    Notizen: In a rat heart transplantation model, permanent graft survival was achieved by anti-thymocyte globulin (ATG) treatment of Wistar Kyoto (WKy) recipients before grafting with PVG/c hearts. Nine hearts with palpable and electrocardiographic (ECG) function 8–20 months after transplantation were studied with histological and immunohistochemical methods. The dominating findings were fibrosis, intimal thickening, thrombosis of vessels, cellular infiltrates, and a high number of cells expressing class II antigens. These findings suggest that reactions compatible with a chronic rejection can be found in long-term surviving rat heart allografts. This rat model may thus be used in further studies of a chronic rejection process.

    Notizen: This communication describes an immunohistochemical analysis of rejected human renal allografts. T-lymphocyte subsets were identified in frozen tissue sections, by Leu 1 (anti- ‘pan’ T lymphocytes). Leu 2a (anti- ‘cytotoxic/suppressor’ T cells), and Leu 3a (anti- ‘helper/inducer’ T cells) monoclonal antibodies. In addition, HLA-DR-positive cells were identified by simultaneous labelling with heterologous anti-HLA-DR antibodies. T cells dominated the cellular infiltrates in acute cellular rejection. Leu-3a-positive cells were more numerous than Leu-2a-positive cells. The Leu-3a-positive cells usually appeared in clusters, whereas the Leu-2a-positive cells appeared scattered in the tissue. HLA-DR-positive non-T cells were found within clusters of T ‘helper/inducer’ cells. The cell pattern shares many features with the findings in detayed-type hypersensitivity reactions.

    Notizen: We have investigated the cytotoxic response to allogeneic I-E and I-A antigens in bulk culture and limiting dilution experiments. A high degree of specific killing could be generated from unprimed T cells of I-E-negative strains upon stimulation with cells expressing I-E. In such conditions, cytotoxic T lymphocytes (CTL) were generated in the absence of a specific proliferative response. The frequency determinations by means of limiting dilution experiments showed that I-E-specific CTL precursors were much less frequent than the K. and D specific precursors. The results suggest the existence of a population of I-E-specific CTL that failed to grow under conditions that allowed the growth of class I-specific CTL.

    Notizen: The effects of eliminating cycling precursors by in vivo administration of high doses of hydroxyurea (HU) in populations of mouse spleen T lymphocytes were studied by membrane immunofluorescence identifying mature Thy 1+, Lyt 2− and Lyt 2+ cell populations and mitogen-induced T-cell growth factor (TCGF) production or reactivity to TCGF. HU treatment results in a sharp decrease in the total number of T cells in mouse spleen, indicating that about 50% of peripheral splenic T lymphocytes have a short survival time in vivo. This depletion is more marked for Lyt 2+ cells than for Lyt 2− cells. The decay of TCGF-reactive spleen cells after drug adminstration is similar to that of the overall spleen T-cell population. Conversely, production of TCGF induced by concanavalin A is predominantly affected by HU treatment, indicating a shorter life span of the cell(s) involved in TCGF production.

    Notizen: The frequency of leucoagglutinin (LA)-inducible, TCGF-reactive T cells has bcen determined using a sensitive limiting-dilution assay. The f'rcqucncy of cells proliferating in TCGF-CM was 1 in 3. and the Frequency of lectin-dependent cytotoxic T lymphocyte precursors (CTL-p) was 1 in 3–4. By analysing individual clonal cultures for proliferation and cytotoxic potential, a selective induction nf CTL-p was detected, as all prolifcrating clones were found to mediate lectin-dependent cytotoxicity.

    Notizen: We have analysed the effects of ultraviolet (UV) irradiation on the expression, by normal splenic cell populations, of cell-mediated lympholysis determinants encoded by the I-A and I-E genes of the H-2 complex. Cytotoxic T lymphocytes specific for class II antigens could be generated from unprimed responder mice when stimulated with UV-treated cells from I-A-and/or I-E-congenic mice in the presence of interleukin 2-containing medium. A cytotoxic T-cell line specific for I-Ek antigens was obtained and maintained upon stimulation with UV-irradiated spleen cells. Secondary la-specific responses were obtained with UV-irradiated stimulators in the absence of added soluble factors. The results show that the failure of UV-irradiated cells to stimulate primary cytotoxic T-cell responses is not due to a selective destruction of class II antigens but rather to the inactivation of a still undefined function of la-positive cells, other than interleukin-1 production, involved in the establishment of high-affinity interactions between the responding T helper cells and the stimulator cells.

