    Notes: When cotyledons of mung bean [Vigna radiata (L.) Wilczek] were treated with spermidine (3 mM) during the first 6 h of imbibition, the development of α-amylase activity in cotyledons during the following 3 days was severely inhibited (75%) This inhibition was due to a slower accumulation of α-amylase protein, which in turn resulted from an inhibition of α-amylase synthesis. The rise in the level of α-amylase mRNA in cotyledons was also inhibited by spermidine treatment. However, the degree of inhibition of mRNA accumulation (40%) was not so marked as that of the activity of α-amylase synthesis (80%). These results are discussed in relation to the mode of action of spermidine on α-amylase expression.

    Notes: Abstract  The genetics of fertility restoration (Rf) of kosena radish CMS has been characterized. The kosena CMS-Rf system is genetically the same as that of the ogura CMS-Rf system. Two dominant genes that act complementary to the restoration of fertility control fertility restoration in kosena CMS. One allele (Rf1) is associated with accumulation of the CMS-associated protein, ORF125. The interaction of Rf1 and another allele (Rf2) was essential for the restoration of fertility in radish, whereas Rf1 alone was sufficient for the complete restoration of fertility in the B. napus kosena CMS cybrid.

    Notes: Abstract The activity of α-amylase (EC 3.2.1.1) in mung bean (Vigna radiata (L.) Wilczek) cotyledons increased markedly in response to wounding. The changes in enzyme activity were in parallel with those in enzyme content. The level of α-amylase mRNA also notably increased in wounded cotyledons and attained its maximum level during the period between 1 and 2 d after wounding. The level of mRNA for phenylalanine ammonia-lyase, which is one of the well-characterized stress-inducible proteins, also increased after wounding, but the increase in mRNA level was faster than that of α-amylase mRNA. On the other hand, the content of mRNA for actin, a ‘housekeeping’ protein, was almost the same in wounded and unwounded cotyledons. The increase in α-amylase mRNA level in wounded cotyledons was severely inhibited by α-amanitin and cordycepin. α-Amylase expression in the first leaves of mung-bean seedlings was also induced by wounding.

    Notes: Abstract A CMS-associated gene, orf125, present in the Japanese radish cultivar Kosena, has a sequence homologous to that of the ogura CMS-associated gene, orf138, except for two amino acid substitutions and a 39 bp deletion in the orf138 coding region. In Kosena radish, orf125 is linked with orfB, whereas the orf125 locus differs in a Brassica napus CMS cybrid derived from protoplast fusion between Kosena radish and B. napus. A novel mtDNA sequence is present in the 3′-flanking region of orf125 in the B. napus kosena CMS cybrid. The orf125 is expressed both in the radish and the B. napus kosena CMS cybrid. Its accumulation is strongly associated with the CMS phenotype in B. napus. Fertility restoration was accompanied by a decrease in the amount of ORF125 in B. napus.
