    Notes: Abstract The glycosaminoglycan (GAG) concentrations in cerebral cortices of two strains of cesarean-derived, barrier-raised rats were compared as functions of growth and senescence. The qualitative distribution of the GAG was similar in both strains of rats at all ages. The GAG present were hyaluronic acid, chondroitin 4(6)-sulfate, heparan sulfate and a non-uronic acid containing glycoconjugate which was partially degraded by endo-B-D-galactosidase (keratanase). The chondroitin 4(6)-sulfate was remarkably constant throughout the lifespan studied and similar in both strains of rats. A non-uronic acid containing component in the GAG fraction increased significantly to a peak at 4 months of age in the Fischer rat and at 6 months of age in the King rat, but subsequently declined to the pre-peak baseline level in senescence. A parallel rise and fall of sulfate and sialic acid in the GAG fraction was observed in both strains of rats. Our data suggest that the non-uronic acid containing component is a sulfated glycoprotein-like glycoconjugate similar to keratan sulfate. The glycoconjugate peak in the cerebral cortex may be associated with the rapid proliferation of glyco-protein-rich glial cells. The gradual decline in keratan sulfate with age may represent the gradual alteration of glial glycoconjugates.

    Notes: Abstract The concentration and distribution of the glycosaminoglycans (GAG) in the cerebellum of two strains of cesarean-derived, barrier-raised rats were compared as a function of growth and senescence. The GAG in the cerebellum were the same as those found in the cerebral cortex: hyaluronic acid, chondroitin 4(6)-sulfate, heparan sulfate and a non-uronic acid containing glycoconjugate degraded by endo-B-D-galactosidase which was labelled “keratan sulfate.” The concentration of the GAG in the cerebellum was half that in the cortex, although the relative distribution of the individual GAG components was the same as in the cerebral cortex. The concentration of the GAG in the cerebellum, relative to the DNA concentration, was one-tenth that in the cerebral cortex. As in the cerebral cortex, chondroitin 4(6)-sulfate was remarkably constant throughout the lifespan studied. In both strains of rats the concentration of “keratan sulfate” showed a peak between 2 and 3 months of age, and then gradually declined. These growth related peaks of keratan sulfate appeared to be associated with the increase of myelinated fibers and/or the proliferation of the Purkinje cell dendrites. The gradual reduction of the “keratan sulfate”: chondroltin 4(6)-sulfate ratio may alter the movement of water and other molecules accounting for the reduction in extracellular space observed by others.

    Notes: Abstract If meningomyelocele is indeed a progressive intrauterine process, then early delivery or possibly intrauterine repair of meningomyelocele becomes an issue. Util-izing the delayed splotch (Spd) mouse, a genetically transmitted neural tube defect model, we looked for evidence of abnormalities of neural tissue exposed to amniotic fluid. Affected embryonic and fetal mice were examined with the light microscope, and also with the transmission and scanning electron microscope. Neuronal development and programmed cell death paralleled normal fetal development. No evidence of inflammation on or within the exposed neural tissue was observed. Because the vascular supply to the alar and basilar plate are different, vascular development was also examined and no difference could be found. In conclusion, we found no evidence of deterioration of the exposed neural tube during the gestational period of a mouse, which suggests that exposure of unneurulated spinal cord to amniotic fluid is not a risk factor to the fetus with a neural tube defect.

    Notes: Smooth muscle cells were demonstrated adjacent to the aqueous outflow pathway in rabbit eyes. The cells were arranged as a discontinuous band, one to three layers thick and extended from the termination of Descemet's membrane to the ciliary body. Most of the cells were oriented with their long axes at right angles to the aqueous plexus, but many were arranged in a direction parallel to the plexus. The organization of the cells suggests that their contraction would exert a bi-directional force, and therefore, affect aqueous outflow resistance.