    Notizen: In a rat kidney transplantation model, DA kidneys were transplanted into Lewis rats. Syngeneic Lewis transplantations were studied as controls. Histologic evaluation was made, and immunohistochemical staining with a single-staining peroxidase–antiperoxidase technique on frozen sections was performed after 2, 4, 6, and 8 days. Antibodies for Ia antigen (Ox 6), ‘suppressor/cytotoxic’ cells (Ox 8), pan-T cells (W 3/13), and ‘helper/inducer’ cells (W 3/25) were used. Allogeneic grafts were almost completely rejected in 8 days. Syngeneic grafts also showed lymphocyte infiltrates, somewhat later than allogeneic ones, but were not rejected. In these infiltrates W 3/25-positive cells dominated, being even more numerous than W 3/13-positive cells. Relatively fewer Ox 8-positive cells were seen in the infiltrates of syngeneic than in allogeneic transplants. Infiltrates occurred later in the renal medulla than in the cortex. Perivascular infiltrates with cells of all investigated phenotypes were seen earlier in allogeneic grafts than in syngeneic ones. All tubular cells within one renal tubule appeared either Ox 6-positive or -negative. With time, all tubules in the allogeneic transplants became Ox 6-positive. Some increase of Ox 6-positive tubules was also seen in syngeneic transplants.

    Notizen: Collagen type II-spedfic long-term cultured T helper cells, derived from the DBA/1 mouse, have been established and characterized. Clones from these T-cell lines could he shown to recognize either species-specific or species-nonspecific determinants on the collagen type II molecule, including determinants on autologous mouse collagen. Induction of arthritis via transfer to both irradiated and normal syngeneic recipient mice was obtained with both collagen type II-specific T-cell lines and an autoreactive and collagen type II-specific T-cell clone. Fewer cells were needed to evoke arthritis in normal than in irradiated recipients. Cells from lines and the clone used for transfer were by immunocytochemistry shown to have T helper phenotype.

    Notizen: The involvement of different sets of lymphoid cells in the development of collagen II-induced arthritis in rats was studied by means of immunohistochemical analyses on frozen sections of tissue from joint biopsy specimens taken at different phases of arthritis development. Particular attention was paid to cells involved in early pannus formation. Accumulation of anti-Ia-reactive cells close to the cartilage surface was seen early in the development of pannus, and the anti-Ia reactive cells could in later phases be seen infiltrating cartilage and crowding bone surfaces at sites of marginal erosion. With the help of monoclonal anti-T-cell subset antibodies and rabbit anti-rat immunoglobulin antiserum, it was demonstrated that synovial infiltration of T lymphocytes, particularly W3/25-reactive T “helper” cells, occurs very early in the development of arthritis, whereas a moderate increase of Ox 8-positive ‘suppressor/cytotoxic’ T cells and a small number of B cells and plasma cells are seen later in the course of the disease. Levels of Ia-expressing cells and of T cells belonging lo different subsets were recorded in peripheral blood by means of immunocytochemical analyses on cell smears; no significant deviations from normal levels were seen during the development of arthritis.

    Notizen: F1, hybrid mice injected with parental T lymphocytes undergo a graft-versus-host (GvH) reaction, one of the consequences being a highly depressed cytotoxic T-lymphocyte (CTL) potential against alloantigens and modified self antigens. In the present study we demonstrate that concanavalin A (Con A) can he used for analysing such GvH-mediated immune suppression. Thus, both alloantigen- and Con A-induced responses were reduced by approximately 80% in F1, suppressed (F1s) animals as compared to F1 control (F1c) mice, Although interleukin 2 (IL-2) production was found to be reduced by approximately 50% it did not account for the reduced CTL response in F1s mice. The addition of IL-2 to Con A-stimulated F1s spleen cell cultures did not reconstitute the response. The results suggest that the suppressive mechanism operates by preventing a large fraction of Lyt-2+ CTL precursors from acquiring IL-2 reactivity. However, a small fraction of CTL precursors, escaped the suppression and differentiated into effector CTL.
