    Notes: Neutron scattering and specific heat measurements have been carried out on a series of NdBa2Cu3O6+x powder samples with varying oxygen concentrations (x=0.94, 0.78, 0.45, 0.3, 0.13) in order to study the effects of oxygen on the magnetic order of Nd. For our highest oxygenated superconducting NdBa2Cu3O6.94 (Tc(approximately-equal-to)92 K), three-dimensional (3D) long range order develops below TN(approximately-equal-to)0.53 K, with a spin configuration which is antiferromagnetic along all three crystallographic axes. For a small reduction of oxygen to x=0.78, we observe drastic effects on the Nd order; 3D order is inhibited and only short range 2D correlations are found at low temperature. However, the 2D correlations develop at a much higher temperature, near 1.5 K. At (nonsuperconducting) x=0.45, the 3D magnetic order is better developed at low temperature, yet long range order still does not occur. For x=0.3, 3D long range magnetic order of the Nd ions is reestablished, developing below TN(approximately-equal-to)1.5 K, three times that of the fully oxygenated material. All of these ordering temperatures are much higher than what is expected based on dipolar interactions alone, and this coupled with our experimental results indicates that exchange interactions play an essential role, and are strongly influenced by the chain layer oxygen.

    Notes: Low-temperature specific-heat measurements were performed between 0.4 and 4 K on Nd–Ba–Cu–O-samples doped with Ca and additional Nd. In these samples, the specific-heat anomaly due to magnetic ordering of the Nd3+ ions as a function of hole concentration behaves similarly to that of oxygen-deficient NdBa2Cu3Ox. In samples with 0〈Tc〈70 K, a broad anomaly due to short range magnetic interaction was found but no indication for long range magnetic ordering could be observed. In the semiconducting samples, 3D-Ising-like anomalies with TN around 1.6 K were measured. In order to explain the behavior of the magnetic anomaly in the oxygen-deficient undoped samples, one could assume that the magnetic interaction depends on lattice parameters or interatomic distances. Our newest results clearly show, however, that the position and shape of the anomaly is mainly controlled by the density of carriers in the superconducting CuO2 planes, and not by structural properties. Thus, the magnetic rare-earth ions are isolated enough from the CuO2 planes not to influence superconductivity (except for Pr), but are not totally decoupled from the rest of the unit cell and the carrier concentration.

    Notes: We report muon spin rotation/relaxation (μSR) measurements of the heavy fermion superconductor UPt3 in external fields Hext(parallel)cˆ. We find that the muon Knight shift is unchanged in the superconducting state, consistent with odd-parity pairing (such as p wave). The transverse field relaxation is observed to be strongly field dependent, decreasing with increasing field. Below Tc the increase is barely detectable in an applied field of 4 kG(parallel)cˆ. On the basis of the high field measurements, we estimate the low temperature penetration depth to be λ(T→0)≥ (R18)11 000 A(ring).

    Notes: Effect of grain size on magnetoresistance (MR) and electrical and magnetic properties of double perovskite Ba2FeMoO6 (BFMO) have been investigated. BFMO samples with different grain size have been prepared by the conventional solid state reaction followed by sintering in a stream of 5% H2/Ar at 1100 °C with various sintering times. The size of grain increases with increasing of sintering time. Temperature dependent magnetization is the same for samples with different grain size. However, electrical resistivity ρ(T) and MR are affected by the grain size; the ρ(T) and the MR value decrease with an increase in grain size. In spite of the different grain size, the ρ(T) shows a metallic behavior below the ferromagnetic transition temperature. Low magnetic field applied to the sample considerably reduces the resistivity over the whole temperature. The magnitude of MR for the sample sintered for 5 h is as large as 31% and 7% with the magnetic field of 0.7 T at 12 and 300 K, respectively. © 2001 American Institute of Physics.

