We conclude that spermine enhances the phosphoinositide turnover in guinea pig cerebral cortical slices elicited by the “metabotropic” excitatory amino acid receptor. The site through which the action of spermine is mediated remains to be defined, but it is apparently distinct from that suggested to modulate N-methyl-D-aspartate receptor activity.

    Notes: Abstract: In this report, we show that under conditions designed to provide an initially uniform incorporation of [3H]inositol into mouse and guinea pig cerebral cortical slices prior to agonist stimulation, the accumulation of 3H-inositol phosphates (3H-InsPx, x = 1–4) induced by histamine in mouse and guinea pig cerebral cortical slices increased in a quasilinear manner with increasing added calcium. Raising the ambient calcium ion concentration failed to reduce the adenosine receptor-mediated inhibition of the histamine-induced 3H-InsPx response in mouse cerebral cortical slices. Similarly, the potentiation of the histamine response by adenosine receptor activation in guinea pig cerebral cortical slices was unaffected by lowering the added calcium ion concentration. The presence of the calcium ionophore A23187 (33 μM) produced 3H-InsPx responses in both mouse and guinea pig cerebral cortical slices, which were not affected by the presence of the stable adenosine analogue 2-chloroadenosine. A23187 also potentiated the accumulation of 3H-InsPx induced by histamine in both species. Both the inhibitory and potentiatory modulations of the histamine response by 2-chloroadenosine in mouse and guinea pig, respectively, were still apparent in the presence of A23187. These results indicate that the histamine-induced 3H-InsPx accumulations in both mouse and guinea pig cerebral cortical slices are sensitive to variations in calcium ion concentrations. However, the adenosine receptor modulations of the histamine responses are relatively insensitive to fluctuations in either extra-or intracellular calcium ion concentrations, and thus cannot be mediated by effects on calcium ion movements.

    Notes: Abstract: In cerebral cortical slices from the guinea-pig, quinoxalinedione derivatives antagonised the generation of 3H-inositol phosphates evoked by the excitatory amino acids quisqualate and DL-α-amino-3-hydroxy-5-methyl-4-isoxalone propionic acid but were without effect on the trans-DL-1-amino- 1,3-cyclopentanedicarboxylic acid and l-glutamate responses. Omission of calcium from the medium reduced the accumulation of 3H-inositol phosphates induced by incubation with trans-DL-1-amino- 1,3-cyclopentanedicarboxylic acid (incubation for 45 min) by 〉 50%, whereas the responses to l-glutamate and the two other amino acid analogues were reduced by ∼20%. Generation of inositol 1,4,5-trisphosphate over a 30-s period by treatment with quisqualate, trans-DL-1-amino- 1,3-cyclopentanedicarboxylic acid, KCI, and carbachol was abolished in the presence of nominally calcium-free medium. l-Glutamate induced a large, rapid increase in inositol 1,4,5-trisphosphate mass (more than threefold), which was, however, unaffected by omission of calcium from the medium. These results indicate that of the excitatory amino acids tested, only l-glutamate may be classed as a metabotropic receptor agonist in guinea-pig cerebral cortical slices with respect to generation of inositol phosphates. The other agents appear to stimulate accumulation of inositol phosphates, at least in part through some mechanism requiring the presence of extracellular Ca2+, presumably Ca2+ entry.

    Notes: Abstract: Addition of histamine (0.1 mM) to guinea-pig hippocampal slices causes a 20- to 30-fold increase in the accumulation of cyclic AMP compared with basal levels. This accumulation represents a balance between cyclic AMP production by adenylate cyclase and cyclic AMP breakdown mediated by phosphodiesterase (PDE). However, brain tissues are known to contain several different PDE isozymes. To determine which are involved in this response to histamine, the effect of isozyme-specific PDE inhibitors on cyclic AMP accumulation was examined in the hippocampus. MB 22948 (0.1 mM), an inhibitor of PDEs I and II, had no significant effect on the response to either 1 μM or 0.1 mM histamine. SKF 94120 (0.1 mM), a PDE III inhibitor, was also without effect in the presence of 1 μM histamine, although with 0.1 mM histamine, it caused a weak (1.25-fold compared with control), but statistically significant, enhancement of cyclic AMP accumulation. However, both rolipram (0.1 mM), a PDE IV inhibitor, and 3-isobutyl-l-methylxanthine (0.1 or 1 mM), an inhibitor of all forms of PDE, significantly increased cyclic AMP accumulation (2.8- to 6.5-fold compared with controls), and the relative size of this effect decreased with increasing histamine concentration. It is concluded that PDE IV is the main PDE isozyme involved in cyclic AMP turnover in guinea-pig hippocampal slices responding to histamine.

    Notes: We describe instrumentation for conducting high sensitivity millimeter-wave cavity perturbation measurements over a broad frequency range (40–200 GHz) and in the presence of strong magnetic fields (up to 33 T). A millimeter-wave vector network analyzer (MVNA) acts as a continuously tunable microwave source and phase sensitive detector (8–350 GHz), enabling simultaneous measurements of the complex cavity parameters (resonance frequency and Q value) at a rapid repetition rate (∼10 kHz). We discuss the principle of operation of the MVNA and the construction of a probe for coupling the MVNA to various cylindrical resonator configurations which can easily be inserted into a high field magnet cryostat. We also present several experimental results which demonstrate the potential of the instrument for studies of low-dimensional conducting systems. © 2000 American Institute of Physics.

    Notes: We assess the relevance of theories of increased decentralization in post-bureaucratic organizations for understanding changes in the organization of the research and development (R&D) function. Analysis of a sample of companies shows that R&D is less decentralized than contemporary accounts suggest, and there are few signs of it becoming more decentralized. We present evidence on the organization of R&D in two sectors (mechanical engineering, and food and drink), which suggests: (1) most firms remain centralized in their approach to R&D, relative to other functions; (2) firms that adopt a more decentralized approach to R&D do so for more complex reasons than have been recognized; (3) inter-functional integration remains a critical issue, for centralized and decentralized organizations alike; (4) introducing market principles into R&D brings risks of market failure.

    Notes: Abstract  Background: Troponin I is used to diagnose myocardial infarction (MI). Its use and pattern of elevation is not well defined in coronary artery bypass graft (CABG) surgery. This study assessed the timing of troponin I elevation in patients undergoing urgent CABG. Method: Patients undergoing urgent isolated-CABG with cardiopulmonary bypass were studied prospectively. Blood samples were taken to measure CK, CK-MB, and troponin I: preoperatively, 7 hours postoperatively, 14 to 18 hours postoperatively, 30 to 48 hours postoperatively, and on postoperative day 4. Electrocardiograms and in-hospital course were recorded. Perioperative MI (PMI) was defined by either (i) ECG criteria of new Q-waves in the presence of CK-MB elevation 〉50 μg/L or (ii) CK-MB 〉 100 μg/L. Results: Of the 50 patients studied, 6 met the criteria for PMI (12%); 2 by criteria (i) and 4 by criteria (ii). In patients not meeting the criteria for MI the troponin I level peaked at 7 hour post-op with a mean of 20.97 μg/L (95% CI, 17.11 to 24.83). At this time, patients who met the criteria for MI had a mean troponin I level of 46.85 μg/L (95% CI, 36.40 to 57.30). Of variables investigated for the 44 patients who did not meet MI criteria, only preoperative troponin I level impacted peak postoperative troponin I. Conclusions: CABG elevates troponin I far beyond current diagnostic benchmarks without the clinical occurrence of a MI and appears to peak during the second postoperative day. An elevated preoperative troponin I may predict an elevated peak postoperative troponin I in patients who do not have a PMI.
