    Notizen: Raman scattering in liquid (and in some cases in solid) isotopic mixtures of HC1 and DC1 is analyzed to prove recent theories by Bratos and Tarjus [Phys. Rev. A 32, 2431 (1985)], Logan [Mol. Phys. 58, 97 (1986)], and Knapp [J. Chem. Phys. 81, 643 (1984)] on vibrational line broadening in liquids. The concentration and temperature dependencies of isotropic [Ji(ω)] and anisotropic [Ja(ω)] line shapes have been studied between triple point (Tt) and critical temperature (Tc). It has been found that in accordance with the Bratos–Tarjus theory, Ji (ω) is much more sensitive of isotopic composition of the liquid than Ja(ω). An analysis of the concentration dependence of the broadening parameters near Tt illustrates the importance of cross correlations between the environmental broadening and the resonant intermolecular coupling. The spectral activity of three-particle resonant transfer also becomes significant. From the change of the maximum of Ji(ω) with isotopic dilution, which is a linear function of mole fraction, the dipole moment derivative δμ/δq is estimated to be more than twice that of its gas phase value. The asymmetry of the isotropic bands of both HC1 and DC1 changes with concentration at constant temperature. With increasing temperature, Ji(ω) of pure and diluted samples narrows as T−0.5 and T−0.3, respectively. Ji(ω) has been found to be intermediate between the slow and the fast modulation limit. From the high frequency wing of Ji (ω) the time constant of the zeroth order memory function was obtained. Its activation energy increases with increasing T. This is in qualitative agreement with the temperature dependence of the Enskog collision time and the spin–rotational correlation time. Taking into account the results of the Bratos–Tarjus theory, orientational correlation times τ(2) are determined from Ja (ω). The Raman method yields τ(2) values which are twice as long as those determined from NMR relaxation.

    Notizen: The oscillation of ion bunches around a strongly negative grid in a double plasma device is studied with one–dimensional particle–in–cell simulation. The system exhibits feedback amplified virtual anode oscillations in the target chamber. The undriven system is shown to perform relaxation processes which can be described by the van der Pol equation for high nonlinearity. The periodically driven system exhibits nonlinear phenomena like mode locking and periodic pulling, which are in quantitative agreement with theoretical predictions for the driven van der Pol oscillator. For large driver amplitudes period doubling and weak chaos are found. © 1997 American Institute of Physics.

    Notizen: Experiments are reported on oscillations that arise in a double plasma device when plasma production is restricted to the source chamber and the separating grid between the two chambers is biased negatively. The free oscillating system shows periodic pulling which is a typical behavior of driven van der Pol type oscillators. The second interacting frequency is identified to be half the ion plasma frequency at the sheath edge on the source side. With the help of particle in cell simulations the concept of virtual anode oscillations (VAO's) as the underlying oscillation mechanism is investigated and the van der Pol character of these is revealed. When applied to the experimental conditions, the VAO-model predicts correct oscillation frequencies. It gives a new interpretation of the scaling of these with plasma density and grid bias, and is compatible with earlier findings. © 1997 American Institute of Physics.

    Notizen: We used transcranial Doppler ultrasonography in 45 patients to investigate if changes in haemodynamics in the major arteries of the brain base occurred after lumbar puncture and whether or not patients with or without post-lumbar puncture headache differ with respect to their cerebral haemodynamic parameters before and after lumbar puncture. Before lumbar puncture, patients with post-lumbar puncture headache differed from patients without post-lumbar puncture headache in that they showed significantly higher flow velocities and significant asymmetry of flow velocities with lateralization to the right (p£ 0.05). Patients without post-lumbar puncture headache, on the other hand, showed non-significant flow velocity lateralization to the left. Forty-eight hours after lumbar puncture, both groups demonstrated symmetrical flow velocities. In addition, only patients with post-lumbar puncture headache showed a significant reduction in the flow velocity of the right middle cerebral artery (p£ 0.05). These findings suggest that it is not only absolute flow velocity that plays a part in the event of headache, the interhemispheric relation of cerebral haemodynamics also plays a fundamental role.

    Notizen: Both, the isotropic and anisotropic Raman line shapes of orthobaric liquid hydrogen and deuterium bromide are measured at 295 K and close to the triple point (185 K). Concentration dependent studies in HBr/DBr mixtures allow an experimental separation of the resonant transfer mechanism from other line broadening mechanisms. It is found that the anisotropic spectra are nearly unaffected by isotopic dilution, whereas the isotropic ones show a blue shift and a narrowing with increasing isotopic dilution. The overall contribution of the transfer mechansim to the line broadening becomes most pronounced at the lower temperature. The results are discussed in terms of a recent Raman line shape theory of Bratos and Tarjus.

    Notizen: Environmental factors are known to regulate the molecular differentiation of neocortical interneurons. Their class-defining transmitter synthetic enzymes are the glutamic acid decarboxylases (GAD); yet, fairly little is known about the developmental regulation of transcription and translation of the GAD-65/67 isoforms. We have characterized the role of neuronal activity, neurotrophins and afferent systems for GAD-65/67 expression in visual cortex in organotypic cultures (OTC) compared with in vivo in order to identify cortex-intrinsic regulatory mechanisms. Spontaneously active OTC prepared at postnatal day 0 displayed from 10 days in vitro (DIV) onwards 12–14% GAD-65/GAD-67 neurons similar to in vivo. However, GAD-65 mRNA was higher, whereas GAD-67 protein was lower, than in vivo. During the first week neurotrophins increased whereas the Trk receptor inhibitor K252a and MEK inhibitors decreased both GAD mRNAs and proteins. After 10 DIV GAD expression no longer depended on neurotrophin signalling. Activity-deprived OTC revealed only 6% GAD-67 neurons and mRNA and protein were reduced by 50%. GAD-65 mRNA was less reduced, but protein was reduced by half, suggesting translational regulation. Upon recovery of activity GAD mRNAs, cell numbers, and both proteins quickly returned to normal and these ‘adult’ levels were resistant to late-onset deprivation. In 20 DIV activity-deprived OTC, only neurotrophin 4 increased GAD-65/67 mRNAs, rescued the percentage of GAD-67 neurons and increased both proteins in a TrkB-dependent manner. Activity deprivation had thus shifted the period of neurotrophin sensitivity to older ages. The results suggested neuronal activity as a major regulator differentially affecting transcription and translation of the GAD isoforms. The early presence of neuronal activity promoted the GAD expression in OTC to a neurotrophin-independent state suggesting that neurotrophins play a context-dependent role.

    Notizen: Substrate-bound guidance cues play an important role during the development of thalamocortical projections. We used time-lapse video microscopy to study the growth behaviour of thalamic axons on different substrates. On embryonic cortical membranes and on a pure laminin substrate, thalamic fibres advanced relatively slowly (∼15 μm/h) and on average their growth cones retracted transiently every ∼5 h. In contrast, on membranes prepared from early postnatal cortex, thalamic fibres grew twice as fast and spontaneous growth cone collapse occurred ∼8 times less often. Experiments in which we used the sugar-binding lectin peanut agglutinin or heat inactivation to change the membrane properties indicated that these differences are due to growth-supporting molecules on postnatal cortical membranes. When offered a choice between embryonic and postnatal cortical membranes, thalamic axons preferred the postnatal membrane substrate. Time-lapse imaging revealed that borders between these two substrates effectively guided thalamic fibres, and in most cases axons changed their direction without collapse of the growth cone. Our results suggest that thalamic axons can be guided by the spatial distribution of growth-promoting molecules in the developing cortex.