    Notes: Upper critical magnetic fields Hc2(T) have been measured resistively for the high-temperature (Tc(approximately-greater-than)90 K) superconductors TmBa2Cu3O9−δ and YBa2Cu3O9−δ in applied magnetic fields H up to 9 T. If H at the resistive midpoint is defined as Hc2, then the upper critical field slopes (dHc2/dT)Tc at the superconducting transition temperatures Tc are respectively −2.8 and −1.1 T/K. The Tm-specimen initial slope is apparently the highest yet reported for 90 K superconductors. Extrapolations from this slope, based on standard, three-dimensional, dirty-limit Werthamer, Helfand, Hohenberg, and Maki theory, yield (1) zero-temperature upper critical fields Hc2(T=0)=99–175 T, where the range is determined by the degree of paramagnetic limitation; (2) Hc2(T=77 K)≈36 T, the upper critical field at the boiling point of liquid nitrogen.

    Notes: We have measured electron paramagnetic resonance (EPR), resistivity, and dc susceptibility from 2 to 300 K for the oxide high Tc superconductors (R)Ba2Cu3Oy (R=Y,Pr,Nd,Eu,Gd,Ho,Er,Tm, or Yb). Selected systems were doped with 3-d ions (Cr,Mn,Fe,Ni,Co, or Zn) or 4-f ions (Gd or Er) which presumably substitute for the Cu or R site, respectively. In the systems studied we have observed an EPR line at low temperatures (T〈40 K), which exhibits an increase in intensity and decrease in field for resonance as the temperature is lowered. The ESR linewidth is also temperature dependent and exhibits a minimum at about 15 K. An additional EPR line that can be associated with a Gd3+, Mn2+ or Er3+ ion was observed for those samples where these ions were present as dilute impurities. In some of the samples another EPR signal is observed with properties that depend on sample preparation conditions. The behavior and origin of all lines will be discussed. The variation of Tc with concentration of the added impurities over the range (1%–15%) will also be presented, and compared with previous studies in other superconducting systems.

    Notes: Background:  Blo t 11 is a high molecular weight allergen from Blomia tropicalis with significant immunoglobulin (Ig)E binding frequency. Native and recombinant Blo t 11 are susceptible to degradation and the isolation and expression of the allergen is problematic thus obtaining sufficient amounts of purified Blo t 11 for antibody production is limiting. DNA-based immunization is an attractive alternative strategy that bypasses antigen purification for antibody production.Objectives:  To use a DNA-based immunization protocol for the production and characterization of Blo t 11 monoclonal antibodies (mAbs).Methods:  The 2625 bp cDNA coding for Blo t 11 was cloned into a mammalian expression vector and immunized intramuscularly with electroporation into mice. Monoclonal antibodies to Blo t 11 were generated using a methylcellulose-based hybridoma cloning kit. These mAbs were utilized for native Blo t 11 isolation and the development of sandwich enzyme-linked immunosorbent assay (ELISA).Results:  Six mAbs recognizing the native and recombinant Blo t 11 were generated and characterized. Native Blo t 11 was affinity purified from Bt extract and its identity was confirmed by matrix assisted laser desorption/ionization – time of flight mass spectrometry. The native Blo t 11 showed IgE reactivity with 67% of mite allergic sera. A two-site ELISA developed showed a detection limit of 100 pg/ml of Blo t 11.Conclusion:  A DNA-based immunization protocol was successfully used to generate Blo t 11 mAbs with a spectrum of distinct epitopes located throughout the whole molecule, and they are useful for immunoaffinity purification and immunoassays.

