    Notes: In the rat two major molecular variants of prolactin are recorded i.e. 23,000 Mr and glycosylated 26,000 Mr. In order to further characterize the glycosylated 26,000 rat prolactin molecular variant, rat pituitary cell lysates were digested with several glycoen-zymes and the digestion products submitted to sodium dodecyl sulphate polyacrylamide gel electrophoresis and subsequent immunoblotting. The results were as follows: treatment with 1) neuraminidase, specific for sialic acid, yielded an Mr decrease of the glycosidic variant from 26,000 to 24,500, 23,800, 23,000 and 22,000; 2) endo-α-N-acetylgalactosaminidase, which releases the disaccharide Gal (β 1–3) GalNac from O-glycans, split 26,000 rat prolactin into a doublet of Mr 26,000 to 25,500; and 3) mixed exoglycosidases from Turbo cornutus caused a gradual Mr shift from 26,000 to 23,000.Affinity chromatography on wheat germ agglutinin Sepharose 6MB and soybean agglutinin agarose of rat pituitary homogenates and competitive inhibition tests showed that glycosylated rat prolactin has distinct affinity for these lectins.From the experimental data it is proposed that glycosylated rat prolactin is O-linked through threonine by the disaccharide Gal (β 1–3) GalNac and possesses at least GalNac, and/or Gal and sialyl residues.

    Notes: Abstract. Growth hormone (GH) and prolactin (PRL) qualify as lymphohaemopoietic growth and differentiation factors, and so does insulin-like growth factor (IGF)-I, which mediates many of GH activities. Although there is only limited evidence that endocrine, paracrine or autocrine GH or PRL play a role in human leukaemia and lymphoma, the expression of these factors or their receptors may have diagnostic or therapeutic implications. Indeed, the participation of GH, PRL or IGF-I in the development or progression of certain haematological malignancies or to the antitumour immune response has been documented. Examples discussed in this review include a rat lymphoma in which the PRL receptor acts as an oncogene; the rat Nb2 lymphoma, which is dependent on PRL for growth; and experiments showing that PRL stimulates natural killer cell activity and the development of lymphokine-activated killer cells.

    Notes: Abstract. Prolactin (PRL) and growth hormone (GH) qualify as lymphoid growth and differentiation factors. Yet, long-standing hyper- or hyposecretion of PRL or GH does not induce any significant alteration of the immune system. Subclinical changes in laboratory parameters (such as chemotaxis or phagocytosis by granulocytes or macrophages or natural killer cell activity) have been reported in some of these conditions. The GH-insulin-like growth factor (IGF)-I axis is dysregulated in ageing, in catabolic states and in critical illness. Immune parameters, such as infection rate, are being monitored during clinical trials with GH or IGF-I. Hyperprolactinaemia may play an aggravating role in systemic lupus erythematosus, in autoimmune thyroiditis and in other autoimmune diseases. The patient may benefit from increased alertness about interactions between the endocrine and immune systems.

    Notes: Summary Cells grown as adhering monolayers have been iodinated using the lactoperoxidase technique with glucose and glucose oxidase instead of H2O2. A high degree of cytologic fixation, as evidenced by electron microscopy, resulted from this treatment even in the absence of lactoperoxidase.
