    Notes: A quartz crystal resonator and an atomic force microscopy were applied for the measurement of local viscoelasticity and surface morphology. For the reduction of signal noise from the quartz crystal resonator, we designed an oscillation circuit based on a referential quartz crystal method. A polystyrene bead-coated quartz crystal was used as a signal quartz crystal, and a bare Au quartz crystal was used as a reference. By approaching the cantilever of atomic force microscopy to the surface of working quartz crystal, the differential resonant frequency in the two quartz crystals showed changes by the interaction between the tip and the quartz crystal. The changes of differential resonant frequency in the two quartz crystals were influenced by the local viscoelasticity. The image resolution of differential resonant frequency was observed under 80 nm for the polystyrene bead-coated quartz crystal. © 1999 American Institute of Physics.

    Notes: A facile chemical route to prepare a well-defined Ni powder on a mass-production scale without external heating was suggested. A highly concentrated Ni complex was formed between NiCl2 and N2H4 solution, and subsequent addition of NaOH solution triggered serial reactions from the Ni complex decomposition via Ni(OH)2 formation to the reduction of a Ni(OH)2 gel into well-defined Ni spheres. The high temperature necessary for the reaction was attained not by external heating but by exothermic reactions such as complex formation and reduction. The preparation of monodisperse and spherical Ni powder was attributed to uniform distribution of the reducing agent (N2H4) within the Ni(OH)2 gel.

    Notes: Penicillium amagasakiense secreted glucose oxidase (GOD) with a highly diminished carbohydrate moiety (aglyco-GOD) in the presence of 20 mg/l tunicamycin. To examine enzymatic properties of aglyco-GOD, this and native GOD (glyco-GOD) were purified to homogeneity from the culture medium with or without tunicamycin. The carbohydrate content of aglyco-GOD was 2.9% and the Mr of it and its subunit were 130 000 and 70 000, respectively. On the other hand, those from glyco-GOD were 20.4%, 150 000 and 80 000, respectively. Nonetheless, they showed remarkably similar catalytic properties to each other: Km values of 3.4 and 2.7 mM and Vmax values of 320 and 270 U/mg protein. pH optima 5.5 and 6.0 were observed for aglyco and glyco-GOD, respectively. Isoelectric focusing showed a strikingly similar pattern with each GOD having one major band of pI 4.2 and 2 minor bands between pI 4.3 and 4.5, which implies that microheterogeneity of GOD was not due to heterogeneous glycosylation. Up to 35°C, both enzyme preparations were stable for 30 min while both were considerably inactivated after 30 min at 45°C. Therefore, we conclude that the carbohydrate moiety plays no significant role in the enzymatic function of P. amagasakiense GOD.

    Notes: Abstract A novel metabolic pathway for creatinine, degradation of creatinine to glycine via N-methylhydantoin, N-carbamoylsarcosine and sarcosine in turn, was found to operate in Pseudomonas putida 77. N-Methylhydantoin, sarcosine and glycine were detected as metabolites from creatinine, and enzyme activities responsible for the degradation of creatinine, N-methylhydantoin, N-carbamoylsarcosine and sarcosine were observed in a crude cell extract, while neither creatinine amidohydrolase activity nor creatine were detected at all. It is suggested that creatinine was utilized as a source of both carbon and nitrogen at the same time, and that N-methylhydantoin was also utilized efficiently.

    Notes: The present work deals with an applicability of the local approach to assess in-plane sizeeffects among different sized compact tension (CT) specimens. To characterize ductile crack growthof typical nuclear materials, SA515 Gr.60 and SA516 Gr.70 carbon steels, finite element analysesemploying modified GTN and Rousselier models as well as fracture toughness tests were carried out.Material damage parameters were calibrated using standard CT specimens and reflected to predictfracture resistance (J-R) curves of larger CT specimens. Since comparison results betweennumerically estimated J-R curves and experimentally determined ones corresponded well, it isanticipated that the local approach might be used as a promising tool for ductile fracture evaluationincorporating the in-plane size effect

    Notes: Summary The soil isolate Cellulomonas cellulans AM8 produces an extracellular l-amino acid oxidase (L-AAO) with broad substrate specificity. The strain produced up to 0.35 unit (U)/ml of the extracellular L-AAO in a simple medium containing glycerol and yeast extract. The enzyme was easily purified up to 30 U/mg protein using Phenyl-Sepharose fast flow. The purified enzyme migrated as single band on sodium dodecyl sulphate-polyacrylamide gel electrophoresis (SDS-PAGE) with a molecular mass of 55 kDa. On native PAGE the molecular mass was approx. 300 000 kDa, which may be due to aggregation. With the exception of glycine, proline, and threonine, all the amino acids normally constituting proteins were oxidized. The V max values from 0.7 to 35.2 U/mg for aspartic acid and lysine, respectively, and the K m values from 0.007 to 7.1 mm for cysteine and valine, respectively, were obtained at 25° C and pH 7.0 in oxygen-saturated solutions. The L-AAO had a pH optimum of 6.5–7.5. It was stable for several months at — 30° C and for some days at 35° C. Ferricyanide served as an electron acceptor with a V max of 50 U/mg and K m for 0.3 mm with phenylalanine as the substrate.

    Notes: The reliability of QFP (Quad flat package) solder joint using Sn-8Zn-3Bi solder pasteunder the thermal shock test was investigated. Considering the environmental restriction such asROHS, the QFP Cu LF (Lead-frame) was coated with lead-free materials (Sn, Sn-3Bi). To analyzethe reliability under thermal shock treatment, the samples were placed in the thermal shock chamber(248K/423K, Dwell time: 30min). During the thermal shock test, the solder joint cross-sectionswere observed every 500 cycles. No crack initiation and propagation was observed through all typeof plated Cu LF. The measured pull strength slightly decreased, as the number of cycles increased.After 1000 cycles, the pull strength of Sn and SnBi plated Cu LF reduced by 30% and 20%,respectively, compared with that of initial condition. Observing the fracture surface morphology byFE-SEM, the fracture mode changed and the IMC fractured area on the both fracture surface wasincreased. The IMC was identified as γ-Cu5Zn8 by chemical composition analysis using EDS. Thereduced pull strength was affected by IMC fracture and fracture mode change. However, the pullstrength does not decrease steeply but gradually. Consequently, the Sn-8Zn-3Bi solder joint showsthe reliable solder joint strength, fracture mechanism, and compatibility with lead-free plated Cu LFduring the thermal shock temperature of 248K to 423K