    Notes: Background: The house dust mite Blomia tropicalis (B. tropicalis) was found to be the most prevalent domestic mite in Singapore. However, its pathogenicity in allergic airway diseases remains to be investigated.Methods: Twenty adults with persistent allergic rhinitis (PAR) were studied. Five had a history of asthma, and all were asymptomatic except one who was under treatment with low-dose inhaled corticosteroid. Nasal challenge was carried out by nasal spray with phosphate-buffered saline (PBS) and with increasing concentrations of crude B. tropicalis extracts (0.6, 6.0 and 60 µg/ml) at 15 min intervals. Subjective symptom scores and absolute number of sneezes were recorded together with objective measurements of spirometry (forced expiratory volume in 1 s, FEV1) and acoustic rhinomanometry (volume of the nasal cavity). These were performed at baseline, 5 min after each incremental challenge, and 30 min, 1 h, 3 h, 5 h and 7 h after the last challenge. Meanwhile, concentrations of mediators in nasal secretions (tryptase, leukotriene C4 (LTC4) and eosinophil cationic protein (ECP)) were measured in nasal aspirate samples at similar time intervals. An identical (control) challenge procedure with PBS alone was repeated in seven patients after a washout period of at least 2 weeks.Results: Significant increases in the subjective and objective nasal symptoms, together with a significant increase of tryptase and LTC4 concentrations in nasal secretion, were found 5 min after each challenge with B. tropicalis, but not with PBS. There was no definitive pattern of the late-phase nasal response in either subjective symptoms or objective measurements. Three patients (3/5) with a history of asthma showed a fall in FEV1 readings (33%, 22% and 11% from baseline, respectively) at 7 h post challenge with concomitant mild wheezing in the night.Conclusions: Our study demonstrates direct evidence of allergic nasal response to B. tropicalis in sensitized adults. It shows that nasal provocation may also provoke concomitant asthmatic symptoms during the late-phase reaction, especially in people with a history of asthma.

    Notes: Background: Asthma and atopy are complex genetic traits, influenced by the interaction of multiple genes and environmental factors. Linkage of these traits to chromosome 5q31–33 has been shown in other populations, but has not been well studied in the Chinese. We studied linkage between asthma and atopy with markers on chromosome 5q31–33 in the Singapore Chinese. This region contains many candidate genes, including the cytokine gene cluster. Methods: We recruited 88 Chinese families with at least two affected offspring, totaling 373 subjects, with 125 and 119 sib-pairs for atopy and asthma, respectively. All individuals were genotyped with 19 polymorphic microsatellite markers spanning a distance of 41 cM along chromosome 5q31–33. Affected sib-pair and multipoint linkage analysis was performed. Results: There was evidence for linkage of the asthma and atopy phenotypes with three markers, D5S2110, D5S2011, and D5S412 (P values of 0.001 to 0.00001). Multipoint analysis further substantiated this (nonparametric linkage scores of 1.8–2.9). These findings suggest that susceptibility genes for asthma and atopy are found in this region in the Chinese. Conclusions: This study has shown linkage of atopy and asthma to chromosome 5q31–33 in a heterogeneous Chinese population. These findings further substantiate the notion that chromosome 5q31–33 contains “universally” important susceptibility genes for these traits.

    Notes: Background House dust mite allergy is closely associated with allergic diseases. Blomia tropicalis mite species is an important clinical species in the tropics. The cDNA clone encoding Blo t 3, a group 3 allergen from B. tropicalis, has been isolated in our laboratory.Objective This study was designed to generate Blo t 3-specific monoclonal antibodies (mAbs) for the detection, characterization and purification of this allergen.Methods Mice were immunized intramuscularly with naked plasmid DNA encoding Blo t 3 gene with in vivo electroporation. Hybridomas were generated by the fusion of the splenocytes to X63-Ag8.653 myeloma cells. Purified native Blo t 3 was obtained by mAb immuno-affinity purification and the allergenicity of native Blo t 3 was determined by human IgE enzyme-linked immunosorbent assay (ELISA).Results A panel of class-switched and high-affinity mAb recognizing a wide spectrum of Blo t 3 epitopes have been generated. These mAbs are useful for western immunoblot assay, sandwich ELISA and affinity purification of native Blo t 3. Allergenicity of native Blo t 3 protein was examined with 44 mite-allergic sera and approximately 57% of the tested sera had positive serum IgE reactivity to the native Blo t 3.Conclusions These results demonstrated that intramuscular injection of naked DNA encoding Blo t 3 gene combined with in vivo electroporation is an effective and simple method to raise monoclonal antibodies that can be used for characterization and purification of Blo t 3.
