    Notes: Concentrations of calcium, strontium, sodium, and potassium were measured along chronological transects of sectioned sagittae from adult red drum (Sciaenops ocellatus), using a wavelength-dispersive electron microprobe. Coarse sampling involved triplicate measurements at the centre of each opaque (winter) and translucent (summer) zone. Fine sampling was performed in duplicate at equidistant points (15 μm apart) spanning four opaque zones (3 years of life). Concentrations of strontium generally increased with distance from the core (age). Other elements showed no consistent long-term trends. Sodium and potassium showed consistent differences between winter and summer otolith zones for ages 6 to 15, but calcium and strontium did not show this seasonal difference. Sampling through these zones on a finer spatial scale confirmed the winter/summer differences as cyclic trends. There was general concordance between annual variation in sodium and potassium concentrations in otoliths and concurrent trends in sea temperature, but significant departures in agreement suggested that temperature was not the immediate determinant of sodium and potassium incorporation. It is suggested that the roughly seasonal patterns of variation in otolith sodium and potassium concentrations may be a result of reproductive activity.

    Notes: Nezumia liolepis and Nezumia stelgidolepis were collected during a National Marine Fisheries Service 1997 groundfish survey along the Pacific coast of North America. Bottom trawling was conducted from 188 to 1260 m depths. Nezumia liolepis were collected from 581 to 1247 m (3·4–5·7°C) and N. stelgidolepis from 285 to 555 m (6·0–8·4°C). The two species had distinct depth and temperature distributions and the majority of the specimens for each species came from narrow depth ranges. Nezumia stelgidolepis attains a larger pre-anal fin length (117 mm) than N. liolepis (83 mm), and age estimates from otolith ring counts indicate ages from 3 to 9 years for N. liolepis and 7 and 13 years for N. stelgidolepis from specimens collected. The gonads of N. liolepis were not reproductively active, and a single N. stelgidolepis possessed eggs of various sizes suggesting batch spawning. N. liolepis and N. stelgidolepis fed benthically, primarily on crustaceans such as amphipods, shrimp, mysids and polychaete worms. Although diet overlap was high, these two species appear to limit competition for resources by habitat separation.

    Notes: A method has been developed for the automated sample pretreatment of organochlorine pesticides (OCP's) and polychlorinated biphenyls (PCB's) in extracts of human milk. This work was part of a regular monitoring program presently carried out at our institute. In this program several hundreds of human milk samples have to be analyzed for the occurrence of PCB's and OCP's.With a normal bore straight phase HPLC system, utilizing column switching we are able to separate the fat from the compounds of interest and, moreover, complete separation of the PCB fraction from the OCP's can be achieved. Under the conditions used to separate the PCB fraction from intefering OCP's column-switching is essential since the retention times for the OCP's vary from 4 minutes for hexachlorobenzene (HCB) to more than 2 hours for dieldrin.1 ml of an extract containing 45 mg of fat is injected on the first (pre)column, the fat is retained on this column and the early eluting HCB, the PCB fraction, and the DDT complex are transferred to the second (analytical) column. Compounds eluting later than p,p′-DDT are collected directly from the precolumn. Meanwhile, the PCB fraction is separated from the rest of the OCP's on the analytical column.Contrary to conventional gravity-controlled chromatography or solid phase extraction the clean-up process can be monitored on-line by UV-detection, thus rendering a fast and reliable optimization of the system. The OCP-fractions collected from the LC are pooled before they are transferred to a high resolution gas chromatograph equipped with a large volume option. The PCB-fraction is injected directly in a HRGC equipped with a concurrent solvent evaporation injection device.The limits of detection for the OCP's (HCB, α-,β- and γ-HCH, β-Hepo (heptachlorepoxide), dieldrin, p,p′-DDE, o,p′-DDT p,p′-DDT and TDE) and the PCB's investigated are at sub-ppb level (fat basis); the recoveries vary from 80 to 100%.
