    Notes: Abstract– The GABA content of the spinal cord and of approx 70 discrete rat brain nuclei is measured with a simple rapid semi-automated fluorimetric assay, after prevention of post-mortem effects with 3-mercaptopropionic acid. We found that microwave irradiation produced decreases in the GABA contents of the microdissected zona reticulata of the substantia nigra, indicating that microwave fixation is not suitable to measure GABA levels in microdissected brain nuclei. In approx 70% of the nuclei in the anterior half of the brain the GABA concentration was found to be between 41 and 90nmol GABA/mg protein. The GABA content varied from 11 to 40 nmol GABA/mg protein in the posterior half of the brain. High GABA levels were found in some hypothalamic nuclei, the globus pallidus and eminentia mediana. An extremely high GABA level was found in the zona reticulata of the substantia nigra. GABA is unevenly distributed in the striatum. The highest concentration was found in the caudal part and in the ventral region at any level of the striatum. In the spinal cord the highest concentration of GABA was in the sacral region.

    Notes: The aim of this study was to characterize neurohypophyseal hormone receptors in the brain of the canary (Serinus canarius) by using autoradiographical and biochemical procedures with a radioiodinated vasotocin analogue, [125 l]d(CH2)5[Tyr(Me)2, Thr4, Orn8, Tyr-NH29]vasotocin ([125 I]-OTA). This synthetic analogue was used previously to identify a population of oxytocin-like receptors in the rat brain that have high affinity for vasotocin. In vitro autoradiography revealed high affinity binding sites for [125 I]-OTA in the canary brain in the archistriatum surrounding the nucleus robustus archistriatalis. Scatchard analysis of [125 I]-OTA binding to a synaptic membrane fraction prepared from the archistriatal region including the nucleus robustus archistriatalis indicated the presence of a single population of binding sites (Kd= 0.05 nM; Bmax= 4 fmol/mg protein).Displacement studies revealed that the order of potency of certain peptides to inhibit [125 I]-OTA binding was as follows: vasotocin (Ki= 0.4 nM) 〉 oxytocin = vasopressin 〉 mesotocin (Ki= 8.0 nM). The administration of testosterone to female canaries did not affect [125 I]-OTA labelling in the archistriatum detected by autoradiography and did not influence [125 I]-OTA binding constants in the membrane binding assay. In conclusion, this study provides morphological and biochemical evidence of a vasotocin-target site in the archistriatum in close association with the central vocal control circuit in the canary brain.

    Notes: A close contact between the dam and the litter is essential for the normal development of the hypothalamic-pituitary-adrenal (HPA) axis in rats and mice. Maternal signals, as licking and feeding, have been shown to sustain the HPA axis of the pups in a hypo-responsive state. Disruption of this mother–pup interaction by 24 h of maternal deprivation activates the otherwise quiescent stress system of the neonates, resulting in an enhanced adrenal sensitivity to adrenocorticotropic hormone (ACTH) and a decreased expression of central HPA markers, such as corticotropin-releasing hormone (CRH). However, the dynamics of these central and peripheral changes over the 24h period are largely unknown. In this study, we examined the time course of some of the central and peripheral indices of HPA activity during 24 h of maternal deprivation. We measured corticosterone and ACTH in the blood as well as CRH, mineralocorticoid and glucocorticoid receptor expression in the brain. Our results demonstrate that each of the components of the HPA axis responds to maternal deprivation at different time points following removal of the mother and with a very specific time course. The main activation of the HPA axis occurred between 4 h and 8 h of maternal absence. By contrast, during the second half of the deprivation period, negativefeedback mechanisms restrained the further increase in ACTH and corticosterone release. We conclude that maternal deprivation triggers a cascade of sequential changes at the various levels of the stress system, and that measuring only one aspect of the system at one time point does not accurately reflect the dynamic alterations of the HPA axis.

    Notes: To investigate the role of brain glucocorticoid (GR) and mineralocorticoid receptors (MR) in centrally evoked blood pressure responses, the effects of intracerebroventricular (i.c.v.) administration of angiotensin II and vasopressin were studied in adrenalectomized rats with and without corticosterone or aldosterone replacement. Five groups were examined: (i) Adrenalectomy (ADX); (ii) ADX + a subcutaneously implanted 20-mg corticosterone pellet (low corticosterone); (iii) ADX + 100 mg corticosterone pellet (high corticosterone); (iv) ADX + 6 µg/24 h aldosterone via Alzet minipump (Aldo); and (v) Sham adrenalectomy (Sham). Pressor responses to 150 ng angiotensin II and 50 ng vasopressin i.c.v. were determined in freely moving rats using biotelemetry. The results show that, compared to sham rats, ADX rats showed significantly reduced pressor responses. This reduction of the pressor response to angiotensin II could be reversed and even further enhanced by replacement of the ADX rats with high corticosterone concentrations. In contrast, with aldostosterone, a depressor type response was observed. Corticosterone replacement could not restore the pressor response to vasopressin. We conclude that the pressor response to centrally administered vasoactive substances is substantially attenuated by removal of the adrenals and that, in the case of angiotensin II, this is due to the lack of high concentrations of circulating corticosterone occupying both MR and GR. However, predominant MR occupancy appears to play an opposite role and attenuates the angiotensin II-induced pressor response.
