Cells of Thiobacillus ferrooxidans obtained by growth on the oxidation of Fe2+ at acid pH are able to also oxidise elemental sulphur. Either with O2 or under anaerobic conditions, Fe3+ can act as a terminal oxidant for S0 oxidation, and observations on these reactions have led to a recent proposal that Fe2+/Fe3+ cycling may be an intermediate in electron transfer from S0 to O2. In this paper we present evidence contrary to this mechanism and also report findings that implicate site 2 energy conservation during S0 oxidation by these cells.
