    Notes: We present coplanar resonator measurements of the nonlinear microwave surface impedance of laser ablated and electron-beam coevaporated YBa2Cu3O7−δ thin films at 8 GHz in zero and applied magnetic fields, including the effects of irradiation with heavy ions. Correlations between the changes in surface resistance and reactance as a function of microwave current suggest that there are different contributions to the nonlinear behavior at low and high microwave currents in both zero and applied fields. In zero field, we suggest that microwave-induced flux lines are responsible for the observed nonlinear behavior, resulting in thermal runaway at large, sample dependent, microwave currents. The observed nonlinearity completely masks any possible contribution from the intrinsic pair breaking expected for a d-wave superconductor. In a magnetic field at relatively low currents, the nonlinearity can be described by the nonlinear dynamics of flux lines pinned by defects with a continuous range of pinning strengths. At higher currents, microwave field nucleation of flux lines is again considered important. On irradiation with 3.6 MeV/nucleon Xe-129 ions, the microwave losses and nonlinearity are significantly decreased in both zero and applied fields, which could have important implications for microwave device applications. © 1999 American Institute of Physics.

    Notes: Abstract Aims/hypothesis. Although retinoid X receptor (RXR) and peroxisome proliferator activated receptor-γ (PPARγ) agonists have antidiabetic effects in hyperinsulinaemic animals, little information exists on their effects after pancreatic beta-cell failure. Thus, we examined if RXR and PPARγ agonists alter distinct metabolic pathways in animals suffering from impaired insulin secretion. Methods. Adverse side effects and antidiabetic responses were measured in db/db mice treated from 14–16 weeks of age with the RXR agonist, LG100268, and/or the PPARγ agonists, BRL49 653 or GW1929. Results. In animals treated with LG100 268 or BRL49653, serum glucose, glycohaemoglobin and the cardiovascular risk factor, fibrinogen, decreased to the same extent. Both of these agonists were equally effective at increasing insulin accumulation in beta cells, although neither agent had an effect on serum insulin concentrations. In contrast, the RXR agonist was less effective than the PPARγ agonists at lowering serum triglycerides and non-esterified fatty acids and increasing interscapular brown fat and body weight. Further, LG100 268 increased serum alkaline phosphatase and liver mass, hepatic fat accumulation, lauric acid hydroxylase activity, catalase-immunostaining and peroxisomal number more than the PPARγ agonists. Moreover, co-treatment with the RXR and PPARγ agonists reduced glucose, triglycerides, non-esterified fatty acids and cholesterol more than either agent alone. Conclusion/interpretation. These data suggest 1) RXR and PPARγ agonists decrease islet degeneration, cardiovascular risk and cachexia during later stages of diabetes, 2) RXR agonists are less effective than PPARγ agonists at decreasing serum lipids and causing weight gain and 3) RXR agonists have a more pronounced effect on liver metabolism (e. g. peroxisome accumulation and hepatomegaly) than PPARγ agonists.     Notes: Abstract Teichuronic acid was the major anionic polymer of Bacillus licheniformis NCIB 6346 durign phosphate-limited (P-limited) growth in the chemostat. This polymer was also present in significant quantities when B. licheniformis was grown under carbon-limited (C-limited) or magnesium-limited (Mg-limited) conditions where teichoic acid predominated in the cell wall. However, the cell wall composition was not of significance in protein export and the parameters for the excretion process were found to be environmental. In particular, two types of extracellular proteins were identified: the first type of enzyme, penicillinase, was only weakly catabolite repressed; was maximally synthesized and secreted during P-limited growth; was unaffected by growth in high Na+ media but its production was inhibited by gramicidin. The second type of enzyme, α-amylase, was strongly catabolite repressed and its export was markedly inhibited during P-limited growth or in the presence of Na+ or gramicidin. It is noteworthy that the penicillinase carries a glyceride-cysteine modification at its N-terminus whilst the α-amylase does not.

    Notes: Abstract Bacillus licheniformis NCIB 6346 was grown under phosphate or magnesium limitation in continuous culture and forward and reverse transitions carried out to examine their effects upon extracellular enzyme production. Analysis of the observed kinetics allowed a distinction to be made between the effects of phenotypic variation and of genetic selection upon exoenzyme production. Furthermore, increases in the phosphate concentration (0.3–30 mM) were directly related to increased phenotypic expression of alpha-amylase. By contrast, penicillinase production was reduced under these conditions but analysis revealed that its phenotypic expression was only indirectly influenced by the concentrations of either magnesium or phosphate present during the transitions.

    Notes: Abstract  In this study we report measurements of the intracellular sodium concentration ([Na+]i) in cardiac myocytes isolated from sub-endocardial and sub-epicardial regions of the rabbit left ventricle. These measurements show that [Na+]i is significantly higher in sub-epicardial than in sub-endocardial myocytes both at rest and during steady-state stimulation at 0.5 Hz. During a 10-min rest the rate of fall of [Na+]i was identical in cells from the two regions but during post-rest recovery [Na+]i rose significantly faster in sub-endocardial cells. The effect that this difference in sodium regulation may have on the rate of recovery of the calcium transient post-rest is discussed. The steady-state differences in [Na+]i do not appear to have the effect on contraction we would expect i.e. steady-state contraction is larger in sub-endocardial cells where [Na+]i is lower. Changes in [Na+]i brought about by altering the frequency of stimulation are associated with the expected changes of contraction. Action potentials were found to be significantly longer in sub-endocardial cells and following rest action potential duration was shortened although regional differences were maintained. No regional differences in this effect of rest or in recovery of the action potential post-rest were found.
