    Notes: Abstract The immunohistochemical occurrence of sarafotoxin (SRTX), a snake venom peptide under strong evolutionary control, was investigated in the pulmonary diffuse neuroendocrine system (PDNES) of newborn cats and rats. By applying the avidin-biotin-peroxidase complex method on serial lung sections, we have demonstrated its distribution and colocalization with different endothelin (ET) isoforms. A light microscopic study revealed apparent immunostaining for SRTX in neuronal components and smooth muscle tissue and in neuroepithelial bodies (NEB), while isolated neuroendocrine cells (NEC) remain unlabelled. Comparison of the SRTX reactivity pattern with that of different ET peptides on adjacent lung sections showed colocalization of SRTX-b with ET-3 in NEB, intrapulmonary ganglion cells and nerve fibres, on the one hand, and with ET-1 in airway and vascular smooth muscle cells, on the other. These findings, in addition to the remarkable functional and structural similarities between SRTX and ET peptides, suggest a common evolutionary origin and biological significance of sarafotoxin and endothelins. Moreover, this is the first time that a toxic peptide has been demonstrated in the PDNES.

    Notes: Summary Two unrelated patients with severe arthrogryposis multiplex congenita (AMC) who died perinatally, are presented. In both, postmortem examination revealed an intact nervous system and striking dystrophic muscle changes, consistent with congenital muscular dystrophy (CMD). Few similar cases have been reported before, but since the condition is not well known, it seems probable that in the past many have been labeled as mere multiple malformations. The possibility of an underlying muscular disorder, either primary myopathic or neurogenic should be considered in any patient with early lethal AMC. Our findings confirm that the fetal akinesia-arthrogryposis sequence is a nonspecific clinical syndrome resulting from various causes of muscular inactivity in utero. The main objective of this report is to provide reasonable guidelines on how to approach the problem of classification. We favor a pathogenetic approach, depending upon careful sampling of the central nervous system and skeletal muscles at autopsy.

    Notes: Summary In an attempt to further our knowledge about the structure and function of the recently identified intrapulmonary Neuro-epithelial Bodies (NEB's) (Lauweryns et al., 1972a, 1972b), lungs of 84 neonatal rabbits and 6 neonatal mice were studied along four different lines of investigation. Several routine and silver staining methods, Falck's fluorescent amine technic and histochemical and electron microscopical techniques were performed. 1. In order to test the probable chemoreceptor function of the NEB's, animals were exposed to hypoxia. Under such circumstances, the corpuscular cells of the NEB's secrete their dense-cored, serotonin-containing vesicles at their basal vascular pole. 2. After reserpine pretreatment, the NEB's of otherwise normal animals reveal a distinct amine depletion, the corpuscular cells exhibiting a decreased yellow fluorescence and ultrastructurally a clearing up of their dense-cored vesicles. 3. Studied on serial sections with the electron microscope, various types of morphologically afferent-like and efferent-like nerve endings, making contact as well with the corpuscular cells as amongst themselves have been detailed. 4. Cytochemically the corpuscular cells react positively with alpha-glycerophosphate dehydrogenase, acetylcholinesterase and Solcia's lead hematoxylin stain for endocrine cells producing polypeptides and amines. It is proposed that the NEB's provide an intrapulmonary, hypoxia-sensitive neuro(chemo-) receptor system in addition to the well established central and peripheral (e.g. carotid body) chemoreceptors. They contain and secrete serotonin and probably also related amines or peptides, which could influence the pulmonary vasoconstrictor response. According to classic morphologic criteria, they possess a dual innervation, both afferent and efferent. Various other possible functions of the NEB's in normal and diseased lungs are briefly proposed.

    Notes: Summary An electron microscopic investigation has revealed that the pulmonary lymphatic valves of adult rabbits are not simple duplicatures of the lymphatic vessel wall. They consist of an uninterrupted central connective tissue core, covered on both sides with a single layer of flattened endothelial cells. Near their insertion in the lymphatic vessel wall, the connective tissue core reveals a distinct thickening being composed mainly of collagen bundles. In the other parts it contains mainly elastic fibers and fine filaments, enclosing also some rather peculiar connective tissue cells. Nervous and muscular elements were not observed. The endothelium is continuous and exhibits no open junctions. The valvular basement membrane is better developed than in lymphatic capillaries. The endothelial cells contain numerous cytoplasmic filaments which might be endowed with contractile properties. The nuclei of the endothelial and the connective tissue cells are irregularly spaced and frequently clustered near the free edge of the valve. These ultrastructural features suggest that the function of the lymphatic valves is mainly passive. They are firmly inserted in the lymphatic vessel wall by collagen fibers and their moving parts are slender and elastic. Their endothelium appears relatively impermeable and is firmly attached to the subjacent connective tissue.

