    Description / Table of Contents: Resumé Cette étude prospective randomisée de 4 ans est destinée à comparer l'efficacité et les taux de complication du Nova T et du MLCu250 insérés aussitôt après un avortement. A la date de l'interruption (30/11/83), tous les malades du premier groupe de 400 femmes avaient au moins 12 mois d'utilisation. A ce stade de l'essai, aucune différence significative n'est apparue entre les deux dispositifs dans aucune des catégories d'interruption habituelles.
    Abstract: Resumen Este estudio prospectivo randomizado, de 4 años, se ha creado para comparar las tasas de efectividad y complicaciones de los dispositivos Nova T y MLCu250 insertados inmediatamente después de un aborto. En la fecha de cierre del estudio (noviembre 30 de 1983), todas las pacientes en la primera cohorte de 400 mujeres, habían completado al menos 12 meses de uso. En el momento de este análisis no se encontró una diferencia significativa entre los dos dispositivos para cualquiera de las categorías de terminaciones típicas.
    Notes: Abstract This 4-year prospective randomized study is designed to compare the effectiveness and complication rates of the Nova T and MLCu250 inserted immediately postabortion. At the cut-off date (30 November 1983), all patients in the first cohort of 400 women had completed at least 12 months of use. At this stage of the trial, no significant differences had emerged between the two devices in any of the standard termination categories.

    Notes: Abstract This paper investigates the optimum control of a heat exchanger having internal heat sources from a reference steady state to a desired value. Both the wall and coolant are treated as distributed-parameter systems. Under certain constraints inherent in the operating conditions and physical limitations of the heat exchanger, the control function of the system, i.e. the heat generation rate which minimizes the deviation of the temperature distribution from an assigned pattern at a given time, is found through the use of a linear programming method. The effects of physical parameters on the optimal control function and the temperature response and distribution are examined. Experimental results are presented which compare favorably with the theoretical analysis. Heat exchangers to which these results apply include the electrical heater, a chemical reactor in which a chemical reaction occurs within the solid walls and a heterogeneous nuclear reactor.

    Notes: Abstract Long-term culture of hepatocytes has been challenged by the loss of differentiated functions. In particular, there is a rapid decline in cytochrome P450 (CYP). In this study, we cocultured rat hepatocytes with 3T3 fibroblasts for 10 days, and examined hepatocyte viability, morphology, and expression of CYP3A. Terfenadine was incubated with the cultures, and its biotransformation was quantitatively analyzed by HPLC. Terfenadine is metabolized by two major pathways:C-hydroxylation to an alcohol metabolite which is further oxidized to a carboxylic acid, andN-dealkylation to azacyclonol. In rat liver, only theN-dealkylation pathway appears to be mediated by CYP3A since anti-rat CYP3A antibody inhibited azacyclonol but not alcohol metabolite formation in incubations of terfenadine with liver microsomes. Freshly isolated rat hepatocytes were seeded on top of confluent 3T3 cells. Cultures were maintained in Williams' E medium supplemented with 10% fetal bovine serum and either 0.1 μmol/L or 5 μmol/L dexamethasone. In pure hepatocyte cultures, viability, as determined by lactate dehydrogenase (LDH) activity, decreased steadily to less than 30% of initial levels by day 10. In cocultures, LDH activity remained high and was 70% of initial levels on day 10. The half-life of terfenadine disappearance was optimally maintained in cocultures treated with 5 μmol/L dexamethasone, and was associated with the increased formation of azacyclonol. On day 5, nearly 50% of added 5 μmol/L terfenadine was converted to azacyclonol within 6 h, whereas the conversion was only 4% on day 1. Western and RNA-slot blot analyses confirmed that treatment with 5 μmol/L dexamethasone induced CYP3A mRNA expression and CYP3A protein expression. This coculture system could offer a useful approach in the study of drugs and xenobiotics metabolized by CYP3A.
