    Notes: Summary Growth of human mammary tumor cells ZR-75-1 is stimulated by estradiol (E2), epidermal growth factor (EGF) and insulin-like growth factor I (IGF-I). In these cells ribosomal protein S6 kinase is activated by EGF, IGF-I, insulin and phorbol 12-myristate 13-acetate (TPA) but not by E2. The human mammary tumor cell line MDA-MB 231, which is E2-receptor negative, has receptors for EGF, IGF-I and insulin but is unresponsive to these factors in terms of growth and S6 kinase activation. The role of protein kinase C (PKC) in the activation of S6 kinase by growth factors and TPA was investigated in ZR-75-1 cells. Down regulation of PKC activity by treatment with TPA for 48-h blocks the stimulation of S6 kinase by TPA but leaves the activation by EGF, IGF-I and insulin unaffected. In intact ZR-75-1 cells staurosporine blocks activation of S6 kinase by EGF and TPA, however with different IC50. The results show that S6 kinase is not activated by estradiol, that its activation by EGF, IGE-I and insulin does not depend on the presence of PKC activity and that its activation by TPA is mediated by a different (PKC-dependent) pathway.

    Notes: Summary Two isoenzymes of cAMP-dependent protein kinases were found in MCF-7 cytosol. The regulatory (cAMP-binding) subunit of protein kinase I (predominant form) has an apparent molecular weight (MW) of 49,000 and the two forms of regulatory subunits of protein kinase II have MWs of 52,000 and 54,000. Substantial amounts of the 49,000 protein cochromatographed on DEAE cellulose with protein kinase II. The quantities of protein kinase holoenzyme activity are strongly influenced by extraction procedures: the use of EDTA and of the protease inhibitor benzamidine can lead to extensive dissociation. On the other hand, high yields of cAMP-dependent protein kinase holoenzyme activity were consistently obtained with 150 mM KCl.

    Notes: Abstract Bioreactor performance studies of the recently developed “horizontal stirred tank” with a volume of 421 have been carried out for fermentation with Trichosporon cutaneum. Quantification on the basis of measured oxygen transfer capacity and power consumption is presented and compared with data for a conventional vertical tank bioreactor. During the experiments it has been observed that two different forms of morphology of Trichosporon, i.e. the “normal” yeast-form (Y) with single cells and a mycelium-form (M) with filamentous cells, are present in the horizontal stirred tank when working with the original strain (DSM 70698). After separation both forms were characterized and later on used for bioreactor performance studies in the horizontal and vertical stirred tank. Results of oxygen efficiency show the drastic effect of the morphology change on bioreactor performance. Finally different bioreactors are quantitatively compared on the basis of oxygen transfer, power consumption and productivity using the reference fermentation system Trichosporon cutaneum.

    Notes: Solid cyclodecyl p-toluenesulfonate decomposes spontaneously at room temperature into p-toluenesulfonic acid and cis-cyclodecene. Pure cyclodecyl p-toluenesulfonate gives by acetolysis a mixture of 80% trans- and 20% cis-cyclodecene. The non-classical, transannular course of these eliminations has been investigated by using starting material labeled with 14C. By systematic degradation of the cyclodecenes obtained it has been found that the amount of transannular hydride shifts in products of spontaneons decomposition differs considerably from that in products of acetolysis. These results explain the disturbing discrepancies observed in earlier experiments, in which partly decomposed cyclodecyl p-toluenesulfonate must have been used.
