    Notizen: Maize (Zea mays L., hybrid INRA 260) was grown in the greenhouse with mineral nutrition of different sulphate concentrations. Mature seeds from these plants were compared for their free amino acid and protein N forms. For the most S-deficient sample, the Asx (asparagine + aspartic acid) content increased by 30% as compared with control, while methionine and cysteine decreased (by 25 and 30%, respectively), as well as glycine, lysine, histidine, arginine and tryptophan. In seeds lowest in S the non-protein N to total N ratio was 77% higher than in the control. Free asparagine dominated in starved seeds (50 mol % of total free amino acids) and was ten-fold more concentrated than in the control, where proline was the predominant free amino acid. Thus the Asx of non-protein N reached 28% of the total mol Asx of the whole starved seed. Altered S nutrition had virtually no effect on the amino acid composition of the main protein fractions, but it significantly changed their ratios. Zeins, which are poor in S-containing amino acids, showed 25% higher level than in seeds supplied with normal S. As a counterbalance, two glutelin subfractions rich in S-containing amino acids, decreased by 36–71% under limiting S nutrition.It is concluded that the plant reacts against S deficiency by modifying its N metabolism. Significant accumulation occurred of free asparagine, which is the main form of N transportation. The biosynthesis of seed storage protein occurred through the accumulation of the highest possible protein quantity allowed by the available S-containing amino acids, i.e. proteins low in S-containing amino acids were preferentially synthesized.

    Notizen: Abstract Elicitins are 10 kDa holoproteins secreted by Phytophthora fungi, that elicit an incompatible hypersensitive reaction, leading to resistance against fungal and bacterial plant pathogens. Comparison of primary sequences of α-elicitins and β-elicitins indicated several potential necrotic activity-determining residues. All of the highly necrotic β-elicitins have a hydrophilic residue (usually lysine) at position 13, whereas in the less necrotic α-elicitins this residue is replaced by a valine. Here, we report the synthesis and expression of a gene encoding a highly necrotic elicitin, β-cryptogein, and we show that the substitution of Lys-13 of this recombinant protein by a valine leads to a drastic alteration to the necrotic activity of the recombinant protein.

    Notizen: Abstract Peptide maps and partial amino acid sequences of the 3 main pectinmethylesterases (PMEs) solubilized from mung bean hypocotyl cell walls demonstrated that these proteins were different isozymes originating from a small multigene family. A cDNA clone encoding the most alkaline PME (PEγ) have been obtained by PCR using degenerate oligonucleotide primers. Combining the protein and nucleotide sequencing data, the complete amino acid sequence of PEγ was determined. The nature protein is composed of 318 amino acids with a calculatedM r of 34 677 and an estimated pI of 9.84 consistent with the values previously obtained by SDS-PAGE and IEF. It shares most of the conserved regions of previously known PMEs. Enzymatic activities of the three isoforms were differently affected by the presence of cations in the incubation medium but, in all cases, infra-optimal cation concentrations induced two opposite effects: a decrease in theV max and an increase in the affinity of the enzymes for their substrate. The presence of cations in the assay modulates both the number of enzyme molecules available to the demethylation reaction and the conformation of the pectin and, in turn, the affinity of the PMEs for their substrate.

    Notizen: Abstract Elicitins form a family of 10-kDa holoproteins secreted by various Phytophthora species. The large-scale purification of parasiticein, a novel elicitin secreted by P. parasitica, led to the determination of its sequence. We have compared the necrotic activities and the primary and secondary structures (determined through circular dichroism) of four elicitins. On tobacco plants, they could be classified into two classes: a, comprising capsicein and parasiticein (less necrotic), and β, comprising cryptogein and cinnamomin (very toxic with a necrosis threshold of 0.1 μg per leaf). The features of elicitin structure which might be involved in the interaction of elicitins with the leaf target cells and that could explain the different necrosis-inducing properties of the two proteins are investigated. About 75% sequence identity was observed between the four elicitins: only two short terminal regions are heterologous, while the central core is mainly conserved. The circular-dichroism spectra showed that the secondary structure of the elicitins was largely conserved. All of them consisted of approx. 50% α-helix with little or no β-structure. Comparisons of the complete sequences, amino-acid compositions, isoelectric points, hydropathy indices and the secondary-structure predictions correlated with the necrotic classification. Alpha elicitins corresponded to acidic molecules with a valine residue at position 13, while β elicitins were basic with a lysine at this position, which appeared to be a putative active site responsible for necrosis induction.

    Notizen: Abstract In culture, Phytophthora fungi — except P. nicotianae — secrete proteins, called elicitins, which cause necrosis on the leaf of the non-host tobacco (Nicotiana tabacum L.) at a distance from the inoculation site, and are responsible for the incompatible reaction. Cryptogein and capsicein are elicitins secreted by P. cryptogea and P. capsici, respectively, and form part of a novel family of 10-kDa holoproteins. On tobacco, the necrotic activity of cryptogein is approx. 100-fold higher than that of capsicein. Using elicitins radioactively labelled in vivo, we have demonstrated that cryptogein and capsicein (i) move from a wound in the stem towards the leaves where they interact directly, (ii) reach their target without undergoing any molecular alteration, (iii) are carried in, and at the same rate as, the sap flow in the xylem, (iv) do not alter the rate of the xylem flow although they are able to provoke drastic damage to the lamina. Consequently, the remote necrotic activity of elicitins does not require any transportable secondary plant elicitor, so the differences in necrotic properties should be due to structural features involved in the interaction of elicitins with the leaf target cells.

    Notizen: Abstract Barley grains (9 samples from 7 cultivars) with nitrogen contents (N) ranging from 1.45 to 4.01% of dry matter were analysed for their amino acid (AA) composition with high accuracy from six different hydrolysates per sample. AA levels in grain increased as linear functions ofN with correlation coefficients close to unity. A comparison with literature data confirmed that the AA composition of any grain sample of normal barley can be predicted from itsN for all phenotypes and genotypes. AAs in grain protein changed as hyperbolic functions ofN which increased for Phe, Pro and Glx but more or less strongly decreased for the other AAs. By plotting AA scores againstN, barley proteins were shown to be always richer than wheat and rye in Val and Phe + Tyr; sometimes richer than both other species forN〈2 (Lys); 2.2 (Leu and Ile); 3.4 (Thr); sometimes intermediate to wheat and rye above the latterN values. They were also intermediate in sulphur AAs forN〈1.9 and drastically poorer forN〉1.9. However, they were richer than both other species in Trp forN〉1.6. The hyperbolic variations of non-protein nitrogen and nitrogen-to-protein conversion factors were determined as a function ofN and also compared with those of wheat and rye.

    Notizen: The possibility of using experimentally determined amino acid composition to assess relatedness between 75 proteins separated by two-dimensional electrophoresis (2-DE) and to identify them was tested on maize. Two independent parameters, the relative Euclidean distance and the correlation coefficient between the amino acid compositions, were evaluated and used. Previous sequence information for 31 out of the 75 proteins made it possible to evaluate the method for the detection of isoforms and for identification. However, the extension of the interrogation beyond maize to all plant sequences raised the problem of false positives that could nevertheless be limited by replications and by using additional information. The efficiency of the method to assess relatedness betwen proteins should make amino acid composition analysis a valuable tool in large protein characterization programs based on 2-DE, by facilitating the transfer of information from one well-documented organ/tissue or genotype to another.
