    Notes: We present coplanar resonator measurements of the nonlinear microwave surface impedance of laser ablated and electron-beam coevaporated YBa2Cu3O7−δ thin films at 8 GHz in zero and applied magnetic fields, including the effects of irradiation with heavy ions. Correlations between the changes in surface resistance and reactance as a function of microwave current suggest that there are different contributions to the nonlinear behavior at low and high microwave currents in both zero and applied fields. In zero field, we suggest that microwave-induced flux lines are responsible for the observed nonlinear behavior, resulting in thermal runaway at large, sample dependent, microwave currents. The observed nonlinearity completely masks any possible contribution from the intrinsic pair breaking expected for a d-wave superconductor. In a magnetic field at relatively low currents, the nonlinearity can be described by the nonlinear dynamics of flux lines pinned by defects with a continuous range of pinning strengths. At higher currents, microwave field nucleation of flux lines is again considered important. On irradiation with 3.6 MeV/nucleon Xe-129 ions, the microwave losses and nonlinearity are significantly decreased in both zero and applied fields, which could have important implications for microwave device applications. © 1999 American Institute of Physics.

    Notes: Abstract Teichuronic acid was the major anionic polymer of Bacillus licheniformis NCIB 6346 durign phosphate-limited (P-limited) growth in the chemostat. This polymer was also present in significant quantities when B. licheniformis was grown under carbon-limited (C-limited) or magnesium-limited (Mg-limited) conditions where teichoic acid predominated in the cell wall. However, the cell wall composition was not of significance in protein export and the parameters for the excretion process were found to be environmental. In particular, two types of extracellular proteins were identified: the first type of enzyme, penicillinase, was only weakly catabolite repressed; was maximally synthesized and secreted during P-limited growth; was unaffected by growth in high Na+ media but its production was inhibited by gramicidin. The second type of enzyme, α-amylase, was strongly catabolite repressed and its export was markedly inhibited during P-limited growth or in the presence of Na+ or gramicidin. It is noteworthy that the penicillinase carries a glyceride-cysteine modification at its N-terminus whilst the α-amylase does not.

    Notes: Abstract Bacillus licheniformis NCIB 6346 was grown under phosphate or magnesium limitation in continuous culture and forward and reverse transitions carried out to examine their effects upon extracellular enzyme production. Analysis of the observed kinetics allowed a distinction to be made between the effects of phenotypic variation and of genetic selection upon exoenzyme production. Furthermore, increases in the phosphate concentration (0.3–30 mM) were directly related to increased phenotypic expression of alpha-amylase. By contrast, penicillinase production was reduced under these conditions but analysis revealed that its phenotypic expression was only indirectly influenced by the concentrations of either magnesium or phosphate present during the transitions.

    Notes: Summary The optimum production of cloned carboxypeptidase G2 from plasmid pNM21 byEscherichia coli was found to be strongly strain- and temperature-dependent. The superior host was strain RV308 and the preferred growth temperature 28°C. Copy number, which decreased during exponential growth of all strains examined, did not relate in these studies to the level of enzyme production: the strain with the highest enzyme yield also having the lowest overall copy number.
