    Description / Table of Contents: Résumé La technique d'immunofluorescence démontre l'existence de cellules sécrétrices de glucagon dans l'îlot pancréatique endocrine du Poisson téléostéen Xiphophorus helleri H. La réaction est faite sur du matériel fixé et inclus pour la microscopie électronique, avec un sérum de lapin anti-glucagon de porc. Le tryptophane est mis en évidence sur les coupes adjacentes par la réaction au DMAB-nitrite; un certain nombre de cellules fluorescentes sont négatives pour cette réaction. La différence entre les résultats donnés par les deux techniques est discutée.
    Notes: Summary By immunofluorescence technique performed on material embedded in epoxyresin, glucagon containing islet cells were demonstrated in the teleost, Xiphophorus helleri H. Rabbit anti-pig glucagon serum was employed. Correlative tryptophan DMAB-nitrite staining shows that only some of the immunofluorescent glucagon cells are positive; this discrepancy is discussed.

    Notes: Summary Crystalline inclusions in parathyroid gland cell nuclei of Rana temporaria were studied by electron microscopy using a specimen tilting stage. Images were analysed by optical diffraction. Results were compared with X-ray and electron microscopic data of trigonal bovine liver catalase to which a striking resemblance of the inclusions was found.

    Notes: Summary Yolk-platelet crystals in Amia calva L., Lepisosteus osseus (L.) and Polypterus bichir have orthorhombic features with unit-cell dimensions a= 8.3...8.8 nm, b= 16.4...16.9 nm and c= 18.6...19.8 nm as determined in electron-diffraction patterns of fixed, epoxy-resin embedded and thinsectioned material. Electron-diffraction patterns, crystal projections and the above unit-cell data make them extremely similar to the orthorhombic yolkplatelet crystals known for amphibians and teleosts. This observation fills a gap in yolk-platelet research and supports the view that the general architecture of yolk platelets has been conserved for nearly 400 million years. It follows that the peculiar platelet architecture itself has physiological significance.

    Notes: Summary Yolk-platelet crystals in the oocytes of the teleost Pelvicachromis pulcher (Cichlidae) were shown, using electron diffraction and tilting of thinsectioned specimens, to possess an orthorhombic lattice with unit-cell sides a=8.3 nm, b=16.6 nm and c=18.0 nm. They thus closely resemble the crystals known for a newt (Triturus sp.) and a frog (Xenopus laevis).

    Description / Table of Contents: Résumé Cinq catégories cellulaires sont décrites dans l'îlot endocrine du poisson téléostéen Xiphophorus helleri. Chacune est définie par un ensemble de caractères histochimiques et ultrastructuraux, ce qui peut être fait avec certitude grâce à l'étude comparée de coupes ultra-fines et sémi-fines successives. Les techniques d'immunofluorescence ont été appliquées à du matériel préparé pour la microscopie électronique, à l'aide de sérums anti-glucagon et anti-insuline de porc. Un des résultats les plus intéressants est la démonstration de l'existence de 2 catégories de cellules A2: les cellules Adams positives, qui ont des grains de sécrétion amorphes et de section circulaire (≪ cellules à grains ronds ≫) et les cellules Adams négatives, dont de nombreux grains sont des cristaux (≪cellules à grains cristallins≫). Les cellules B et les cellules A1 constituent deux autres types cellulaires. Les ≪cellules claires≫, qui ne réagissent à aucune des techniques employées, représentent un type cellulaire distinct des précédents. L'influence de la fixation se révèle très importante, aussi bien sur les affinités tinctoriales des cellules que sur leurs caractères ultrastructuraux. The authors are grateful to Mrs. L.G.     Notes: Summary By means of correlative light and electron microscopy, five pancreatic islet cell categories are described in the teleost fish, Xiphophorus helleri, each of which has specific light microscopic appearance and fine structure. Different histochemical techniques have been used, including immunofluorescence with antiporcine insulin and glucagon sera. In addition to B- and A1-cells, two categories of A2-cells have been observed, both reacting with antiporcine glucagon serum: A2-cells with round granules gave a positive reaction for tryptophan; A2-cells with crystalline granules gave a negative reaction with the same staining technique on the same section. The “clear cells”, the last category, were not specifically stained by any of the staining methods carried out in this investigation. The influence of fixation on staining affinities and on ultrastructure was shown to be considerable.
