    Notes: Abstract Generally, many wrinkles form on the human face, and temporary wrinkles eventually become permanent. We evaluated the effects of temporary skin fixation on wrinkle formation after UVB irradiation using the back skin of hairless mice. In the group treated with UVB irradiation immediately after production using cyanoacrylate resin of an artificial groove parallel to the midline, wrinkles formed parallel to the midline, an uncommon direction for wrinkle formation in this mouse model. These wrinkles did not disappear even when the skin was stretched. No such changes were observed in the group in which only the temporary groove alone was produced without UVB irradiation. In 3-D surface parameter analysis, all roughness parameters in the group treated with UVB irradiation immediately after production of an artificial groove were significantly increased relative to the age-matched control group. In contrast, no differences were observed between the group in which only the temporary groove alone was produced without UVB irradiation and age-matched controls. The results of this study suggest that both a temporary groove in the skin and UVB irradiation are necessary for wrinkle formation in the back skin of hairless mice.

    Notes: An attempt ha been made to determine the adsorption ability of surfactants by the human horny layers in vivo. For this purpose, a technique utilizing an acidic dye such as indigo carmine was applicable and convenient. Our in vivo study estimating adsorbing residual amounts of surfactants has revealed that there is a positive correlation between adsorbing and roughening effects. It is suggested that the pathogenesis of hand roughness is primarily related to the cumulative injury of the stratum corenum by adsorbing residual surfactant molecules.

    Notes: Anionic surfactants have various degrees of membrane labilizing action on the isolated lysosome in vitro. This action has been found to correlate with the degrees of skin roughness induced by the surfactants using the circulation method applied to the forearm of human subjects. It has been shown that the acid phosphatase activity in the human horny layer decreases during the in vivo application of the surfactants by the circulation method. The magnitude of the decrease in acid phosphatase activity has been found to correlate to the degree of skin roughness and to in vitro lysosome labilization induced by the surfactants.

    Notes: In order to analyze the role of various surfactants in the abnormal desquamation process of the epidermis in keratodermia type hand dermatitis, the lysosome labilizing effect of surfactants has been investigated by measuring the release of enzymes from lysosomes in vitro using spectrophotometric and gel chromatographic techniques. It has been found that the labilizing effect of surfactants decreases in the order of cationic, anionic, and nonionic surfactants and varies with its alkyl chain length. The relationship of the labilization and skin roughness inducing effect is also discussed.

    Notes: Chemical and ultrastructural studies were conducted to define the relationship between type of melanogenesis and fine structures of melanosomes in normal human epidermal melanocytes. Chemical analysis of epidermal melanin demonstrated that the ratio of eumelanin/pheomelanin varied individually, ranging from 1.31 to exclusively eumelanic. Ultrastructural analysis of fine structures of melanosomes revealed that spheroid melanosomes were frequently observed in melanocytes of the epidermis whose eumelanin/pheomelanin ratio was less than 5. Conversely, ellipsoid melanosomes predominated in melanocytes of the epidermis whose ratio was more than 10.On the basis of these findings, it seems reasonable to conclude that 1) normal human epidermal melanocytes synthesize both eumelanin and pheomelanin and 2) pheomelanin synthesis may be characterized by the presence of spheroid melanosomes whereas eumelanin synthesis is ascribed to ellipsoid melanosomes.

    Notes: Tyrosinase-related protein 1 (TRP1) maps to the brown locus in mice. Although the specific function of TRP1 has been in dispute, mutations in its structural gene result in the formation of brown rather than black melanin. We have investigated the melanogenic function of TRP1 by using immune-affinity purification of the protein and also by using transfection of its gene into fibroblasts to study its characteristics. We show that TRP1 has the ability to oxidize DHICA, a melanogenic intermediate derived from DOPAchrome. In addition, TRP1 has the ability to interact with tyrosinase and significantly stabilize the latter's catalytic function.

    Notes: Further advances leading to more sophisticated and effective suppression of melanogenesis and melanoma growth based on clarification and utilization of common vital factors involved in both processes are reviewed. Induction of depigmentation has been achieved by both glycosylation and its processing inhibitors, which have been found to be critical for the maturation and transport of tyrosinase from ribosomes through GERL-coated vesicles into premelanosomes. Kojic acid, a copper chelating melanogenic inhibitor, can induce inhibition of isolated tyrosinase activity as well as melanization in living pigment cells in in vitro and in vivo systems. This depigmenting effect was found to be due to a concurrent decrease in both eu- and pheomelanin formation. Malignant melanoma principally has accentuated melanosome genesis, which has been utilized to accumulate selectively 10B into melanoma cells using 10B-dopa analogue. Subsequent thermal neutron irradiation induces 10B(n, α)7Li reaction which releases high LET particles within a range of 10–14 μm thus erradicating selectively melanoma at the cellular level. This new therapy has been applied to a human melanoma lesion for the first time, and a successful therapeutic effect on melanoma has been obtained.

    Notes: Purification of tyrosinase inhibitors of hamster melanomas was carried out using tyrosinase binding affinity column chromatography. This method enables the isolation of tyrosinase inhibitors with a 124-fold purification index as compared to that of crude preparation after dialysation. The purified inhibitors consist of a mixture of 5000–6000 and a 310 molecular weight fraction. They also show characteristics of polypeptides which contain glycine, glutamic acid, serine, proline and alanine as main amino acids.

    Notes: The site of condensation and activation of tyrosinase prior to melanosome melanogenesis has been investigated using subcellular fractionation and enzymic analysis of so-called Golgi apparatus of melanoma cells. Two different functional units have been found in the so-called Golgi fraction, the tyrosinase-rich GERL-coated vesicle unit and the TPPase-rich Golgi unit. The tyrosinase-rich unit consists of a tubular structure with a cisternal portion corresponding to the three-dimensional structural characteristics of GERL, and spheroid vesicles, corresponding to the currently accepted criteria for coated vesicles. This fact suggests, in agreement with electron microscopic observations, that the GERL-coated vesicle unit is a specialized portion of smooth endoplasmic reticulum having the function of activating and condensing the tyrosinase synthesized by ribosomes.

    Notes: Further purification of highly active tyrosinase inhibitors by DEAE-Sephadex column chromatography has revealed that the mode of action of the inhibitors from melanomas with different melanogenic status is organelle specific. The inhibitor from amelanotic melanoma has a relatively strong suppressive effect on the tyrosinases in GERL and coated vesicle containing fractions. On the other hand, the inhibitor from melanotic melanoma has no substantial effect on these tyrosinases, although melanosomal tyrosinase is inhibited by both. The present evidence suggests that the defect of melanization in amelanotic melanoma in vivo is related to the tyrosinase inhibition of the smooth membrane system including GERL and coated vesicle by the highly active inhibitor characteristic of amelanotic melanoma.
