    Notes: Objective: To provide a synthesis of information learned by the veterinary community during the care and treatment of search and rescue dogs in a large-scale urban disaster.Summary: Disaster medicine requires several stages. The most important stage is the planning stage. To function effectively and safely in a disaster situation, a disaster response plan and prior training in disaster response are essential. The execution of a disaster plan requires coordination and communication with multiple agencies. Treatment of search and rescue dogs involves front line field care for life-threatening emergencies, a first aid station or MASH unit in close proximity to the disaster and/or base of operations for stabilization and general medical care, and local full service veterinary hospitals for specialty and extended care.New or Unique information provided: This report is a summary of experiences gained in preparing for and implementing veterinary care in a major disaster. Medical conditions that affected search and rescue dogs and were treated by veterinary disaster responders at the World Trade Center are presented.

    Notes: Fusarium culmorum is one of the most important Fusarium species causing head blight infections in wheat, rye, and triticale. It is known as a potent mycotoxin producer with deoxynivalenol (DON), 3-acetyl deoxynivalenol (3-ADON), and nivalenol (NIV) being the most prevalent toxins. In this study, the effect of winter cereal species, host genotype, and environment on DON accumulation and Fusarium head blight (FHB) was analysed by inoculating 12 rye, eight wheat, and six triticale genotypes of different resistance levels with a DON-producing isolate at three locations in 2 years (six environments). Seven resistance traits were assessed, including head blight rating and relative plot yield. In addition, ergosterol, DON and 3-ADON contents in the grain were determined. A growth-chamber experiment with an artificially synchronized flowering date was also conducted with a subset of two rye, wheat and triticale genotypes. Although rye genotypes were, on average, affected by Fusarium infections much the same as wheat genotypes, wheat accumulated twice as much DON as rye. Triticale was least affected and the grain contained slightly more DON than rye. In the growth-chamber experiment, wheat and rye again showed similar head blight ratings, but rye had a somewhat lower relative head weight and a DON content nine times lower than wheat (3.9 vs. 35.3 mg/kg). Triticale was least susceptible with a five times lower DON content than wheat. Significant (P = 0.01) genotypic variation for DON accumulation existed in wheat and rye. The differences between and within cereal species in the field experiments were highly influenced by environment for resistance traits and mycotoxin contents. Nevertheless, mean mycotoxin content of the grain could not be associated with general weather conditions in the individual environments. Strong genotype-environment interactions were found for all cereal species. This was mainly due to three wheat varieties and one rye genotype being environmentally extremely unstable. The more resistant entries, however, showed a higher environmental stability of FHB resistance and tolerance to DON accumulation. Correlations between resistance traits and DON content were high in wheat (P = 0.01), with the most resistant varieties also accumulating less DON, but with variability in rye. In conclusion, the medium to large genotypic variation in wheat and rye offers good possibilities for reducing DON content in the grains by resistance selection. Large confounding effects caused by the environment will require multiple locations and/or years to evaluate FHB resistance and mycotoxin accumulation.

    Notes: The performance of the Rapid Infusion System was evaluated in the laboratory. Using a conventional mixture of two units of packed red cells, two units of fresh frozen plasma and 500 ml crystalloid, a single line and a driving pressure of 300 mmHg, the highest flow in our study was 970 ml.min−1 (2.8 mm catheter, no stopcock). With a 1.6 mm venous cannula the measured flow was 640 ml.min−1. Additional diluting of the standard ‘blood cocktail’ did not add much to the performance of the system. When primed with tap water 21°C (12°C respectively), the fluid at the outlet of the system reached a maximum temperature of 37.8°C (37.4°C) after 6 min at a flow of 400 ml.min−1. At flows higher than 1150 ml.min−1 (priming with 12°C tap water: 800 ml.min−1), the system slowed down to flows of 700 to 1000 ml.min−1 in order to maintain an adequate temperature. We conclude, that the Rapid Infusion System is a valuable tool for situations where a rapid but controlled replacement with warmed blood at rates up to at least 1100 ml.min−1 is needed. The use of large bore intravenous catheters and avoiding additional resistors such as standard 3-way stopcocks is highly recommended.