    Notes: Summary Neonatal rabbit neuro-epithelial bodies (NEB) were investigated under various experimental conditions with light microscopy, microspectrography, morphometry and electron microscopy. (1) Hypoxia causes a decreased amine fluorescence intensity and an increased secretory exocytosis of dense core vesicles (DCV). Otherwise the NEB appear structurally normal. (2) Hypercapnia also produces a decreased fluorescence and an increased exocytosis; ultrastructurally, however, the dense core of DCV fragmentizes. (3) Hyperoxia does not appear to affect significantly either fluorescence or exocytosis. (4) The uptake of biogenic amines such as 5-HTP and L-DOPA was demonstrated by fluorometry and electron microscopy. (5) Reserpine, on the other hand, provokes an amine depletion with a decrease of the NEB fluorescence and an ultrastructural palor of the DCV. (6) Intratracheally administered nicotine is accompanied by a decreased fluorescence and a distinct exocytosis of fragmented DCV. The reaction of NEB to hypoxia and hypercapnia suggests that these corpuscles could be intrapulmonary chemoreceptors (in addition to the classically known central and peripheral chemoreceptors), inducing a reflex reaction through the liberation of DCV at the corpuscular sensible nerve endings and via the CNS. In addition, they may subserve a local intrapulmonary effect by modulating directly the hypoxic and hypercapnic pulmonary vasoconstriction and thus the V/Q ratio.

    Notes: Summary Lungs of fetal, neonatal and adult rabbits and of various other adult mammals were investigated. Several routine and silver staining methods, Falck's fluorescent amine technic and various electron microscopic techniques were performed. As in the human infant lung (Lauwerynset al., 1972) each of these techniques revealed the occurrence throughout the intrapulmonary airways of intramucosal corpuscles or so-called Neuroepithelial Bodies, composed of more or less parallely oriented, non-ciliated cylindrical cells which reach from the basement membrane to the airway lumen and display a prominent cytoplasmic argyrophilia, a less pronounced argentaffinity and an intense yellow fluorescence. Ultrastructurally, they are granulated, containing mainly two types of dense-cored vesicles of which the first type exhibits a positive reaction for serotonin (technic of Jaim-Etcheverryet al., 1968). They contain intracorpuscular nerve endings which form synaptic end formations upon the granulated cells. Though the functions of these serotonin producing Neuroepithelial Bodies remain furthermore unsettled, they seem to be related also to the recently reported AFG (Argyrophil, Fluorescent and Granulated) cells (Lauwerynset al., 1969, 1970a) and might be involved in various mucosal bronchial and bronchiolar neurosecretory processes; most probably they are chemo-, stretch- and/or tactile neuroreceptor organs modulated by the central nervous system.

    Notes: Abstract. Effects of postnatal age and neonatal thymectomy on the numbers and characteristics of pulmonary neuroepithelial bodies (NEB) were investigated in 14-day- compared with 2.5-month-old hamsters. Left lung sections were stained for the marker PGP 9.5 and used for light-microscopic quantification, while the right lungs were processed for an electron-microscopic survey of the NEB ultrastructural features. For the first time, it is clearly demonstrated that, depending on the sampling method, the number of NEB may rise or fall with age; when considering the entire lung volume, the actual number of NEB doubles, whereas when studying a constant surface area, their number apparently decreases. Also, the proportion of alveolar NEB as well as luminal contact increase on normal development. In neonates, in contrast to older animals, apoptosis is clearly present in NEB, and approximately 10% of the NEB are associated with inflammatory cells. In some cases, the dead cells have properties of both apoptosis, disintegration and cytoplasmic degeneration. The presence of intracorpuscular neutrophilic granulocytes correlates with cellular death and innervation of the NEB. Thymectomy causes only minor effects on the pulmonary neuroendocrine system. It is argued that development of the NEB and of their innervation continue during the postnatal period.

    Notes: Summary The ultrastructural localization of serotonin in neuroepithelial bodies (NEB) of neonatal rabbits was examined by use of the recently developed immunogold-staining method. In the granulated epithelial cells of NEB serotoninimmunoreactive material could be demonstrated only in dense-cored vesicles (DCV) of the first type. No immunoreactivity was observed in DCV of the second type. The present findings support the previously made suggestion that the corpuscular NEB cells contain two distinctive DCV types which react differently with the immunogold-staining procedure for identifying serotonin. NEB, known as intrapulmonary sources of serotonin (and peptide substances), may play a prominent role in lung physiology.

    Notes: Abstract. In a previous study, direct epithelial-stromal cell contacts via foot processes perforating the basement membrane were investigated in bronchioles of postnatal hamster lungs. In the present study, analogous postnatal epithelial foot processes have been found on stromal cells exhibiting active elastogenesis as determined by morphological criteria. In addition, elastin is prominently present close to the epithelium, but less so in the deeper layers, in the bronchioles of the 14-day-old hamster. The elastin often has an asymmetrical distribution around the subepithelial fibroblast with preference for secretion toward the epithelium. Thus, elastogenesis can now be linked to epithelial-stromal interactions at the temporal, functional, and morphological levels.
