    Notes: To investigate a potential candidate material for making artificial red blood cells to supplement blood transfusion, the X-ray structure of porcine haemoglobin at 1.8 Å resolution was determined as part of research towards synthesizing human blood. Porcine haemoglobin was crystallized by the vapor-diffusion method, producing crystals of dimensions 0.3–0.5 mm after successive seeding. The crystals belong to the orthorhombic space group P212121, with unit-cell parameters a = 68.10, b = 72.27, c = 114.85 Å. The initial phase was determined by the molecular-replacement method, using human oxyhaemoglobin as a model. The final R factor was 21.1% for 36 820 reflections after validation of 574 water molecules. The r.m.s. deviations of bond lengths, angles, torsion angles and improper angles from their ideal values are 0.017 Å, 3.0, 20.6 and 1.8°, respectively. The average B factor is 33.63 Å2 for the haemoglobin molecule and 50.53 Å2 for the water molecules. The structure could be superimposed on a 2.8 Å resolution structure with an r.m.s. difference of 0.59 Å in main-chain atomic positions and 1.27 Å in side-chain atomic positions. Porcine and human haemoglobins are compared. A tentative model for artificial blood is proposed based on the complementarity relationship of the surface charges between haemoglobin and the surrounding cell membrane.

    Notes: Abstract The use of plasticizers to affect the properties of drug-free, self-adhesive Eudragit E-100 films with higher transparency was tested for possible transdermal drug delivery. Triacetin was found to be an effective first plasticizer for Eudragit E-100 polymer. In order to improve the flexibility and adhesiveness of Eudragit E-100 film plasticized with triacetin, a more flexible and adhesive, secondary plasticizer was added. Plasticizer–polymer compatibility was evaluated by measuring transparency, surface topography, and solubility. Secondary plasticizers with a low molecular weight and a solubility parameter similar to that of Eudragit E-100 polymer and triacetin were compatible. Further, a lower molecular weight or higher concentration of the secondary plasticizers might lead to greater plasticizing action, reduce tensile strength, and increase film elongation, independent of the hydrophilicity of the plasticizer. The adhesive strength of Eudragit E-100 film under a 180° peel test was also affected by the molecular weight and solubility parameter of the secondary plasticizers used. The results indicate that PEG 200, propylene glycol, diethyl phthalate, and oleic acid can serve as a secondary plasticizer to improve the transparency, flexibility, and adhesion of Eudragit E-100 film.

    Notes: Cephalosporin C was extracted from diluted or whole broth by PEG/salt aqueous two-phase systems. Parameters such as PEG molecular weight, salt type, pH, and salt concentration were investigated for finding a suitable extraction system. In PEG 600/ammonium sulfate or phosphate systems, Kc (partition coefficienct of cephalosporin C) was observed to be larger than 1, with Kd (partition coefficient of desacetyl cephalosporin C) being smaller than 1. The particular values of these coefficients would imply that the difficult separation of cephalosporin C and desacetyl cephalosporin C could possibly be achieved via the aqueous two-phase extraction. The addition of surfactants, water-miscible solvents, and neutral salts for enhancement of the separation efficiency was also investigated. The addition of surfactants to the system did not affect the separation efficiency substantially. Kc would increase whereas Kd decreased as a result of the addition of acetone, MeOH, EtOH, IPA, and n-BuOH. Meanwhile both Kc and Kd would decrease whenever neutral salts, NaCl, KCl, Kl, or KSCN, were added. The partitioning behavior of cephalosporin C and desacetyl cephalosporin C in filtered, whole, and different batches of broth was notably quite similar to that of diluted broth. The recovery yield of cephalosporin C in whole broth extraction was observed to be a function of centrifugal force used in phase separation. © 1994 John Wiley & Sons, Inc.

    Notes: The facilitated transport of penicillin G (Pen G), through a supported liquid membrane with Amberlite LA-2 dissolved in 1-decanol, supported on a microporous polypropylene membrane, were studied. The distribution coefficient was obtained from a batch extraction experiment. The effects of flow rate, carrier concentration, initial concentration of Pen G, and the pH of feed and stripping phases on the transport rate of Pen G through the supported liquid membrane were also investigated. The results are in agreement with theoretical predictions, and it is demonstrated that the transport of Pen G through the supported liquid membrane is controlled simultaneously by mass transfer across both aqueous and liquid membranes. © 1993 John Wiley & Sons, Inc.

    Notes: Γ-ray attenuation method was applied to determine the axial fluidizing bulk density profile for an air-solid aggregative fluidized bed. Except for beds fluidized at very high air velocities two distinct density zones were noted. The density of the bed in the lower portion of the column was relatively constant for a particular set of operating conditions and did not vary with bed height. The density in the top portion of the bed fell rapidly with bed height.The average density of the lower zone was correlated as a function of the operating variables. The density in the top zone was represented by a one-tail normal distribution curve. A somewhat better correlation was obtained by a two-parameter equation at high air velocities. The effects of operational variables, such as air velocity, static bed height, particle size, and particle-size distribution on these correlations were investigated.Friction-factor correlations based on two different definitions of bed height were also obtained.

    Notes: A method has been designed for making spherical microencapsulation of activatedcarbon particles for use in the removal of metabolic wastes and/or toxins from blood. This bench-scale method was able to make better uniformity in particle size, and at a higher particle production rate. The material properties such as particle size, particle porosity, electromicroscopy of surface morphology, surface hardness, and pore-size distribution were characterized in order to conclude with their adsorption capacity for specific toxin solutes. The performance characteristics of these self-prepared particle encapsulation was also evaluated against the commercially available hemoperfusion particles, e.g., Kuraray's DHP (Japan). The encapsulation particles were also coated with polyhydroxyethyl methacrylate (PHEMA) for blood compatibility purpose.

    Notes: The concentration of reactants at an interface cannot be determined directly. The concentration at the interface has been estimated by measuring the interfacial tension at the interface between two immiscible solutions. An aqueous phase, de-ionised water containing penicillin G, and an organic phase containing amberlite LA-2 were used as a typical system to study the kinetics of interfacial reaction. The interfacial tension was measured by a spinning-drop tensiometer at 25 ± 0·1°C. The interfacial tension was found to correlate well with the concentrations of penicillin G, Amberlite, LA-2 and proton in each respective phase.