    Notes: Melanocortins are thought to be involved in neuronal development and regeneration. Proopiomelanocortin (POMC), the precursor of α-melanocyte stimulating hormone (α-MSH), γ-MSH, ACTH, and β-endorphin, becomes detectable in rat hypothalamic neurons from gestational day (E) 12.5. We recently described stage- and region-specific ontogenetic patterns of binding sites for the α-MSH analogue [125I]-Nle4, d-Phe7-α-MSH ([125I]-NDP), with the first localizations in epithalamus and sympathetic chain at E13. [125I]-NDP binds to all known melanocortin receptors, including MC3-R and MC4-R, the predominant melanocortin receptors in nervous system. To identify the receptor type expressed during ontogeny, the developmental pattern of MC3-R and MC4-R mRNA was investigated by in situ hybridization in fetuses and offspring of time-pregnant Long Evans rats between E14 and postnatal day (P) 27. MC4-R mRNA was found to be the predominant species during the entire fetal period. It was localized in all fetal areas exhibiting distinct [125I]-NDP binding, starting with sympathetic ganglia and epithalamus (E14), and including sensory trigeminal nuclei (E16), dorsal motor nucleus of vagus (E16) and cranial nerve ganglia, inferior olive (E18) and cerebellum (E18), striatal regions (E16), and entorhinal cortex (E22). In contrast, MC3-R mRNA was detectable only in the postnatal period, with a fast increase in expression in the ventromedial and arcuate nuclei. The early presence of MC4-R mRNA in central and peripheral nervous system and transient regional peaks of mRNA expression, often concomitant with periods of neural network formation, suggest a role of this receptor type in early ontogeny. The MC3 receptor may be involved in analogous processes during postnatal development.

    Notes: We have used in situ hybridization to investigate estradiol regulation of estrogen receptor (ER) mRNA in regions of the mediobasal hypothalamus which contain ER and are related to specific neuroendocrine functions. Ovariectomized rats were treated with oil or 10 μg estradiol benzoate for 2, 4, 18 or 24 h. Brains were sectioned and hybridized with a [3 H]single-stranded DNA probe prepared from the pORF cDNA of the human ER gene and exposed to autoradiographic emulsion for 4 months. Specificity of labeling was determined by counting the number of grains over cells in hypothalamic regions known to bind estradiol, compared to cells in the thalamus and cortex, and by comparing with sections pretreated with ribonuclease or hybridized with a [3 H]single-stranded message-sense (control) probe. Labeling for ER mRNA was distributed differently than glucocorticoid and thyroid hormone receptor mRNAs, and was regulated by estrogen differently than progestin receptor mRNA. These differences indicated specific hybridization for ER mRNA. ER-expressing cells constituted 11.5% of the cells in the dorsomedial nucleus, 30% of the cells in the arcuate nucleus and 43% in the ventromedial nucleus, in close accordance with previous studies of ER autoradiography and binding. Quantitative analysis showed that the highest level of ER mRNA was present in the ovariectomized controls. ER mRNA levels fell 42% (ventromedial), 64% (arcuate), or remained unchanged (dorsomedial) 18 h following estradiol benzoate treatment. The pattern of decrease was similar for cells in the ventromedial nucleus and arcuate nucleus. These data show that estrogen regulation of ER mRNA in brain parallels that reported for MCF-7 cells and rat uterus. These results also demonstrate that in situ hybridization can be used to detect and measure the relative level of a low abundance mRNA in a heterogeneous tissue in which only 12% to 40% of the cells in limited regions express the message.

    Notes: The Diazepam Binding Inhibitor/Acyl-CoA Binding Protein (DBI/ACBP) has been implicated in different functions, as acyl-CoA transporter and as an endogenous ligand at the GABAA receptor and the peripheral benzodiazepine receptor (PBR). The latter is thought to be involved in control of steroidogenesis. We studied the ontogeny of DBI/ACBP and PBR mRNA expression in embryos and offspring of time-pregnant Long Evans rats by in-situ hybridization with 33P-endlabelled oligonucleotides. Both mRNAs were present in embryo and placenta at gestational day (G)11, the earliest stage studied. DBI/ACBP mRNA was strongly expressed from embryonic through mid-foetal stages in central nervous system (maximum in neuroepithelium), cranial and sympathetic ganglia, anterior pituitary, adrenal cortex, thyroid, thymus, liver and (late foetal) brown adipose tissue, moderately in testis, heart, lung and kidney. In brain, a late foetal decrease of DBI/ACBP mRNA was followed by an increase at postnatal day 6. Peripheral benzodiazepine receptor mRNA expression started very low and increased to moderate levels in adrenal cortex and medulla, testis, thyroid, brown adipose tissue, liver, heart, lung, salivary gland at mid- to late-foetal stages. Data suggest a significant role of DBI/ACBP at early developmental stages. Both proteins may be involved in the control of foetal steroidogenesis. However, differences in developmental patterns indicate that additional functions may be equally important during ontogeny, such as the involvement in lipid metabolism in the case of DBI/ACBP.
