    Notizen: Abstract Methods for measuring neutrophil adherence, phagocytic-nitroblue tetrazolium (NBT) reducing activity and chemiluminescence were applied to canine whole blood as means for routine assessment of neutrophil functions. The phagocytic-NBT reduction test appeared to be useful for monitoring the NBT reducing activity of phagocytic cells associated with phagocytic functions. Ethylene diamine tetraacetic acid suppressed both the adherence and the phagocytic-NBT reducing activity of neutrophils. Increased phagocytic-NBT reduction and an enhanced chemiluminescence response were observed in dogs with neutrophilia. These methods provide a rapid and practical screening procedure for measuring selected phagocytic functions in canine whole blood.

    Notizen: Abstract Clonal assays for haematopoietic progenitors were performed on mononuclear cells isolated from bone marrow samples collected from the sternebrae of normal horses. Colony-forming units-erythroid (CFUE), burst-forming units-erythroid (BFU-E), colony-forming units-granulocyte/monocyte (CFU-GM) and colony-forming units-fibroblastic (CFU-F) were assayed, and reference values for clinically normal horses were established. The mean numbers of CFU-E, BFU-E and CFU-GM per 5 × 104 cells were 329 ± 48, 30 ± 5 and 131 ± 13, respective. The mean number of CFU-F was 49 ± 6 per 2 ± 10 cells. The numbers of CFU-E and BFU-E were linearly related to the concentrations of fetal bovine serum (FBS), bovine serum albumin (BSA) and the cell number plated. The number of CFU-E were increased in the presence of erythropoietin (EPO) but its presence was not required for colony growth. BFU-E had an absolute requirement for EPO but the number of BFU-E was not linearly related to dose of EPO. Methods for clonal assays of equine hematopoietic progenitors are described.

    Notizen: Abstract Pathogenic mechanisms that underlie feline leukaemia virus subgroup-C (FeLV-C) induced erythroid aplasia are unknown. FeLV-C infection is associated with higher serum levels of interferon-γ (IFN-γ) and tumour necrosis factor-α (TNF-α), which may act synergistically to cause haemopoietic suppression. In the present studies, the synergistic effects of TNF-α and IFN-γ on feline bone marrow progenitors in vitro were evaluated. Bone marrow mononuclear cells from specific-pathogen-free cats were exposed to TNF-α (100 and 200 pg/ml) and IFN-γ (100 or 200 units/ml), alone or in combination, for 2 h before plating for clonal assays of colony forming units. Our results show that TNF-α and IFN-γ in combination caused marked suppression of feline colony forming units-erythroid (CFU-E), burst forming units-erythroid (BFU-E), and colony forming units-fibroblasts (CFU-F), whereas colony forming units-granulocyte/macrophage (CFU-GM) were minimally affected. The same concentrations of TNF-α and IFN-γ alone had minimal effects on CFU-E, BFU-E and CFU-F. These results suggest that TNF-α and IFN-γ may play a significant role in regulating haemopoiesis in cats and may be involved in the pathogenesis of erythroid aplasia in cats infected with feline leukaemia virus.

    Notizen: Abstract The use of orally administered interferon-α as a treatment for retroviral disease was evaluated in the experimental model of feline leukaemia virus (FeLV)-induced erythroid aplasia. Progressive anaemia, FeLV viraemia, and leukopenia developed in cats inoculated with the Kawakami-Theilen isolate of feline leukaemia virus (FeLV-KT). A treatment regimen with orally administered recombinant interferon-α or natural interferon-α as employed in this study had no significant effects on viraemia, course of disease, or differential leucocyte counts. The results of this study did not reveal any beneficial effects of human interferon-α administered by the oral route for treatment of cats with experimentally induced FeLV infection.

    Notizen: Abstract Tumour necrosis factor-α (TNF-α) levels were measured in supernatants of macrophage cultures infected in vitro with feline leukaemia virus (FeLV) and in serum, bone marrow plasma, and supernatants of bone marrow mononuclear cell (BMMC) cultures from cats infected with an anemogenic strain of FeLV. TNF-α levels significantly increased (p 〈 0.05) in the supernatants of in vitro FeLV-infected macrophage cultures compared with uninfected cultures. TNF-α levels in serum and culture supernatants of BMMC from FeLV-infected cats were also significantly increased (p 〈 0.05) compared with baseline values. Pentoxifylline is a dimethylxanthine derivative that inhibits TNF-α production. Pentoxifylline treatment of FeLV-infected macrophage cultures caused a significant decrease in TNF-α levels. Pentoxifylline given orally to FeLV-infected cats from week 0 to week 5 post-inoculation did not cause significant inhibition of TNF-α levels. Although a trend toward higher numbers of erythroid and fibroblast progenitors was observed in pentoxifylline-treated cats, the changes were not significantly different. The persistent increase in TNF-α levels in serum and culture supernatants of BMMC from FeLV-infected cats suggests that TNF-α may be important in the pathogenesis of hematopoietic suppression.
