    Notizen: —The activity of thiamine pyrophosphatase was determined in segments and segment subfractions from normal and crushed sciatic nerves of the cat. Activity of the enzyme was increased in segments near the site of compression. The highest activity of the enzyme was in a fraction rich in tubular and vesicular elements resembling those observed in intact compressed nerves. On morphological grounds these structures have been interpreted as originating from Golgi-like structures.

    Notizen: Abstract: Cyclic AMP-responsive element binding protein (CREB) is critically involved in many important brain functions, including the formation of long-term memory. CREB is the best characterized member of a family of transcription factors (CREB/ATF family) recognized to be important nuclear targets for intracellular signal transduction systems. Here we show, by using different approaches, that CREB is unexpectedly localized to mitochondria of the rat brain. Controlled subcellular fractionation of hippocampus and cerebral cortex showed that both synaptic and nonsynaptic mitochondria exhibited immunoreactivity to the phosphorylated form of CREB (pCREB). Moreover, CREB extracted from synaptic mitochondria is able to be phosphorylated by the catalytic subunit of protein kinase A and dephosphorylated by protein phosphatase 1 or 2B. DNA mobility shift assays showed the presence of binding activity to the calcium—cyclic AMP-responsive element in mitochondrial extracts from hippocampus; this binding complex was specifically supershifted by an anti-CREB antibody. Immunoelectron microscopic analysis of hippocampal subcellular fractions revealed that pCREB immunoreactivity is localized in close association with the inner mitochondrial membrane. These results, together with recent findings describing the presence and phosphorylation of CREB in developing dendrites, suggest that CREB may participate in different mechanisms involved in the communication between extracellular signals and the expression of genes.

    Notizen: Summary The present knowledge on the methodology, specificity, localization, cytochemical significance and experimental variations of the zinc iodide-osmium tetroxide (ZIO) staining are reviewed. Some new findings supporting the concept that ZIO reacts with-SH groups in some structures under specific conditions are also communicated. In this respect the effect of in vitro pretreatment with-SH reagents on ZIO staining, already described in different kinds of synaptic vesicles, was studied in rod photoreceptor outer segments. It was observed that the intra-and extradiskal electron opaque deposits, present in untreated rod outer segments directly stained with ZIO at 4°C for 2 h, were enhanced by previous incubation with dithioerythrytol (DTE), which protects-SH groups and reduce-S-S-bridges; when N-ethyl-maleimide (NEM), a-SH blocking agent was used directly or after DTE, the electron opaque deposits were considerably reduced or abolished. Furthermore the influence of previous incubation with cysteine on ZIO staining was studied in the rat pineal gland. In the controls ZIO made at 4°C for 2 h, reacts strongly with the matrix of the small granulated synaptic vesicles in the pineal nerves; no electron opaque deposits are seen in the membranes of the vacuolar system. Previous incubation with cysteine 5 mM in phosphate buffer makes negative ZIO reaction in the matrix of the synaptic vesicles while with a concentration of 50 mM cysteine, a partial recovery is observed. In both concentrations the membranes of the vacuolar system appeared covered by patchy electron opaque deposits.

    Notizen: Abstract In this paper the demonstration is made that membrane vesicles (liposomes) containing the muscarinic receptor can be formed by polyethylene glycol (PEG) precipitation of detergent extracts of bovine atrial membranes. The incorporation of the muscarinic receptor in these vesicles may be related to the restoration of the heterogeneity and nucleotide modulation of muscarinic agonist binding by PEG precipitation of atrial detergent extracts, previously reported. Vesicles are also formed when detergent solubilized asolectin lipids, alone or in combination with membrane detergent extracts, are precipitated by PEG. The structure of the vesicles seems depend on their lipid and protein composition and the procedure employed for the removal of the dispersing medium. These results indicate that PEG precipitation could be used for the reconstitution of the muscarinic receptor into the liposomes of exogenous lipids.

