    Notes: Isolate-specific resistance of 71 cultivars and breeding lines of wheat (Triticum aestivum) to septoria tritici blotch was evaluated in six field trials in the Netherlands, Switzerland and the UK between 1995 and 1997. Each plot was inoculated with one of six single-pycnidium isolates of the pathogen Mycosphaerella graminicola. There were strong interactions between wheat lines and M. graminicola and the line-by-isolate interactions were stable over the six trials. Lines with specific resistance or specific susceptibility to each of the isolates were identified. Specific resistance to isolate IPO323 was especially common, being carried by 22 lines from 10 countries. The results confirm that line-by-isolate interactions in septoria tritici blotch of wheat are effective in adult plants in field conditions, and are not simply confined to seedlings. Wheat lines with good, quantitative resistance to all or most isolates were identified, including lines from Brazil, the USA and seven European countries. These may be useful as sources of resistance in wheat breeding.

    Notes: Abstract Monospore isolates of Mycosphaerella graminicola considered to originate from one ascus were analysed by the polymerase chain reaction (PCR) with 32 RAPD primers. Eighteen of these revealed three classes of polymorphisms, which enabled a RAPD-based tetrad analysis. Four pairs of isolates resulting from a single diploid nucleus were determined. A procedure to cross these isolates was developed to investigate the mating system. Three of six crosses were successful, and the segregation of mating types in accordance with the tetrad analysis strongly points to a bipolar heterothallic mating system in M. graminicola. Random ascospore progenies from the successful crosses, each comprising 54 isolates, were studied with three primers to determine the mode of inheritance of the RAPD markers. Mendelian segregation and recombination of RAPD markers was observed in all progenies.

    Notes: Summary Winter wheat cultivars released in the Netherlands before 1930 carried durable resistance to yellow rust. Cultivars released in the period between 1930 and 1950 often were durably resistant while recent cultivars infrequently showed durable resistance. This durable resistance was not difficult to transfer to new cultivars. Twenty nine older cultivars with durable resistance and eight recent non-durably resistant cultivars were tested in the seedling stage and in the adult plant stage against 12 West-European yellow rust races and against some non-European races in the seedling stage only. The adult plant tests were carried out in ‘race nursery’ tests in the Flevopolder. Per race nursery all 37 cultivars, planted in hills of about 20 plants on both sides of the highly susceptible cv. Michigan Amber, were exposed to one race. The infection type of each cultivar-race combination was scored on 0 to 9 scale once in the seedling stage and twice in the adult plant stage. In the race nurseries the percentage leaf area affected was evaluated three times to be used to calculate the area under the disease progress curve (AUDPC). This AUDPC multiplied with the mean infection type in the field gave the susceptibility index (SI). The infection types were classified into resistant (R), intermediate (I) or susceptible (S) when the infection types were 0 to 3, 4 to 6 or 7 to 9, respectively. Four categories of resistance were discerned on the basis of the three infection type scores: 1) RRR, overall resistance; complete or near-complete resistant at all stages. 2) SRR, adult plant resistance, complete- or near-complete resistant at the adult plant stage only. 3) SRS and SSR, temperature sensitive resistance, the resistance changed from the one evaluation data to the other. 4) SSS and an SI lower than that of Michigan Amber, partial resistance. The frequencies of overall, adult plant and temperature sensitive resistance were 1.4, 52 and 54% in the older cultivars and 40, 62 and 22% in the recent ones, respectively. Among the older cultivars all had a fair to high level of partial resistance, the SI being on average only 20% of that of Michigan Amber, while most cultivars also seemed to carry temperature sensitive resistance. The partial resistance of the recent cultivars was of a much lower level with a mean SI compared to that of Michigan Amber of 61%. Partial resistance was highly correlated (r = −0.94) with the mean resistance scores from the Dutch Recommended Cultivars Lists. It was concluded that partial resistance and temperature sensitive resistance were the major components of the durable resistance in the older cultivars.

    Notes: Abstract The in vitro production of pycnidia bySeptoria tritici was examined on six media reported to induce the formation of fruiting bodies. Among 26 freshly isolated cultures from various parts of the world, consistent differences in growth type were found which were only partially influenced by nutritional and environmental conditions. Cultures with yeast-like growth produced hardly any pycnidia or pseudopycnidia, while cultures with intermediate or mycelial growth types produced them frequently. Incubation in continuous darkness induced intermediate to mycelial growth types rather than yeast-like growth types in some cultures, and concomitantly the production of more pycnidia. Potato-dextrose agar induced intermediate to mycelial growth types and production of (pseudo)pycnidia more often than V8 agar and wheat leaf extract agar, which had previously been reported to be especially beneficial to (pseudo) pycnidium formation byS. tritici. Isolates with a consistently yeast-like growth type, producing (virtually) no fructifications under any of the experimental conditions, were slightly stimulated to form pseudopycnidia on water agar supplemented with sterile pieces of maize, wheat or carnation leaves.