    Notizen: Abstract Neuronal ATPases comprise a wide variety of enzymes which are not uniformly distributed in different membrane preparations. Since purified vesicle fractions have Mg2+/Ca2+-ATPase, the purpose of the present study was to know whether such enzyme activities have a preferential concentration in a synaptic vesicle fraction in order to be used as markers for these organelles. Resorting to a procedure developed in this Institute, we fractionated the rat cerebral cortex by differential centrifugation following osmotic shock of a crude mitochondrial fraction and separated a purified synaptic vesicle fraction over discontinuous sucrose gradients. Mg2+/Ca2+-ATPase activities and ultrastructural studies of isolated fractions were carried out. It was observed that similar specific activities for Mg2+/Ca2+-ATPases were found in all fractions studied which contain synaptic vesicles and/or membranes. Although the present results confirm the presence of Mg2+ and Ca2+-ATPase activities in synaptic vesicles preparations, they do not favor the contention that Mg2+/Ca2+-ATPase is a good marker for synaptic vesicles.

    Notizen: Summary The uptake and storage of 5-hydroxydopamine after electrical nerve stimulation was investigated in the rat pineal nerves. The drug was injected i.p. immediately after the stimulation of the afferent trunks of both superior cervical ganglia for 20 min, with sq pulses of 1 msec, 25 Hz and 25 V. The animals were killed by decapitation 10 and 30 min after the injection, fixed in glutaraldehyde-osmium tetroxide and processed for electron microscopy. The stimulation was performed under chloral hydrate anesthesia. Anesthetized non stimulated, stimulated-non injected and injected-non stimulated were used as controls. It was found that the storage of the false transmitter, whose presence is revealed by an intense osmiophilia in the synaptic vesicles, when tissues are fixed in glutaraldehyde osmium tetroxide, is significantly reduced in rats injected after stimulation. Evidence is reported supporting the concept that the uptake and the storage of the false transmitter are not prevented by previous nerve stimulation although they may be affected in some way.

    Notizen: Summary The morphology of monoaminergic synaptic vesicles of rat pineal nerves was investigated after fixation with calcium-containing solutions of control and electrically stimulated glands. Stimulated and nonstimulated glands were fixed either in glutaraldehyde or in glutaraldehyde-osmium tetroxide with and without calcium chloride and processed for electron microscopy. It was observed that an electron dense particle, located in the vesicle core, appeared within the small and large vesicles when calcium was present in the fixation fluid. The size and electron density of these particles, as well as the proportion of vesicles showing these particles, depended on the concentration of calcium in the fixative, and the particle could be discerned in unosmicated tissues indicating that the electron opacity was produced by the presence of calcium ions. Although postfixation with OsO4 enhanced the electron density of these particles, it could be established that osmium and calcium affinities for the central core of monoaminergic vesicles are independent of each other and primarily due to different core components. Following electrical stimulation, these electron dense particles become significantly reduced or abolished. A decrease in calcium deposits after stimulation has also been described in cholinergic synaptic vesicles. The hypothesis is formulated that depolarization induces a change in the calcium-binding site apparently resulting in a loss of the calcium ions normally trapped by the vesicle. Calcium leaving the vesicle would then increase the calcium concentration in the cytoplasm thus contributing to the release of neurotransmitter, and would be recaptured by synaptic vesicles under the conditions of relatively mild stimulation at the resting state. Thus synaptic vesicles could act as one of the systems of sequestration for calcium ions instrumental in process of depolarization and release of neurotransmitter.

    Notizen: Summary Electronmicroscope observations have been made on compressed sciatic nerves and preganglionic afferents to the superior cervical ganglia of rats. After 6 hours, the proximal regenerating stumps of both myelinated and unmyelinated axons become filled with enlarged neurotubules and vesicles. Granulated vesicles of 750–900 Å, having a dense core become abundant in all types of regenerating axons and they increase in number after 24 hours. The vesicular material is formed by dilatation and pinching off from neurotubules. The existence of a neurotubular system within the axon, connected with the Golgi complex at the perikaryon and involved in the formation of vesicles, is postulated. The presence of granulated vesicles in all types of regenerating axons and nerve terminals is discussed in relation with their possible functional significance. The distal stumps of compressed sciatic nerves show, after 6 hours, a considerable increase in membranous material within the axoplasm mainly represented by multivesicular and lamellar bodies. This reaction, which is interpreted as being autolytic, is compared with the regenerative reaction of the proximal stump.
